高温胁迫对盐藻和塔胞藻叶绿素荧光动力学的影响 预览 被引量:21

Effects of high temperature stress on chlorophyll fluorescence kinetics of Dunaliella salina and Pyramimonas sp.

摘要 利用调制式叶绿素荧光仪(Water-PAM)测定了高温胁迫(35~50 ℃)下盐藻(Dunaliella salina)和塔胞藻(Pyramimonas sp.)叶绿素荧光动力学特性的变化.测定的主要参数有:光系统Ⅱ(PSⅡ)的最大光能转化效率(Fv/Fm),PSⅡ的潜在活性(Fv/Fo),PSⅡ的实际光能转化效率(諴SⅡ=yield),光合电子传递效率(ETR),光化学淬灭(qP)和非光化学淬灭(NPQ).结果表明,与对照组(25 ℃)相比,高温胁迫下2种绿藻的叶绿素荧光参数Fv/Fm、Fv/Fo、諴SⅡ、ETR和qP均明显降低,并且随着胁迫温度的升高,胁迫时间的延长,下降幅度也逐步增大;高温胁迫下盐藻的NPQ先升高后下降.塔胞藻的NPQ随时间的变化趋势与温度有关,35 ℃和45 ℃处理下,塔胞藻的NPQ随着处理时间的延长而逐渐下降;40 ℃处理下NPQ则先下降后上升.盐藻的耐热性较好,在40 ℃和45 ℃下处理10 min后,经过一定的时间各荧光参数可恢复到接近对照水平,在50 ℃下处理10 min后各荧光参数则不能恢复.塔胞藻的耐热性较差,在35 ℃和40 ℃处理10 min后,经过一定的时间后各荧光参数可基本恢复,而在45 ℃下处理10 min后则不能恢复.本研究还对高温胁迫下2种绿藻的响应机制以及叶绿素荧光技术在筛选耐高温微藻品系中的应用进行了初步探讨. Chlorophyll fluorescence can be readily measured by a range of fluorometers,which provides a quick,non-invasive,non-destructive assessment of the efficiency of photochemical conversion by PSⅡ,thus it has become an increasingly powerful tool and widely used techniques in studies of photosynthesis in marine algae,particularly for investigations of stress physiology of microalgae.There are several chlorophyll fluorescence parameters that have been correlated to photosynthetic processes.One of the most commonly used parameters is the ratio of variable to maximal chlorophyll fluorescence(Fv/Fm),the maximal photochemical efficiency of PSⅡ or the maximal quantum yield of PSⅡ,which has been shown to be a sensitive indicator of photosynthetic performance and photoinhibition.Moreover,other fluorescence parameters,such as actual photochemical efficiency of PSⅡ in the light(ΦPSⅡ=yield),the relative electron transport rate(rETR),the photochemical quenching(qP) and the non-photochemical quenching(NPQ) are also used in investigating photosynthesis by many plant physiologists.Microalgae plays an important role in aquatic primary productivity.Some microalgal species,which have high nutritional value and can grow well in large-scale cultures,are commonly used in many mariculture systems as food for certain aquatic animals.The growth and photosynthesis of microalgae are dependent on light,salinity,temperature and nutrient.Among the different culture conditions,temperature is one of the most important environmental factors limiting algae production.In this paper,the modulated chlorophyll fluorescence kinetics of Dunaliella salina and Pyramimonas sp.under high temperature stress(35-50 ℃) were investigated with Water-PAM chlorophyll fluorometer.The results showed that the parameters Fv/Fm,Fv/Fo,ΦPSⅡ,ETR and qP decreased under high temperature stress in the two microalgae.The changing degree of the above parameters increased with the increase of temperature and exposure time.NPQ in the two microalgae exhibited different
