In vitro experimental study of the antitumor potency of snake venom nerve growth factor

摘要 目的:探讨蛇毒神经生长因子(NGF)对人肿瘤细胞增殖的抑制作用.方法:采用噻唑蓝(MTT)法、台盼蓝拒染法和集落形成作为观察指标,以NGF5,10,15,20μg/ml处理4种人肿瘤细胞(人白血病细胞株K 562、人胰癌细胞株JF305,人肺癌细胞株AGZY-83a和人胃癌细胞株MG-803),选用不同作用时间,分别观察NGF对上述细胞的影响.阿霉素(Adr)为阳性对照药.结果:NGF对4种人肿瘤细胞均有细胞毒性作用,K 562和JF 305比后两者显示出更好的量效关系(P均<0.05).NGF处理24,48和96 h后K 562和JF 305细胞的生长均受到抑制(P<0.05,P<0.01).并有显著的剂量效应关系.处理JF305细胞24,48和96 h,NGF的半抑制浓度(IC50),时间-效应关系亦明显;NGF对K 562细胞的生长抑制作用以48 h为显著.NGF还可显著抑制JF 305细胞的集落形成,并有明显的量效关系(P<0.01),其IC50为12.86μg/ml.结论:NGF对4种人肿瘤细胞均具有细胞毒性和生长抑制作用,对K 562、JF 305作用更显著. Objective: To observe the inhibitory effects of nerve growth factor(NGF) on human tumor cell lines. Methods: The MTT colorimetric method, trypan blue exclusion,and colony formation was u sed. Adr used as positive control. Four kinds of human tumor cell lines, K 562,J F 305, AGZY-83a and MG-803 were treated with NGF at different final concentrat ions of 5,10,15, and 20 μg/ml. The effects of NGF on the cells mentioned above were observed at different intervals after the cell were exposed to NGF. Results:NGF had obvious cytotoxicity on these cell lines. There were obvious dose-response relationship in test of K 562 and JF 305 cells (P< 0.05). K 562 and JF 305 were exposed to NGF at different intervals of 24,48, and 96 h, the growth of both cell lines was inhibited obviously and the dose-r esponse relationships were showed distinctively. At 24, 48, and 96 h intervals a fter JF 305 cells were exposed to NGF,there was obvious time-reponse relationsh ip. The inhibitory effect of NGF on the growth of K 562 reached peak at 48h time point. NGF also possessed inhibitory effect on colony formation of JF 305 cells with satisfied dose-response relationship (P<0.01)and the IC 50 was 12.83 μg/ml. Conclusion:NGF possesses distinct cytotoxicity and growth inhibitory effect on the four kind of human tumor cells especially o n K 562 and JF 305
