大枣多糖对免疫抑制小鼠白细胞介素2及其受体水平的影响 预览 被引量:21

Effects of jujube polysaccharide on the levels of interleukin-2 and its receptor in immunosuppressive mice

摘要 目的:探讨大枣多糖对脾细胞产生和分泌白细胞介素2(interleukin-2,IL-2)和血清可溶性白细胞介素2受体(solubleinterleukin-2 receptor,SIL-2R)水平的影响,以及作用机制.方法:用放血与环磷酰胺并用致免疫低下小鼠为研究对象,分为大、中、小剂量(剂量分别为400,200,100 mg/kg)的大枣多糖水溶液组,当归补血口服液组(6.6mL/kg,原药液稀释3倍)及同体积生理盐水组,另设完全空白对照组,每组6只.造模同时给药,连续7 d,测定小鼠脾细胞IL-2的产生及活性和血清SIL-2R水平.结果:大、中、小剂量大枣多糖组和当归补血口服液组均可显著提高1:2和1:4稀释度免疫抑制小鼠脾细胞IL-2的产生和活性,与模型组比较,差异有显著性意义(t=5.2~8.4,P<0.01),以中剂量大枣多糖组对免疫抑制小鼠脾细胞IL-2产生及活性影响作用为最强.大、中、小剂量大枣多糖组及当归补血口服液组均可显著降低血清SIL-2R水平,与模型组比较,差异有显著性意义(t=14.1~16.3,P均<0.01),以中剂量大枣多糖对血清SIL-2R水平的影响为最显著.结论:大枣多糖可促进免疫抑制小鼠脾细胞产生和分泌IL-2,降低血清IL-2R水平. AIM: To investigate the effect of jujube polysaccharide on the levels of interleukin 2 (IL 2) produced and excreted from spleen cells and serum soluble interleukin 2 (SIL 2R) and its action mechanism. METHODS:Immunodeficiency mice induced by combination of bleeding and injecting cyclophosphamide were divided into with high, middle and low dosage groups of jujube polysaccharide solution(with the dosage of 400, 200, 100 mg/kg respectively), danggui buxue oral solution group(6.6 mL/kg, 3 times of dilution as original solution) and iso volume physiological saline group and complete control group, with 6 mice in each group. Administration was carried out with the same time of model establishment for successive 7 days. The formation and activity of IL 2 in spleen cells and the level of serum SIL 2R in mice were measured.RESULTS: Formation and activity of IL 2 in spleen of immunosuppressive mice in the jujube polysaccharide solution with high, middle and low dosage groups and danggui buxue oral solution group were significantly improved with the dilution degree of 1∶2 and 1∶4. The differences had significant meaning as compared with model group (t=5.2 to 8.4, P< 0.01). Among the above mentioned groups, jujube polysaccharide solution with high dosage group had the strongest effect on the formation and activity of IL 2 in spleen of immunosuppressive mice. Jujube polysaccharide solution with high, middle and low dosage groups and danggui buxue oral solution group could significantly decrease the level of serum SIL 2R, and the differences had significant meaning as compared with model group (t=14.1 to 16.3, all P< 0.01). Among them, jujube polysaccharide solution with high dosage group had the most significant effect on the level of serum SIL 2R.CONCLUSION: Jujube polysaccharide can promote the secretion of spleen cell IL 2 in and reduce the level of serum SIL 2R in the immunosuppressive mice.
