The study measured the leaf reflectivity of the flue-cured tobacco variety ‘Zhongyan101’ and anatomically observed the leaf tissue structure of the varieties to analyze the relation between the two and investigate the effects of leaf maturity,leaf position and nitrogen rate on them. It was shown that as the nitrogen rate increased,the leaf position moved up and the leaf maturity decreased,the thickness,palisade tissue thickness and spongy tissue thickness,palisade tissue thickness/spongy tissue thickness ratio and specific weight of the leaves all tended to increase and the near-infrared reflectivity of the fresh leaves increased;there appeared a significant positive correlation between the thickness, palisade tissue thickness, spongy tissue thickness and palisade tissue thickness/spongy tissue thickness ratio, and the near-infrared reflectivity of the leaves. The leaf thickness could be used to better estimate the leaf near-infrared reflectivity and the determination coefficient of the estimation equation was 0.871^＊＊ （P〈0. 001）.
