Sm(Ⅲ)(HE)3配合物与DNA作用方式的光谱研究 预览

摘要 应用电子吸收光谱和荧光光谱手段,以吖啶橙(AO)作探针研究了Sm(Ⅲ)(HE),与DNA的相互作用.研究表明,苏木素(HE)与Sm(Ⅲ)形成了配离子,其结合比nSm(Ⅲ):nHE=1:3,Sm(Ⅲ)(HE)3与DNA的结合常数K=1.36×10^6L·mol^-1.同时研究了酸度、盐效应等对Sm(HE)3与DNA相互作用的影响,Scatchard图表明该配合物对吖啶橙与DNA的结合为非竞争性抑制.实验综合表明,Sm(Ⅲ)(HE)3与DNA之间的结合主要为外部静电作用方式. The interaction of Sm (Ⅲ ) (HE) 3 with DNA was studied by using acridine orange (AO) as a probe, which have been confirmed by absorption, fluorescence spectra and viscometric. The results indicate that there was a complex of Hematoxylin and Sm. The binary complex ratio of nsm(Ⅲ) : nHe = 1 : 3, The binding constant of Sin( Ⅲ) ( HE)3 with herring sperm DNA is K = 1.36×10^6 L· mol^-1. At the same time, we studied how the interaction of Sm( Ⅲ) (HE) 3 with DNA was effected by the changes of pH and salt number. Scatchard plots show uncompectitive inhibition of Sin( Ⅲ) (HE)3 hinging to DNA. All experimental results show that the mode of binding of Sin( Ⅲ )( HE)3 to DNA is mainly electrostatical binding.
