神经营养因子3基因转染的施万细胞在脊髓损伤大鼠中的实验研究 预览 被引量:1

摘要 目的探讨神经营养因子3基因转染的施万细胞在脊髓损伤大鼠中的作用,以进一步了解其在脊髓损伤治疗中的应用价值。方法将90只健康Wistar大鼠采用Allen打击法建立脊髓损伤模型,将其随机分为A组(空白对照组)30只、B组(神经干细胞移植组)30只和C组(神经干细胞联合神经营养因子3基因转染的施万细胞移植组)30只。三组大鼠分别于移植前、移植后1、3个月进行BBB评分、皮质诱发电位及神经功能相关指标评估及比较。结果移植后1、3个月B组和C组的BBB评分中0~7分比例(B组:70%、60%,C组:50%、30%)、皮质诱发体感和运动电位[B组皮质诱发体感电位:(201.37±23.06)、(227.63±26.48)μV,C组皮质诱发体感电位:(241.36±27.34)、(278.53±31.46)μV;B组皮质诱发运动电位:(150.23±22.67)、(193.57±27.18)μV,C组皮质诱发运动电位:(186.72±26.83)、(242.57±30.56)μV],均优于A组[90%、90%,皮质诱发体感电位:(53.86±6.75)、(56.96±7.35)μV,皮质诱发运动电位:(32.64±4.07)、(34.15土4.12)μV]。另外,B、C组神经功能相关指标也均明显优于A组,而C组则优于B组,且C组移植后3个月神经功能指标水平明显优于移植后1个月。差异均有统计学意义(均P〈0.05)。结论神经营养因子3基因转染的施万细胞在脊髓损伤大鼠中的作用较佳.有利于脊髓损伤的改善,对改善患者的感觉、运动等指标均发挥着积极作用。 Objective To study the effect of neurotrophic factor 3 gene transfected Sehwann cells in rats with spinal cord injury, in order to further understand its application value for spinal cord injury treatment. Methods 90 healthy Wistar rats were adopted Allen attack method to establish spinal cord injury model. The rats were randomly divided into group A (blank control group) with 30 cases, group B (neural stem cell transplantation group) with 30 cases and group C (neural stem cell and neurotrophic factor 3 gene transfected Schwann cells transplantation group) with 30 cases. Then the BBB score, cortical evoked potential and nerve function related indexes of the three groups before transplantation and after transplantation at l and 3 month were evaluated and compared. Results The BBB score 0 to 7 points rates (group B: 70%, 60%, group C: 50%, 30%), cortical somatosensory and motor evoked potential [group B cortical somatosensory evoked potential: (201.37土23.06), (227.63土26.48) μV , group C cortical somatosensory evoked potential: (241.36土27.34), (278.53土31.46)μV; group B cortical motor evoked potential: (150.23土22.67), (193.57土 27.18)μV, group C cortical motor evoked potential: (186.72土26.83), (242.57土30.56)μV] of group B and group C after transplantation at 1 and 3 month were all better than those of group A [90%, 90%; cortical somatosensory evoked potential: (53.86土6.75), (56.96土7.35) μV; cortical motor evoked potential: (32.64土4.07), (34.15土4.12) μV]. Nerve function related indexes in group B and C were all better than those in group A, and those indexes in group C were better than those in group B, which in group C after transplantation at 3 month were better than those at 1 month, there were all significant differences (all P 〈 0.05).
