丹参酮ⅡA抑制胃癌细胞株BGC823增殖活性研究 被引量:1

Study on the effect of tanshinone ⅡA on the proliferation of BGC823 cells

摘要 目的探讨丹参酮ⅡA对胃癌细胞株BGC823的抑制增殖活性及其可能的作用机制。方法应用MTF比色法检测不同浓度丹参酮ⅡA对BGC823细胞的增殖能力的影响,应用tit—PCR法检测给药后bel-2,bax以及c—myc基因mRNA表达水平的变化。结果MrITI’结果显示:用浓度0、2.5、5、10、20及40斗M丹参酮ⅡA处理24h后,BGC-823细胞抑制率分别0.00%、12.23%、21.79%、46.35%、77.32%和95.93%。克隆形成实验结果显示,低浓度的丹参酮ⅡA处理后的BGC.823细胞的增殖能力显著受到抑制。另外,PCR研究表明,在给药24h后,丹参酮ⅡA显著降低bcl—2和c—myc基因表达。结论丹参酮ⅡA对BC,C823细胞具有明显的抑制增殖以及诱导凋亡的作用,其可能的作用机制是通过降低bcl-2/bax的比值以及癌基因c-my~的表达有关。 Objective To observe the effect of tanshinone ⅡA on the proliferation of BGC823 cells and to in- vestigate its possible mechanism. Methods MTT assay was used to explore the effects of tanshinone ⅡA on the pro- liferation of BGC823 cells. Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to examine mRNA levels of bel-2, bax and c-myc genes. Colony formation experiment was used to test the anchoring dependent growth of BGC-823 cells. Results The inhibitory rate of tanshinone ⅡA (0,2.5,5,10,20,40μM) on BGC-823 cells was O. 00%, 12.23% ,21.79% ,46.35% ,77.32% ,95.93% after treament for 24h. The colony forming efficiency rate was reduced remarkably. Tanshinone ⅡA down-regulated the expression of bcl-2, c-myc and up-regulated the mRNA expression of bax. Conclusion Tanshinone ⅡA reversed the malignant phenotype of BGC-823 cells and induced the apoptosis of BGC-823 cells through down-regulated bcl-2/bax ratio and c-myc expression.
