目的 研究内毒素血症Wistar大鼠血清TNF-α与心肌细胞凋亡的关系及EPO对其的影响.方法 将180只Wistar大鼠随机分为对照组、脂多糖LPS组及EPO处理组,分别在处理后2h、4h、6h、12h、24h断头取血并取其心脏.采用ELISA法测定血清TNF-α,用TUNEL法检测原位凋亡.结果 ①LPS组TNF-α浓度在2h时达到最大值,且明显高于对照组（P＜0.05）,随后TNF-α浓度开始下降,并在6h时降到正常,与对照组之间无统计学差异（P＞0.05）.LPS＋EPO组TNF-α浓度在2 h时达到最大值,且明显低于LPS组（P＜0.05）,随后开始下降,在6h时降到正常值;②LPS组心肌细胞凋亡指数在2h时达到最大值,且明显高于对照组（P＜0.05）,随后开始下降,并在4h、6h、12h、24h时均明显高于对照组（P＜0.05）.LPS＋EPO组心肌细胞凋亡指数在2h时达到最大值,且明显低于LPS组（P＜0.05）,随后开始下降,并在4 h、6 h、12 h、24 h时均明显低于LPS组（P＜0.05）;③心肌细胞凋亡指数与血清TNF-α存在明显的直线正相关性.结论 血清TNF-α参与了内毒素血症Wistar大鼠的心肌损伤的病理过程,与心肌细胞凋亡呈正相关性,EPO通过抑制TNF-α的产生而延缓心肌细胞凋亡,对内毒素血症Wistar大鼠的心肌具有保护作用.
Objective To study the relationship of rat serum TNF-α of endotoxin blood disease and the apoptosis of myocardial cells and EPO＇s effects on it. Methods 180 Wistar rats were randomly divided into the control group, lipopolysaccharide LPS group and EPO treatment group. All the rats were respectively beheaded 2h, 4h, 6h, 12h and 24h after treatment for their blood samples and their levers were also taken out. Their TNF-α were tested by way of ELISA, and their in situ apoptosis were tested through the method of TUNEL. Results ①The TNF-α concentration of the LPS group reached its highest level in 2 hours, and it was obviously higher than that of the control group （P〈 0.05）, then the concentration began to decrease, and resumed its normal level in 6 hours. It was statistically insignificant （P〉0.05）. The TNF-α concentration of LPS＋EPO group reached its peak in 2 hours, and it was obviously lower than that of the LPS group （P〈0.05）, then it began to decrease, and became normal in 6 hours; ②The apoptosis index of myocardial cells of the LPS group reached its peak in 2 hours, and it was obviously higher than that of the control group （P〈0.05）, then it began to decrease, it was respectively and obviously higher than that of the control group in 4h, 6h, 12h and 24h （P〈0.05）. The apoptosis index of myocardial cells of the LPS＋EPO group reached its peak in 2 hours, and it was obviously lower than that of the control group （P〈0.05）, then it began to decrease, and was respectively and obviously lower than that of the LPS group in 4h, 6h, 12h and 24h （P〈0.05） ; ③ The myocardial apoptosis index had an obviously positive linear correlation with TNF-α. Conclusion The serum TNF-α is involved in the myocardial pathological injury process of Wistar rats with endotoxin blood disease, it has a positive correlation with myocardial cell apoptosis.
