灭幽汤对幽门螺杆菌相关性胃炎脾胃湿热证模型小鼠Toll样受体2、4的影响 预览 被引量:10

Effects of Mieyou Decoction on TLR2 and TLR4 in H. pylori-associated Gastritis Mice ofSpleen-stomach Damp-heat Syndrome

摘要 目的 探讨灭幽汤治疗幽门螺杆菌(Hp)相关性胃炎脾胃湿热证的作用机制.方法 将70 只BALB/c小鼠随机分为对照组、模型组、胃三联组和灭幽汤高、低剂量组(中药高、低剂量组),每组14 只,采用复合因素建立BALB/c 小鼠Hp 相关性胃炎脾胃湿热证模型.成模后各给药组给予相应药物,连续14 d,采用免疫组化检测Toll 样受体(TLR)2、TLR4 蛋白表达,实时荧光定量PCR 检测TLR2、TLR4 基因表达,ELISA 检测血清白细胞介素(IL)-6、IL-8 的表达.结果 模型组TLR2、TLR4 蛋白和基因表达水平及血清IL-6、IL-8 含量均高于对照组(P 〈0.01);中药高、低剂量组和胃三联组TLR2、TLR4 蛋白和基因表达及血清IL-6、IL-8 含量均低于模型组(P 〈0.01);中药高剂量组TLR2、TLR4 蛋白和基因表达水平及血清IL-6、IL-8 含量虽低于胃三联组,但差异无统计学意义(P 〉0.05).结论 灭幽汤可能通过抑制TLR2、TLR4 及下游炎症因子的表达达到治疗Hp 相关性胃炎脾胃湿热证的目的. Objective To investigate the mechanisms of Mieyou Decoction in the treatment of Hp-associated gastritis of spleen-stomach damp-heat syndrome. Methods Seventy BALB/c mice were randomly divided into control group, model group, high concentration of Mieyou Decoction group, low concentrations of Mieyou Decoction group, and gastric triad group, with 14 rats in each group. BALB/c mice of Hp-associated gastritis of spleen-stomach damp-heat syndrome models were established by composite factor. After the success of modeling and continuous 14-day administration, the expressions of TLR2, TLR4 protein, TLR2 mRNA, and TLR4 mRNA were detected by immunohistochemistry and qPCR. ELISA was used to detect the expressions of IL-6 and IL-8 in the serum. Results Compared with control group, the expressions of TLR2, TLR4 protein, mRNA, IL-6, and IL-8 in model group significantly increased (P<0.01);the expressions of TLR2, TLR4 protein, mRNA, IL-6, and IL-8 in high concentration of Mieyou Decoction group, low concentrations of Mieyou Decoction group, gastric triad group were lower than model group, with statistical significance (P<0.01);the expressions of TLR2, TLR4 protein, mRNA, IL-6, and IL-8 in high concentration of Mieyou Decoction group were higher than gastric triad group, but the differences were not statistically significant (P>0.05). Conclusion Mieyou Decoction may play a role in the treatment of Hp-associated gastritis of spleen-stomach damp-heat syndrome through intervention in the expressions of TLR2 and TLR4.
