目的分析江苏省2013年流行性腮腺炎病毒（Mu V）的基因特征。方法针对2013年分离到的139株Mu V小疏水蛋白（small hydrophobic protein,SH）基因316个核苷酸片段进行反转录-聚合酶链反应（RT-PCR）扩增,并对该PCR产物进行序列测定,将139株Mu V与世界卫生组织（WHO）Mu V基因型参考株一起进行分子流行病学研究。结果江苏省136株Mu V分离株属于F基因型,核苷酸和氨基酸同源性分别为93.4%～100%和84.3%～100%。3株Mu V分离株属于G基因型,核苷酸和氨基酸同源性分别为95.6%～100%和93%～100%。Mu V分离株在SH基因编码的氨基酸保守位点也发生了变化。结论江苏省2013年主要流行的Mu V基因型为F基因型,同时也出现G基因型的流行,Mu V毒株与疫苗株S79遗传距离差异较大。F基因型参考株与江苏省2013年的F基因型Mu V毒株间序列差异较大,说明江苏省流行的Mu V发生了一定程度的变异。另外,还发现所有F基因型和G基因型Mu V在SH基因上分别存在5个特异性突变,而其他基因型Mu V在这些位点上均未发生改变。
Objective To analyze the genetic characteristics of mumps virus（ Mu V） in Jiangsu in 2013. Methods In the study,the fragments containing 316 nucleotides of small hydrophobic protein（ SH） genes from 139 mumps virus isolates in Jiangsu in 2013 were amplified by RT- PCR method,the PCR products were directly sequenced. The 139 strains of Mu V were compare with WHO mumps reference strains for molecular epidemiological study. Results 136 isolated strains were F genotypes,the homology of nucleotide and amino acid among the 136 isolated strains were 93. 4%- 100% and 84. 3%- 100%,respectively. Three isolated strains were G genotypes,the homology of nucleotide and amino acid among the three isolated strains were 95. 6%- 100% and 93%- 100%,respectively. Amino acids conserved sites of SH gene encoding in Mu V isolates also changed. Conclusion F genotype of mumps virus was the predominant genotype in Jiangsu in 2013,and G genotype was also found,the S79 genetic distance difference between Mu V strains and vaccine strains was large. The sequence difference between mumps virus F reference strains and mumps viruses F genotype strains in Jiangsu in 2013 were large. It suggested that genetics evolution happened on these mumps virus in some extent. Furthermore,comparing with other genotype mumps strains from other countries,there were five nucleotides mutations in SH gene in all F and G genotype mumps viruses. However,these mutations were not found in the other genotype of mumps virus.
