电针对自发性胰岛素抵抗模型大鼠血管内皮p38MAPK的影响

Effects of electroacupuncture therapy on the p38MAPK expression in vascular endothelium of rat models with spontaneous insulin resistance

摘要 目的:探讨电针对自发性胰岛素抵抗大鼠血管内皮p38MAPK及心肌超微结构的影响。方法:以8只雄性LETO大鼠作为空白组,将16只雄性OLETF大鼠随机分为两组,即电针组及模型组,每组8只。空白组及模型组进行正常饲养,不给予其它处理。电针组针刺双侧内关、三阴交、足三里及肾俞,并予同侧足三里及三阴交加电针刺激。每日1次,每次20min,持续4周。治疗结束后,禁食12h,次晨眼眶静脉窦采血。检测各组大鼠空腹血糖(FPG)、空腹胰岛素(FINS)、稳态模型评估的胰岛素抵抗指数(HOMA-IR)、C肽(C-P)。白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)、丙二醛(MDA),主动脉内皮p38MAPK蛋白表达及左心室心肌超微结构。结果:模型组FPG、FINS、C-P、HOMA-IR、TNF-α、IL-6、MDA的血液水平及p38MAPK蛋白表达水平较空白组显著升高(P〈0.05),SOD水平较空白组降低(P〈0.05);电针组的FPG、FINS、C-P、TNF-α、IL-6、MDA的血液水平及p38MAPK蛋白表达的水平较模型组显著降低(P〈0.01,P〈0.05),SOD水平较模型组升高(P〈0.05)。电镜显示,针刺干预能改善心肌细胞的病理状态。结论:针刺能改善胰岛素抵抗及炎性状态,有助于内皮功能障碍的改善,对心肌病理变化具有一定的良性作用。 Objective:To investigate the effects of electroacupuncture therapy on the expression of p38-MAPK protein in vascular endothelium and the myocardial ultrastructure of rat models with spontaneous insulin resistance.Methods:Eight male Long-Evans Tokushima(LETO) rats were taken as the blank group and sixteen Otsuka Long-Evans Tokushima Fatty(OLETF) rats were randomly divided into two groups,electroacupuncture group and model group,8 in each group.Rats in the blank group and model group were normal feeding with no treatments.Rats in electroacupuncture group were given acupuncture at bilateral Neiguan(PC6),Sanyinjiao(SP6),Zusanli(ST36) and Shenshu(BL23),and electroacupuncture was given to the same side of Zusanli(ST36) and Sanyinjiao(SP6) for 20 min per day and for 4 weeks in total.At the end of the experiment,blood was taken from orbital veins on the next morning after 12 hours' fasting.The fasting plasma glucose(FPG),fasting insulin(FINS),homeostasis model assessment of insulin resistance(HOMA-IR),C-peptide(C-P),interleukin-6(IL-6),tumor necrosis factor alpha(TNF-α),superoxide dismutase(SOD),malondialdehyde(MDA),expression of p38 MAPK protein in aortic endothelium and myocardial ultrastructure of the left ventricle of rats in all groups were detected.Results:Compare with the blank group,the levels of FPG,FINS,C-P,HOMA-IR,TNF-α,IL-6,MDA and p38 MAPK expression of the model group were greatly higher(P〈0.05),SOD level was significantly lower(P〈0.05).Compared with the model group,the levels of FPG,FINS,C-P,TNF-α,IL-6,MDA and p38 MAPK expression of the electroacupuncture group significantly decreased(P〈0.01,P〈0.05),SOD level was significantly higher(P〈0.05).Acupuncture could improve the pathological state of myocardial cells seen from electronmicroscope.Conclusion:Acupuncture can regulate insulin resistance and inf lammatory state,improve endothelial dysfunction and positively af fect the pathological changes of the myoc
