枸杞多糖对去卵巢大鼠肝脏HMGCR、L-FABP及LPL的调控作用研究

摘要 目的:通过观察枸杞多糖对去卵巢大鼠肝脏中HMGCR、L-FABP及LPL蛋白表达的影响,探究枸杞多糖对去卵巢大鼠脂代谢的作用机制。方法:30只SPF级健康雌性SD大鼠随机分为假手术组、去卵巢组、补佳乐组、枸杞多糖高剂量组及枸杞多糖低剂量组。全自动生化分析仪检测大鼠血脂水平,HE染色观测各组大鼠肝脏组织形态学变化,Western Blot方法检测肝脏中HMGCR、L-FABP及LPL蛋白表达。结果:血脂检测发现,与假手术组相比,去卵巢组大鼠血清TC、LDL-C水平显著升高、HDL-C水平显著下降(P〈0.01);与去卵巢组相比,补佳乐组与枸杞多糖高剂量组TC水平显著下降(P〈0.01),补佳乐组与枸杞多糖高、低剂量组LDL-C水平显著下降(P〈0.01 or P〈0.05)。HE染色发现,去卵巢组可见散在的脂肪空泡,高剂量枸杞多糖治疗后脂肪空泡减少。Western Blot结果显示,与假手术组相比,去卵巢组大鼠肝脏HMGCR、L-FABP蛋白表达上调,LPL蛋白表达下调(P〈0.01);与去卵巢组相比,枸杞多糖高剂量组HMGCR、L-FABP与LPL蛋白表达呈相反趋势(P〈0.01)。结论:高剂量枸杞多糖可能通过下调肝脏中HMGCR与L-FABP,上调LPL蛋白表达改善脂代谢紊乱。 Objective:To explore the effect of LBP on lipid metabolism in ovariectomized rats through observing the blood lipid,liver histolomorph and the exprressions of HMGCR,L-FABP and LPL protein in liver.Methods:Thirty SPF healthy female SD rats were randomly divided into sham operation group,ovariectomized group,Progynova group,LBP high dose group and LBP low dose group.Blood lipid levels were detected by fully automatic biochemical analyser.Liver tissue morphology was detected by HE staining.The exprressions of HMGCR,L-FABP and LPL protein in liver were detected by Western Blot.Results:Compared with the sham operation group,TC and LDL-C were significantly increased and HDL-C was significantly decreased in ovariectomized group(P〈0.01).Compared with ovariectomized group,the levels of TC and LDL-C in Progynova group and LBP high dose group were significantly decreased(P〈0.01 or P〈0.05).HE staining found that there were fat cavitations in ovariectomized group livers,while fat cavitations in LBP high dose group were significantly decreased.According to the results of Western blot,compared with the sham operation group,the exprressions of HMGCR and L-FABP protein were significantly increased and LPL protein was significantly decreased in ovariectomized group(P〈0.01).Compared with ovariectomized group,expression tendency was opposite(P〈0.01 or P〈0.05).Conclusions:High dose LBP may improve the lipid metabolism disorders through increasing the exprressions of HMGCR,L-FABP and LPL protein of livers in ovariectomized rats.
