Expression of NDRG4 in ovarian cancer

摘要 目的 探讨卵巢上皮性肿瘤中N-myc下游调节基因4(N-myc downstream regulated gene4,NDRG4)的表达水平与卵巢上皮性恶性肿瘤发生发展及预后的关系。方法 采用免疫组化法及Realtime-PCR检测了90例卵巢上皮性癌组织(包括30例卵巢粘液性腺癌、30例浆液性腺癌、30例子宫内膜样腺癌)、60例正常卵巢组织标本中NDRG4的表达情况,并结合临床耐药性及预后参数进行分析。结果卵巢上皮性肿瘤组织中及卵巢上皮性交界性肿瘤组织中NDRG4表达低于正常卵巢组织,差异具有统计学意义(P〈0.05,P〈0.01)。RT-PCR结果显示,NDRG4在在卵巢癌组织中的表达明显低于正常组织;90例卵巢癌样本与60例正常卵巢标本通过每组4例卵巢癌组织及2例正常卵巢组织以RealtimePCR方法相比,NDRG4在正常卵巢组织的表达明显低于卵巢癌组织,趋势明显。结论 NDRG4作为一种新型抑癌基因与卵巢上皮性癌的临床变量、预后及放化疗药物的耐药有关。 Objective To investigate the relationship between the expression level of NDRG4 and the occurrence, development and prognosis of epithelial ovarian tumor. Methods Expres- sion of NDRG4 in 60 cases of normal ovarian tissue and in 90 cases of epithelial ovarian cancer (including 30 cases of ovarian mucinous adenocarcinoma, 30 cases of serous adenocarcinoma, 30 cases of endometrial adenocarcinoma) was detected by immunohistochemical staining and Reahime-PCR, and were analyzed combined with clinical drug resistance and prognosis param- eters. Results The expression of NDRG4 in epithelial ovarian tumor and epithelial junctional tumor of ovary,was lower than that in normal ovarian tissue (P 〈 0.05, P 〈 0. 01 ). RT-PCR results showed that the expression of NDRG4 in ovarian cancer tissues was significantly lower than that in normal tissue. Ninty cases of ovarian cancer samples and 60 cases of normal ovarian specimens from each group of 4 cases of ovarian carcinoma tissues and 2 cases of normal ovarian tissue compared with RT-PCR method, the expression of NDRG4 in normal ovarian tissues was significantly lower than in ovarian cancer tissues, and the trend was obvious. Conclusion NDRG4 is related to the clinical variables, prognosis and the resistance to chemotherapy of ovarian epithelial cancer.
