尾吊复合隔离导致的大鼠抑郁样行为研究 被引量:1

Study on Depression-like Behavior Induced by Tail Suspension Combined with Isolation in Rats

摘要 目的研究2周的尾吊复合隔离环境所导致的大鼠抑郁样行为。方法 24只雄性SD大鼠随机平均分为3组:对照(Con)、尾吊(tail suspension,TS)、尾吊复合隔离(tail suspension combined with isolation,TS+I)。造模前后检测摄食量和体重变化,造模结束后依次进行空场、新奇事物探索、强迫游泳实验。结果 TS和TS+I组大鼠体重和摄食量低于对照组,且TS+I组最低。TS和TS+I组大鼠的主动性活动显著少于对照组,且TS+I组大鼠最少。TS和TS+I组大鼠探索潜伏期变长,且探究行为减少,其中TS+I组大鼠表现最差。TS和TS+I组大鼠的攀爬次数少于对照组,TS+I组大鼠不动时间最长。结论 2周的尾吊复合隔离环境导致的抑郁样行为比单一尾吊更为严重,此模型可用于航天抑郁发病机制研究及对抗药物开发。 Objective To investigate the depression-like behavior induced by tail suspension combined with isolation in rats. Methods Twenty-four male SD rats were randomly divided into 3 groups: control(Con),tail suspension(TS) and tail suspension combined with isolation(TS + I). Four typical methods including food intake and bodyweight variation test,open field test,novelty exploration test and forced swimming test were deployed to assess emotional activity after 2-week simulation. Results The body weight and food intake of TS and TS + I group were significantly reduced,and the TS + I group was the lowest group. In the open field test,the locomotor activity of TS and TS + I group were lower than those of Con group,and the locomotor activity of TS + I group was the least. In the novelty exploration test,the latency of TS and TS + I group were longer than those of control group,meanwhile,the rats spent less time on the novelty. Among the three groups,the performance of TS + I group was the worst. In the forced swimming test,the climbing behavior in TS and TS + I group was decreased significantly,the immobility time of TS + I group was longer than those of TS and Con group. Conclusion Compared with tail suspension,tail suspension combined with isolation can lead more serious depression-like behavior in rats. These results could provide an appropriate animal model for the study on pathogenesis of depression and countermeasure drug development under space circumstances.
