目的 探讨microRNA-124（miR-124）表达水平及其对非小细胞肺癌（NSCLC）患者诊断和临床意义。方法 选取辽宁省肿瘤医院自2016年1月至2017年1月收治并确诊的NSCLC患者75例作为肺癌组,并选取同期60例健康体检者作为健康组。采用实时定量PCR对miR-124血清含量进行测定,并利用ROC曲线评价其对肺癌诊断的灵敏性和特异性。结果 肺癌组患者血清miR-124表达水平（1.22±0.24）,显著高于健康组受试者的（0.86±0.19）,两组间比较,差异有统计学意义（P〈0.05）。肺癌组患者血清中miR-124表达水平与患者年龄、性别以及肿瘤直径无关（P〉0.05）,而与吸烟史、淋巴结转移情况以及临床分期有关（P〈0.05）。ROC曲线面积为0.902,95%可信区间84.91~95.40。结论 NSCLC患者miR-124血清水平显著高于正常人,对于诊断和评估肺癌有较大的参考价值,可能是肺癌尤其是NSCLC的检测指标之一。
Objective To investigate the expression level of microRNA-124（miR-124） and its clinical significance in the diagnosis of non-small cell lung cancer（NSCLC）. Methods A total of 75 NSCLC patients treated in hospital from January 2016 to January 2017 were enrolled as the lung cancer group and 60 normal physical examination people were enrolled as the normal control group at the same time. MiR-124 content was measured by real-time quantitative PCR and the sensitivity and specificity for the diagnosis of lung cancer of the miR-124 were evaluated by the ROC curve. Results The serum level of NSCLC in patients with miR-124 （ 1.22 ＋ 0. 24）was significantly higher than that of normal subjects （0. 86 ＋ 0. 19 ）, P 〈 0. 01. The serum levels of miR-124 in patients with NSCLC were little correlated with the age of patients, sex and tumor diameter（P 〉 0. 05 ）. While, there were independent of smoking history, lymphatic metastasis and clinical stage （ P 〈 0. 01 ）. The ROC curve area is 0. 902,95 % CI = 84. 91 - 95.40. Conclusion The serum level of NSCLC in patients with miR-124 is significantly higher than that of normal subjects. It is of great reference value for the diagnosis and evaluation of lung cancer, and may be an index for detecting lung cancer, especially for NSCLC.
