美金刚对罗哌卡因致小鼠惊厥作用的影响 预览

摘要 目的观察美金刚(memantine,Mem)对罗哌卡因致小鼠惊厥作用的影响。方法将小鼠随机分为4组:罗哌卡因组(R组)、美金刚5mg·kg^-1、10mg·kg^-1、20mg·kg^-1组(Mere5、Mem10、Mem20组)。R组给予生理盐水灌胃,Mem5、Mem10、Mem20组给予相应剂量的美金刚灌胃,30min后各组均腹腔注射罗哌卡因89.4mg·kg^-1。观察小鼠的惊厥潜伏期、惊厥持续时间和惊厥级数。结果与R组相比,Mere5组和Mem10组惊厥潜伏期延长,惊厥持续时间明显缩短,差异有统计学意义(P〈0.05或P〈0.01),Mem10组惊厥级数的平均秩次降低,差异有统计学意义(P〈0.05);Mere20组惊厥潜伏期和惊厥持续时间均缩短(P〈0.05)。结论美金刚在5~10mg·kg^-1范围内能拮抗罗哌卡因致小鼠惊厥作用。 Objective To observe the influence of memantine on ropivaeaine - induced convulsion in mice. Methods Mice were randomly divided into four groups: a ropivaeaine (R) group, and memantine 5 mg·kg^-1( Mem5 ) , 10 mg·kg^-1 ( Meta10 ) and 20 mg·kg^-1 ( Mem20 ) groups. Normal saline was administered intragastrically in the R group, while the corresponding dose of memantine was administered intragastrieally in the Mem5 , Meml0 and Mem20 groups. After 30 min, 89.4 mg·kg^-1 ropivacaine was injected intraperitoneally in each group. The convulsion latency, convulsion duration and convulsion level of mice were observed. Results Compared with the R group, the Mem5 and Mere10 groups showed remarkably extended convulsion latency and shortened convulsion duration ( P 〈 0. 05 or P 〈 0.01 ) , while the Meml0 group also presented markedly reduced average convulsion level ( P 〈 0.05 ). Both the convulsion latency and duration were reduced in the Mem2o group, compared with the R group ( P 〈 0.05 ). Conclusions Memantine can antagonize ropivacaine -induced convulsion in mice at 5 to 10mg·kg^-1.
