聚酰胺-大孔树脂联用纯化川射干总异黄酮

Purification of Total Isoflavones from Iris Tectorum with Polyamide-macroporous Resin

摘要 目的:寻找一种有效富集川射干总异黄酮的方法并对工艺条件进行初步探索。方法:样品溶液以聚酰胺吸附,水洗除杂,将除杂后的聚酰胺置于大孔吸附树脂柱顶部,以一定体积分数的乙醇溶液洗脱富集总异黄酮。以总异黄酮收率和纯度为指标,通过单因素实验和正交试验筛选最优的富集总异黄酮的工艺条件。结果:聚酰胺与AB-8树脂联用能有效富集总异黄酮。最佳工艺条件为提取液与聚酰胺比为1∶1,与树脂比为1∶3,以40%乙醇洗脱,收集洗脱液7 BV,色谱柱径高比1∶30,在此纯化条件下异黄酮纯度由19.71%增加到53.43%,收率为60.83%。结论:该方法优于目前常用的总异黄酮富集方法,且工艺简单,具有良好的前景。 Objective: To explore the enriching method for the total isoflavones from Iris tectorum Maxim. with polyamide-macroporous resin. Methods: Polyamide adsorbing sample was washed with water firstly to eliminate the impurities,and then was loaded on the top of macroporous resin. The total isoflavones were enriched by eluting with ethanol. The best conditions for the enrichment of isoflavones were screened by single factor test and orthogonal test with the purity and the recovery rate of total isoflavones as the indices. Results: AB-8 resin combined with polyamide could effectively enrich the total isoflavones. The preferred conditions for the procedure were as follows: the weight of polyamide and dry AB-8 resin was one-fold and three-fold of the extract liquids( 1 ∶ 1 and 1 ∶ 3),respectively,the volume of 40% ethanol for the elution was 7-fold of the column volume,and the column diameter ratio was 1 ∶ 30.The purity of the total isoflavones before and after the purification process was 19. 71% and 53. 43%,respectively. And the recovery was 60. 83%. Conclusion: The method is superior to the others for enriching total isoflavones with more convenient operation,which exhibits good application prospects.
