阿拉善沙漠双峰驼的微卫星多态性与体尺性状的关联分析

Correlation between microsatellite polymorphism with body size traits of Alashan Desert Bactrian camels

摘要 为了探讨所选择的微卫星座位作为阿拉善沙漠双峰驼群体生长指标遗传标记的可行性,利用PCR技术和非变性聚丙烯酰胺凝胶电泳技术,检测17个微卫星标记在阿拉善沙漠双峰驼群体中的多态性,并与体尺等性状进行关联分析。结果:17个微卫星标记在阿拉善沙漠双峰驼群体中共检测到68个等位基因,平均有效等位基因数为2.836 7,平均预期杂合度为0.619 2,多态信息含量在0.268 8~0.717 1之间,平均多态信息含量为0.546 1。有7个微卫星标记与阿拉善沙漠双峰驼体高、体长、胸围、管围和体重5个体尺性状有不同程度的显著关联,其中标记YWLL29和标记VOLP32与阿拉善沙漠双峰驼的体高、体长、胸围、管围、体重性状有关联;标记LCA33与体高、胸围、体重、体长性状有关联;标记LCA37与胸围、体重、管围性状有关联;标记CVRL01与体长、体重、胸围性状有关联;标记LCA90和标记CMS104仅与体高性状有关联。本研究结果可为阿拉善沙漠双峰驼体尺性状的标记辅助选择提供参考依据。 In order to investigate the feasibility of microsatellite loci to be selected as genetic markers of growth indexes for Alashan desert camel population,the polymorphism of 17 microsatellite markers were detected in the Alashan desert camel population by PCR technology and non-denaturing polyacrylamide gel electrophoresis. Then,association between microsatellite markers and body size traits of the animlas was analyzed. The results showed that a total of 68 alleles were detected in the Alashan desert camel population,with an average effective allele number of 2. 8367,the average expected heterozygosity of 0. 6192. The PIC value of 17 microsatellite markers ranged from 0. 2688 to0. 7171,with an average PIC of 0. 5461. Seven microsatellite markers(LCA33,LCA37,LCA90,CMS104,CVRL01,YWLL29,and VOLP32) were associated with five body size traits including body height,body length,chest circumference and body weight at different significant levels. Of the seven microsatellite markers,YWLL29 and VOLP32 were associated with body height,body length,chest circumference,shin circumference and body weight. LCA33 was associated with body height,chest circumference,body weight and body length.LCA37 was associated with chest circumference,body weight and shin circumference. CVRL01 was associated with body length,body weight and chest circumference. LCA90 LCA90 and CMS104 were only associated with body height. The results of this study provide a reference for marker assisted selection body measure traits of Alashan desert camels.
