QHF复方对人肝癌Huh7细胞生长凋亡、侵袭能力的作用及其对肿瘤干细胞表面标志物的影响 预览

摘要 目的 研究QHF复方对人肝癌Huh7细胞生长凋亡、侵袭能力的作用,并初步研究其对肿瘤干细胞表面标志物的影响.方法 体外培养人肝癌Huh7细胞,MTT法检测QHF复方对细胞生长、增殖的影响,流式细胞仪检测QHF复方对Huh7细胞周期及凋亡的影响,Transwell实验检测QHF复方对Huh7细胞侵袭能力的影响,并采用流式细胞仪检测肿瘤干细胞表面标志物CD133、CD44的表达情况.结果 QHF复方能抑制Huh7细胞增殖,且这种抑制作用在一定程度上具有剂量和时间依赖性;QHF复方能诱导细胞凋亡,使其生长周期阻滞在S期;QHF复方能够降低Huh7细胞的侵袭能力;QHF复方能够下调肿瘤干细胞表面标志物CD133、CD44的表达.结论 中药QHF复方能够抑制Huh7细胞的生长、侵袭,这种抑制作用可能与肿瘤干细胞表面标志物CD133、CD44表达的下调有关. Objective To explore the influence of Chinese herbal compound QHF on proliferation, apoptosis, invasion of human hepatoma cell Huh7 and expression of surface markers of cancer stem cells.Methods Human hepatoma cells Huh7 were cultured in vitro. Cell proliferation inhibition rate was detected by MTT assay. Cell cycle, apoptosis and cell surface marker CD133 and CD44 expressions were detected by Flow cytometry. Cell invasion was observed by Transwell experiments.Results The QHF could inhibit the proliferation of Huh7 cells, and the inhibitory effect had a certain degree of dose and time dependence. The QHF could promote the liver cancer cell apoptosis and significantly blocked cell grouth cycle in S phase. The QHF could reduce the invasion of Huh7 cells. The QHF could down-regulate the expressions of CD133 and CD44 in cancer stem cell surface markers.Conclusion The QHF can inhibit the proliferation and invasion of human hepatoma cell Huh7. This inhibition may be related to the reduction of CD133 and CD44 expressions in cancer stem cell surface markers.
