Hsa_circRBM23在肝细胞癌细胞系的表达及其对HepG2细胞增殖和迁移的影响 预览

摘要 目的探讨hsa_eircRBM23在肝细胞癌(HCC)细胞系的表达及其对HepG2细胞增殖活力及迁移能力影响。方法采用实时定量聚合酶链反应(RT-PCR)检测hsa-circRBM23在正常肝细胞系HL-7702及HCC细胞系HepG2、HHCC、HUH7和BEL-7402中的表达。转染hsa_circRBM23siRNA下调hsa_circRBM23在HCC细胞系HepG2中的表达,采用活细胞计数试剂盒(CCK-8)、EdU检测、细胞创伤愈合实验和Transwell细胞迁移实验对转染后HepG2细胞及对照组的细胞增殖活力、细胞迁移能力进行检测。结果与正常肝细胞系HL-7702比较,HCC细胞系HepG2、HHCC、HUH7和BEL-7402中的hsa_circRBM23表达量均明显升高(P〈0.01)。与对照组比较,hsa-circRBN23siRNA明显降低了hsa-circRBM23在HepG2细胞中的表达(P〈0.01)。下调hsa-circRBM23表达后,HepG2细胞的增殖活力、抑制创伤愈合、抑制细胞迁移能力下降,且HCC细胞s期细胞的百分比降低(P〈0.01)。结论Hsa_circRBM24可能在HCC发生机制中发挥一定作用,下调hsacircRBM24表达可下调HCC细胞的增殖和迁移。 Objective To investigate the expression of hsa_ circRBM23 in hepatocellular carcinoma(HCC) cell lines and explore the role of hsa_circRBM23 in the growth and migration of HepG2. Methods The expressions of hsa_circRBM23 in the normal liver cell line HL-7702 and HCC cell lines including HepG2, HHCC, HUH7 and BEL-7402 were determinated by real-time quantitative polymerase chain reaction(RT-PCR). Hsa_circRBM23 siRNA was transfected to HepG2 cells to reduce the expression of hsa_circRBM23. Cell viability and cell migration were determinated by CCK-8 and EdU wound healing assay and Transwell cell migration assay. Results Compared with the normal liver cell line HL-7702, the expression of hsa circRBM23 was significantly up-regulated in HCC cell lines including HepG2, HHCC, HUH7 and BEL-7402 ( P 〈0. 01 ). Hsa_circRBM23 siRNA could significantly suppressed the expression of hsa_circRBM23 in HepG2 cell line than that in the control group( P 〈 0.01 ). Downregulation of hsa_circRBM23 turned to significantly increase the viability of HepG2 cells, inhibit the wound closure,weaken the ability of migration and reduce the percentage of S phase cells compared with the control group(P 〈0.01 ). Conclusion Hsa_circRBM23 may play an important role in the tumorigenic mechanism of HCC and the down-regnlation of hsa_circRBM23 may suppress the growth viability and migration of HCC cell line.
