不同发育时期小鼠腓肠肌Myostatin表达的研究

Study on myostatin expression in gastrocnemius muscle at different developmental stages in mice

摘要 肌生成抑制素(myostatin,MSTN)是肌肉生长发育的反向调节因子,在骨骼肌中大量表达,其在成肌细胞的增殖、分化和融合过程当中扮演着重要的调控作用。本试验通过HE染色、免疫组织化学和PCR等技术研究了各个发育阶段小鼠腓肠肌中Myostatin的定位及其表达差异。结果,Myostatin在幼年、性成熟、体成熟和老年4个时期的小鼠腓肠肌中均有表达,且表达量存在明显的差异,Myostatin在腓肠肌中表达量最高的是在体成熟时期,最低的是在幼年时期,而在性成熟与老年时期的表达量则低于体成熟时期的小鼠。上述结果表明,Myostatin基因可能参与了小鼠骨骼肌的生长发育,这为提高肉用家畜产肉率和防治肌肉疾病提供了理论基础。 Myostatin(MSTN),a negative regulatory factor for muscle growth and development,is expressed in skeletal muscle and plays an important role in the proliferation,differentiation and fusion of myoblasts.In this experiment,HE staining,immunohistochemical and PCR techniques were used to study the localization and differential expression of myostatin in the gastrocnemius of mice at different developmental stages.In result,myostatin was expressed in mice gastrocnemius of young,sexual maturation,body maturation and aged,and there was significant difference in the expression of myostatin.It was the highest in body maturation mice,the lowest in young mice,but slightly lower in sexual maturation and aged mice than in body maturation mice.The above-mentioned results showed that the myostatin gene may be involved in the growth and development of mice skeletal muscle,which provided a theoretical basis for improving the meat production of domestic animals and the prevention and treatment of muscle diseases in animal.
