高效液相色谱测定明日叶中查尔酮的含量 预览

摘要 以明日叶为原料,乙醇为提取溶剂,用高效液相色谱法测定明日叶中查尔酮含量。采用单因素和正交试验对料液比、乙醇浓度、提取时间和提取温度进行了考察。结果表明:对查尔酮的含量影响最大的是料液比,超声时间对明日叶中查尔酮含量的影响最低,乙醇浓度和超声温度对查尔酮含量影响不大。最佳工艺参数为乙醇浓度80%,提取温度80℃,料液比1∶30(g/mL),提取时间为30min,此时查尔酮得率为22.765mg/g。 The chalcone from Angelica keiskei was extracted by ethanol solution and determined by high performance liquid chromatography(HPLC).The single factor experiment and orthogonal test were studied for the influence degree of the solid-liquid ratio,ethanol concentration,extraction time and extraction temperature.The results showed that the ratio of solid to liquid had the greatest impac and the ultrasonic time had minimal impact and the concentration of ethanol and the ultrasonic temperature had little effect on the content of chalcone.The best conditions for the chalcone:the ethanol concentration was 80%,the best temperature was 80℃,the solid-liquid ratio was 1∶50(g/mL)and the optimum time was 20 min.Under these conditions,the yield of chalcone was 22.225 mg/g.
