摘要 氧化应激是血管损伤的重要因素,而血管受损后其收缩和舒张功能失调是血压异常的关键,因此阐明氧化应激在高血压发病中的具体作用及其机制对高血压的防治具有重要意义。血管氧化损伤与活性氧类(ROS)的过量生成密切相关,内皮型一氧化氮合酶(eNOS)作为血管内皮合成一氧化氮的关键酶,能拮抗ROS的生成、保护血管内皮。而eNOS活性受小凹蛋白、钙调蛋白、热激蛋白及血管紧张素Ⅱ等多种蛋白调节,其能使一氧化氮和ROS保持动态平衡。此外,血管促氧化酶系统和抗氧化酶系统是ROS生成、血管氧化损伤的又一重要因素。因此,通过调节血管氧化应激水平防治高血压可能成为未来的研究方向。 Oxidative stress is an important factor in vascular injury,and the dysfunction of systolic and diastolic function after vascular injury is the key to abnormal blood pressure.Therefore,to elucidate the role and mechanism of oxidative stress in the pathogenesis of hypertension,has great clinical significance for the prevention and treatment of hypertension.Vascular oxidative damage is closely related to the excess generation of reactive oxygen species(ROS).Endothelial nitric oxide synthase(eNOS)is a key enzyme in the synthesis of nitric oxide in vascular endothelial cells,which can antagonize the formation of ROS and protect the vascular endothelium.But the activity of eNOS is regulated by caveolin-1,calmodulin,heat shock protein and angiotensinⅡand other proteins,which can keep the dynamic balance of nitric oxide and ROS.In addition,the vascular pro-oxidant and antioxidant enzyme systems are another important factor in the formation of ROS and oxidative damage of blood vessels.Therefore,the prevention and treatment of hypertension by regulating the level of vascular oxidative stress may become a new research direction.
