黔产辣蓼的显微鉴别与金属元素的检测研究 预览

摘要 目的通过显微鉴别的方法对黔产辣蓼的根、茎、叶等显微特征进行对比研究,测定其金属元素的含量,为其质量控制及开发利用提供实验依据。方法采用常规的显微技术分别观察辣蓼的根、茎、叶的组织结构,同时采用电感耦合发射光谱法(ICP-OES)测定辣蓼中22种金属元素含量。结果鉴定了黔产辣蓼的显微特征,黔产辣蓼叶表面可见腺毛与非腺毛,气孔多为平轴式,偶见不等式或不定式,副保卫细胞常为2~4个。根横切面可见维管束为有限外韧型;茎横切面中柱鞘纤维束断续排列成环,韧皮部狭窄,髓部薄壁细胞内含草酸钙簇晶。检测出辣蓼中含有丰富的Ca、Mg、Al、K、Fe、Mn、Ag、Zn等元素,精密度RSD为0.244%~2.681%,辣蓼中的Pb、As、Cu、Hg、Cd等有害重金属元素含量均未超标,符合2015年版中国药典的质控标准。结论黔产辣蓼中有害重金属含量均符合《药用植物及制剂进出口绿色行业标准》要求,本实验从显微鉴定及重金属元素的测定方面为黔产辣蓼的药用安全性评价提供实验依据,同时也为辣蓼的综合开发利用提供理论基础。 Objective To study the microstructures of roots, stems, leaves and seeds of Polygonum flaccidum Meissn. by microscopic identification, and determine the contents of metal elements, to provide experimental basis for its quality control and development and utilization. Methods The microstructures of roots, stems and leaves of Polygonum flaccidum Meissn. were observed by conventional microscopic techniques. At the same time, 22 kinds of metal elements were determined by inductively coupled emission spectroscopy (ICP-OES). Results Glandular and non-glandular hairs could be seen on the surface of the leaves of Polygonum flaccidum Meissn., whose stomata were mostly flat-axis type, occasionally inequality or infinitive, and the number of the subsidiary cells was often 2-4. The vascular bundles were of limited external toughness in the cross section of the root;in the transverse section of stem, the columnar sheath fibrous bundles were arranged intermittently into rings, the phloem was narrow, and the calcium oxalate clusters were found in the medullary parenchyma cells. The results showed that Ca, Mg, Al, K, Fe, Mn, Ag and Zn were rich in Polygonum flaccidum Meissn., and the experimental precision RSD was 0.244 %-2.681 %;the contents of harmful heavy metals such as Pb, As, Cu, Hg and Cd did not exceed the standard, in line with the quality control standards listed in 2015 version of the Chinese Pharmacopoeia. Conclusion The content of harmful heavy metals in Polygonum flaccidum Meissn. produced in Guizhou meets the requirements of the Green Industry Standard for Import and Export of Medicinal Plants and Preparations. This study provides experimental basis for medicinal safety evaluation of Polygonum flaccidum Meissn. from Guizhou in terms of microscopic identification and determination of heavy metal elements. At the same time, it also provides a theoretical basis for the comprehensive development and utilization of Polygonum flaccidum Meissn.
