神经生长因子对新生大鼠高胆红素血症细胞凋亡以及相关基因的影响 预览

Effect of nerve growth factor on the apoptosis of cells and related genes in neonatal rats with hyperbilirubinemia

摘要 目的 研究神经生长因子对新生大鼠高胆红素血症细胞凋亡以及含半胱氨酸的天冬氨酸蛋白水解酶3(Caspase-3)、核因子-κB(NF-κB)水平的影响。方法 将105只SD新生大鼠随机分为对照组、模型组和干预组,每组35只。模型组、干预组大鼠1 h内腹腔注射剂量为100μg/g的胆红素溶液1次,对照组腹腔注射0.5 ml的生理盐水。通过腹腔注射100μg/g的胆红素溶液建立高胆红素血症大鼠模型。造模后,按50μg/kg腹腔注射神经生长因子,持续给药3 d。造模后12 h、24 h、36 h、48 h、72 h,观察三组大鼠神经行为的变化;脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL)测定神经细胞的凋亡率;免疫印迹试验(Western Blot)检测NF-κB、Caspase-3蛋白的表达水平。结果 与对照组相比,模型组大鼠活动明显减少,出现翻滚、抽搐、对外界刺激的反应迟钝等神经异常行为,细胞的凋亡率和Caspase-3蛋白水平显著增加(P﹤0.05),NF-κB水平显著增加(P﹤0.05);与模型组相比,干扰组大鼠活动增加,神经异常状况显著减轻,细胞的凋亡率、Caspase-3和NF-κB蛋白水平显著降低(P﹤0.05),且具有时间依赖性。结论 神经生长因子可能通过降低Caspase-3和NF-κB蛋白水平抑制神经细胞的凋亡,保护高胆红素血症大鼠的神经损伤。 Objective To investigate the effects of nerve growth factor on the apoptosis and the expression of caspase-3,NF-κB of neonatal rat with hyperbilirubinemia.Methods 105 SD newborn rats were randomly divided into control group,model group and intervention group,with 35 rats in each group.The model group and the intervention group were intraperitoneally injected with bilirubin solution with a dose of 100μg/g for 1 h,and the control group was intraperitoneally injected with 0.5 ml of normal saline.The rats model of hyperbilirubinemia were established by intraperitoneal injection of 100μg/g bilirubin.After modeling,the nerve growth factor was intraperitoneally injected at 50μg/kg.d by 3 d.At 12 h,24 h,36 h,48 h,72 h after modeling,the changes of neurobehavioral changes in each group were observed.The apoptosis rate of nerve cells was measured by terminal deoxynucleotidyl transferasemediated dUTP-biotin nick end labeling(TUNEL).The expression level of NF-κB and Caspase-3 protein were detected by Western Blot.Results Compared with the control group,significant neurobehavioral abnormalities were found in the rats of model group,cell apoptotic rate and the level of Caspase-3 protein were significantly increased(P﹤0.05),and a significant increase in NF-κB increased significantly(P﹤0.05).Compared with the model group,abnormal behavior of rats in intervention group were obviously improved,and the apoptosis rate,Caspase-3 and NF-κB protein levels were significantly decreased with time-dependent(P﹤0.05).Conclusion Nerve growth factor protect nerve injury in hyperbilirubinemia rats may by decreasing the protein level of Caspase-3 and NF-κB to inhibit cell apoptosis.
