基于网络药理学探究人参治疗心肌缺血的作用机制 预览 被引量:1

摘要 目的:利用网络药理学技术,探索人参改善心肌缺血的作用机制。方法:通过中草药综合数据库获取人参相关活性成分的化学结构;通过中草药活性成分数据库和治疗靶点数据库筛选人参活性成分的作用靶点,建立人参活性成分靶点数据集;通过人类孟德尔遗传综合数据库筛选心肌缺血相关的靶标,建立心肌缺血靶点数据集;通过蛋白质-蛋白质相互作用网络构建人参和心肌缺血的交互靶标;采用Cytoscape软件构建“成分-靶点-疾病”交互网络图,并进行基因本体(gene ontology,GO)富集分析及基于京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)的生物通路富集分析,研究人参改善心肌缺血的作用机制。结果:筛选出人参中59个活性成分,40个交互蛋白及15个人参与心肌缺血共同直接作用的蛋白靶点;GO分析条目24个,包括钙离子运输的负调节、外源性凋亡信号通路的负调控、活性氧代谢调节及一氧化氮生物合成过程等;KEGG信号通路43条,包括甲状腺激素信号通路、鞘脂信号通路、FcεRI信号通路、血管内皮生长因子信号通路及肿瘤坏死因子信号通路等。结论:本研究通过网络药理学初步揭示了人参改善心肌缺血的多维调控网络,为其的临床应用提供了理论依据. OBJECTIVE:To predict into the mechanism of ginseng in treatment of myocardial ischemia based on network pharmacology.METHODS:The chemical structure of ginseng related active ingredients was obtained from the comprehensive database of Chinese herbal medicine.Through the database of active ingredients of Chinese herbal medicine and the database of therapeutic targets,the active ingredient targets of ginseng were screened and the data set of active ingredient targets of ginseng was established.The target set of myocardial ischemia was established by screening the target of myocardial ischemia through Mendelian genetic database.The interaction targets of ginseng and myocardial ischemia were constructed by protein-protein interaction network.Cytoscape software was used to construct the"component-targeted disease-related"interaction network graph,and the gene ontology(GO)enrichment analysis and the biological pathway enrichment analysis based on Kyoto encyclopedia of genes and genomes(KEGG)were conducted to study the mechanism of ginseng in treatment of myocardial ischemia.RESULTS:Fifty-nine active ingredients,40 interactive proteins and 15 protein targets directly affected by myocardial ischemia in ginseng were screened.There were 24 GO analysis items,including negative regulation of calcium ion transport,negative regulation of exogenous apoptosis signal pathway,active oxygen metabolism regulation and nitric oxide biosynthesis process.There were 43 KEGG signaling pathways,including thyroid hormone signaling pathway,sphingolipid signaling pathway,FcεRI signaling pathway,vascular endothelial growth factor signaling pathway and tumor necrosis factor signaling pathway.CONCLUSIONS:This study preliminarily revealed the multi-dimensional regulatory network of ginseng in treatment of myocardial ischemia through network pharmacology,which can provide theoretical basis for its clinical application.
