不同药材中迷迭香酸甲酯及迷迭香酸的含量测定 预览

Quantification of Methyl Rosmarinate and Rosmarinic Acid in Different Traditional Chinese Medicine

摘要 建立了一种简便、快速、准确的LC-MS/MS法,对不同中药材中迷迭香酸甲酯及迷迭香酸的含量进行定量.通过单因素考察法建立了样品提取方法:向0.1 g 药材粉末中加入25 mL 50%甲醇,超声15 min进行提取.以0.1%甲酸-水和0.1%甲酸-乙腈为流动相,流速为300 μL/min,采用Waters BEH C18柱(2.1×50 mm, 1.7 μm)进行梯度洗脱,柱温为40 ℃,使用电喷雾离子源,在多反应监测模式下采用负离子扫描检测.迷迭香酸甲酯和迷迭香酸在线性范围0.1~100 ng/mL ( r =0.999)和0.1~500 ng/ml ( r =0.999)内,线性关系良好,重复性、日内和日间精密度的RSD<5%,加样回收率在90%~110%范围内,且RSD<5%,可用于迷迭香酸甲酯和迷迭香酸在植物中的含量测定. A simple, rapid and accurate LC-MS/MS method is established for quatification of methyl rosmarinate and rosmarinic acid in traditional Chinese medicine (TCM). The sample processing method is developed by single factor investigation as follows: 0.1 g TCM powder is extracted with 25 mL solution containing 50% methanol under ultrasonic condition for 15 min. The mobile phase consists of 0.1% formic acid-water and 0.1% formic acid-acetonitrile solution, and the flow rate is 300 μL/min. Separations are performed on a Waters BEH C18 column (2.1 × 50 mm, 1.7 μm) at 40 °C. The mass spectrometry is conducted with the electrospray ionization (ESI) source in negative ion mode. Methyl rosmarinate and rosmarinic acid in the range of 0.1-100 ng / mL (r =0.999) and 0.1-500 ng / mL ( r =0.999) show good linearity, respectively. The intra- and inter-day precision is consistent with the requirements of regulation and the RSD<5%. The range of recovery is 90%-110%, and the RSD<5%. The result indicates that this method can be applied to the quantification of methyl rosmainate and rosmarinic acid in TCM.
