目的探讨诱导型一氧化氮合酶(iNOS)、血小板衍生生长因子(PDGF)-B、脂多糖(LPS)在布-加综合征(BCS)大鼠模型中的表达及意义。方法部分结扎大鼠肝后段下腔静脉构建BCS模型。实验分为对照组(n=20)、模型组(n=20)和假手术组(n=20),各组又分4亚组(1、3、6、12周组)。收集肝组织作免疫组织化学、苏木精-伊红和Masson染色,检测i NOS、PDGF-B和LPS表达。结果模型组LPS值均显著高于对照组和假手术组(P=0.001)。模型组i NOS、PDGF-B mRNA和蛋白表达均显著高于对照组和假手术组(P=0.001);各亚组i NOS、PDGF-B mRNA和蛋白表达差异均有显著统计学意义(P=0.001)。模型组iNOS与PDGF-B、LPS呈正相关;肝纤维化与LPS呈正相关,与PDGF-B呈负相关。结论BCS病损与修复是一复杂过程,i NOS、PDGF-B和LPS可能分别在BCS病程不同阶段发挥侧重不同的作用,如何调节它们在纤维化中的平衡,也许是值得进一步研究的方向。
Objective To investigate the expression and significance of inducible nitric oxide synthase(i NOS),platelet-derived growth factor(PDGF)-B and lipopolysaccharide(LPS)in rat models with Budd-Chiari syndrome(BCS).Methods BCS model was established by partial ligation of inferior vena cava in the posterior segment of the liver.The experimental rats were divided into control group(n=20),model group(n=20)and sham group(n=20).Liver tissues were collected for immunohistochemistry,HE and Masson staining,and the expression levels of i NOS,PDGF-B and LPS were determined.Results The LPS value in model group was higher than that in both control group and sham group(P=0.001).The mRNA and protein expressions of i NOS and PDGF-B in model group were higher than those in both control group and sham group(P=0.001).Statistically significant differences in mRNA and protein expressions of i NOS and PDGF-B existed between each other among the subgroups(P=0.001).In model group i NOS was positively correlated with PDGF-B and LPS;liver fibrosis was positively correlated with LPS and negatively correlated with PDGFB.Conclusion The damage and repair of BCS is a complicated process.The i NOS,PDGF-B and LPS may play different roles in different stages of BCS.How to regulate their balance in liver fibrosis may be a direction that deserves further study.
