翠云草总黄酮对肺癌细胞生长的抑制作用 预览

摘要 目的:检测翠云草总黄酮对肺癌A549细胞和H460细胞生长的抑制作用。方法:翠云草总黄酮以5~40μg﹒mL^-1的浓度作用于A549细胞和H460细胞,以CCK8试剂检测细胞增殖,流式细胞仪检测细胞周期,免疫印迹检测ERK蛋白的表达和ERK的磷酸化水平。结果:翠云草总黄酮能抑制A549细胞和H460细胞的生长,抑制效果随着剂量的增加和作用时间的加长而增强;与对照组相比,翠云草总黄酮使A549细胞周期明显阻滞于G0/G1期,使H460细胞明显阻滞于S期和G2/M期;与对照组相比,翠云草总黄酮使A549细胞中p-ERK显著降低,使H460细胞中ERK和p-ERK均显著降低(P<0.05)。对于细胞周期和ERK蛋白的影响,10μg﹒mL^-1和20μg﹒mL^-1的翠云草总黄酮差异不显著(P>0.05)。结论:翠云草总黄酮对肺癌A549细胞和H460细胞的生长增殖、周期进程具有抑制作用;这种抑制作用可能与其降低ERK通路的活性有关。 Objective: To investigate the inhibition effect of total flavones of Cuiyuncao(TFS) on lung cancer A549 cells and H460 cells. Method: 5 ~ 40 μg﹒mL^-1 TFS was used on A549 cells and H460 cells. CCK8 reagent was used to test the growing inhibition rate of cells. Cell cycle was detected by flow cytometer. Western blotting was used to detect express of ERK protein and phosphorylated ERK protein. Result: TFS inhibited the growth of A549 and H460 cells. The inhibition effect increased with dose of TFS and time. Compared with control group, TFS caused A549 cells arrest at G0/G1 stage and H460 cells arrest at S and G2/M stages. Compared with the control group, TFS significantly decreased phosphorylated ERK in A549 cells while decreased both phosphorylated ERK and ERK in H460 cells(P<0.05). When used at concentration of 10 μg﹒mL^-1 and 20 μg﹒mL^-1, the effect on cell cycle and ERK protein was not significant(P>0.05). Conclusion: TFS has inhibition effect on lung cancer A549 cells and H460 cells through inhibiting cells growing and cell cycle progressing. The inhibition effect may be related to the down-regulation of ERK activation.
