右归饮诱导骨髓间充质干细胞修复兔激素型股骨头坏死的实验研究 预览

Experiment study on the repair of rabbit steroid femoral head necrosis with mesenchymal stem cells induced by Yougui Decoction

摘要 目的观察补肾中药右归饮诱导兔自体骨髓间充质干细胞修复兔激素型股骨头坏死的疗效,并探究其病理生理机制。方法将24只新西兰白兔随机分为右归饮组、激素对照组、空白对照组3组,每组8只。右归饮组及激素对照组使用马血清联合地塞米松注射制作早期股骨头坏死模型,造模开始4周后(第5周起)右归饮组予右归饮汤剂,每日10ml/kg灌胃,激素对照组、空白对照组均使用生理盐水,每日10ml/kg灌胃。每2周留取血液样本测定各组甘油三酯、总胆固醇含量及甲状腺激素水平。各组兔自第1次注射马血清(空白对照组为生理盐水)起分别于第10、14、18周末以过量麻醉方法处死各2只,行HE染色对照观察股骨头软骨下骨各细胞形态及空骨陷窝数目,同时分离纯化各组兔股骨髓腔内间充质干细胞,行MTT、ALP定量对比各组间间充质干细胞增殖及向成骨方向分化的能力。结果与激素对照组相比,各时间段右归饮组甘油三酯及总胆固醇含量均明显降低(P<0.05),甲状腺激素水平明显升高(P<0.05),空骨陷窝数明显较对照组减少而骨小梁数目明显较多(P<0.05),MTT法显示右归饮明显增强兔自体间充质干细胞增殖能力(P<0.05),ALP定量显示右归饮组与激素对照组相比明显具有更强的促成骨反应(P<0.05)。结论右归饮对兔激素性股骨头坏死股骨近端自体骨髓间充质干细胞的增殖和成骨活性具有较为明显的促进作用,且能显著降低该模型的血脂含量。 Objective To observe the curative effect of Yougui Decoction on induced rabbit autologous bone marrow mesenchymal stem cells( MSCS) to repair rabbit steroid femoral head necrosis,and to explore its mechanism. Methods24 New Zealand white rabbits were randomly divided into Yougui Decoction group,hormone control group and blank control group,8 cases in each group. The early femoral head necrosis model was made by using horse serum combined with dexamethasone injection in the Yougui Decoction group and the hormone control group. The model was given the Yougui Decoction every day for 10 ml/kg in the Yougui Decoction group after 4 weeks. In the blank control group and hormone control group,10 ml/kg of normal saline was filled intragastric daily. Blood samples were taken every two weeks to determine triglycerides,total cholesterol and thyroid hormone levels in each group. From the first injection of horse serum( the blank group was normal saline),two rabbits in each group were killed by excessive anesthesia at the weekend of 10 th,14 thand18 th,respectively. HE staining was used to observe the morphology of each cell and the number of hollow bone pits in the subtrochanteric osteochondral bone of the femoral head. At the same time,the rabbit femoral bone marrow mesenchymal stem cells in each group were isolated and purified. MTT and ALP quantitative were used to compare the proliferation and differentiation of mesenchymal stem cells. Results Compared with the hormone control group,the contents of triglycerides and total cholesterol were significantly decreased in the Yougui Decoction group at all times( P < 0. 05),while the thyroid hormone level was significantly increased( P < 0. 05). The number of hollow socket was significantly lower than that of the hormone control group,while the number of bone trabeculae was significantly higher( P < 0. 05).
