‘鸭梨’及其自交亲和性芽变‘金坠梨’花粉ABC转运蛋白家族分析 预览

Proteomic Analysis of ABC Transporters from Self-incompatible ‘Yali’ and Its Spontaneous Self-Compatible Mutant ‘Jinzhuili’

摘要 自交亲和性的‘金坠梨’是由自交不亲和性的‘鸭梨’芽变而来,为探究‘金坠梨’突变机理,综合分析了二者花粉蛋白质组差异表达蛋白质。在489个差异表达蛋白质中,发现了参与自交不亲和识别机制的ABC转运家族蛋白;KEGG富集分析发现,ABC转运蛋白被显著富集,其中PyABCB9-1,PyABCB9-2,PyABCC2,PyABCC3,PyABCD1,PyABCG31等6个蛋白显著下调;生物信息学分析表明,6个ABC转运蛋白不同家族在物种间具有很高的保守性;这6个显著下调蛋白可能影响了‘金坠梨’花柱S-RNase进入‘自己’花粉管的运输过程而导致其亲和性突变。 ‘Jinzhuili’ is a spontaneous self-compatible mutant of ‘Yali’. The mechanism of self-incompatibility in pear was studied at proteome level by using Isbaric Tags for Relative and Absolute Quantitation (iTRAQ). ABC transporters implicated in self-incompatibility recognition mechanism were found among 489 differentially expressed proteins. As a result, ABC transporters were significantly enriched by KEGG. And six down-regulated ABC transporters were obtained, including Py ABCB9-1, Py ABCB9-2, Py ABCC2, Py ABCC3, Py ABCD1 and Py ABCG31. Bioinformatics analysis showed that they had good conservativeness among species. These six ABC transporters might affect the transportation of S-RNase into the pollen tube of ‘Jinzhuili’, and then lead to self-compatible mutation in ‘Jinzhuili’.
