花姜酮对人绒毛膜癌JEG-3细胞增殖和细胞周期的影响 预览

摘要 目的研究花姜酮对人绒毛膜癌JEG-3细胞增殖和细胞周期的影响。方法应用终浓度为10、25、50μmol/L的花姜酮处理人绒毛膜癌JEG-3细胞24、48和72 h,对照组加入MEM(minimum Eaglemedium)完全培养液,CCK-8(Cell Counting Kit-8)检测细胞增殖,流式细胞术检测细胞周期。结果CCK-8检测细胞增殖结果显示,25μmol/L和50μmol/L花姜酮组与对照组比较,细胞增殖受到抑制,且具有浓度依赖性(P<0.05);50μmol/L花姜酮组随着时间的延长,细胞增殖受到抑制,具有时间依赖性(P<0.01)。流式细胞仪测定细胞周期结果显示,25μmol/L和50μmol/L花姜酮处理人绒毛膜癌JEG-3细胞72 h,G0/G1期峰值变低,细胞数量减少(P<0.01);G2/M期峰值变高,细胞数量增多(P<0.01),细胞周期阻滞在G2/M期。结论花姜酮通过抑制绒毛膜癌JEG-3细胞增殖及阻滞细胞周期从而抑制肿瘤的生长,花姜酮有望成为绒癌化疗方案的新型辅助治疗药物。 Objective To investigate the effects of Zerumbone(ZER)on proliferation and cell cycle of human choriocarcinoma JEG-3 cells.Methods Human choriocarcinoma JEG-3 cells were treated with ZER with a final concentration of 10,25 and 50μmol/L for 24 h,48 h and 72 h.ZER was completely replaced with MEM(minimum Eagle medium)in the control group.The methods of CCK-8(Cell Counting Kit-8)and flow cytometric were used to detect proliferation and cell cycle,respectively.Results The results of CCK-8 assay showed that ZER significantly inhibited the proliferation of JEG-3 cells in dose-dependent manner(P<0.05)and time-dependent manner(P<0.01).The results of flow cytometric method showed that the G0/G1 phase cells decreased(P<0.01),the G2/M phase cells increased(P<0.01),and cell cycle arrest was observed in G2/M phase after treatment of ZER.Conclusion ZER may inhibit the growth of tumor by inhibiting the proliferation and cell cycle in human choriocarcinoma JEG-3 cells.This study provides a new idea for the treatment of choriocarcinoma,and ZER is expected to be a new adjuvant therapy for choriocarcinoma chemotherapy in the future.
