bHLH转录因子在调节植物生长发育中起着重要作用。本研究利用转录组测序结果从栽培种花生丰花1号中克隆得到AhbHLH63基因。序列分析显示,AhbHLH63基因有两个剪接体AhbHLH63-1.1和AhbHLH63-1.2,其ORF分别长1 188 bp和1 152 bp,分别编码395个氨基酸和383个氨基酸组成的蛋白。亲缘关系分析表明,AhbHLH63蛋白含有一个预测的bHLH结构域,与其他植物的bHLH63具有较高的同源性,与苜蓿等的同源蛋白亲缘关系较近。实时荧光定量PCR分析表明,AhbHLH63-1.1和AhbHLH63-1.2表达模式基本一致,均为组成型表达,在花中表达量最高,茎和叶中次之,在根中表达量最低;在花生种子的不同发育时期,发育中期表达量较高。
bHLH transcription factors play the important roles in regulating plant growth and development. In this study, an AhbHLH63 gene was cloned from cultivated peanut variety Fenghua 1 based on the results of transcriptomic sequencing. Sequence analysis showed that the AhbHLH63 gene had two kinds of splicesome, AhbHLH63-1.1 and AhbHLH63-1.2, which had the open reading frame as 1 188 bp and 1 152 bp and encoded 395 and 383 amino acids respectively. Phylogenetic analysis indicated that the AhbHLH63 protein contained a predicted bHLH domain, which had high sequence identity with bHLH63 or homologous proteins from other plants, and had the highest similarity with that of alfalfa. Quantitative RT-PCR analysis showed that the expression profile of AhbHLH63-1.1 was consistent with that of AhbHLH63-1.2, which displayed the constitutive patterns. AhbHLH63 gene had the highest expression in flower, followed by stem and leaves, and the lowest in root. The highest expression level of AhbHLH63 gene appeared at the mid-developmental stage during the development of peanut seed.
