顺式和反式四苯乙烯衍生物的性质和应用研究

摘要 分别采用实验测定和理论计算比较了顺式和反式四苯乙烯类光敏剂(TPETH)产生单线态氧的能力。基于ABDA的实验结果表明顺式异构体比反式产生多10%的单线态氧。TD-DFT的理论计算表明顺式异构体的激发态S1与T1的能级差较小,容易发生体系间的能量传递,从而有利于单线态氧的产生。利用TPETH开发了一个靶向Caspase-8的荧光探针(ZH9-1)。并利用该探针评价了香豆素类化合物对Caspase-8的抑制活性,结果显示所选择的香豆素类化合物中,化合物28对Caspase-8表现出一定程度的抑制活性。首次研究了顺反异构体产生单线态氧的能力,为聚集诱导发光分子的进一步应用提供了参考。新开发的靶向Caspase-8的荧光探针将会为发现该酶的抑制剂提供帮助。 The obility of TPETH to produce singlet oxygen was compared by experimented and theoretical calculations.Based on ABDA measurement results,the cis isomer produced 10%more singlet oxygen than the trans isomer.The theoretical calculation using TD-DFT showed that singlettriplet energy gap of cis isomer was smaller than that of trans isomer,and the energy transfer between systems was easy to occur,which was conductive to generate more singlet oxygen.An AIEgen-based probe for Caspase-8 was synthesized to evaluate the activety of coumarins against Caspase-8.It turned out that the compound 28 had moderate inhibition towards Caspase-8.In this paper,the ability of cis and trans isomers to generate singlet oxygen was studied for the first time,which could help with the further application of luminescent molecules with the property of aggregation-induced emission.Caspase-8 targeting AIE probe could help to identify inhibitor of the protease.
