大灰藓与桧叶金发藓醇提物体外抗肝癌作用初步探究

Study on Anti-hepatocarcinoma Effects of Ethanol Extracts from Hypnum plumaeforme and Polytrichum juniperinum in Vitro

摘要 近年来,天然植物药的抗肿瘤作用受到研究者的广泛关注。苔藓作为植物界的一员,虽然中草药已有记载,但是对于其作用机理研究较少。因此,本研究初步探讨了两种苔藓醇提物的抗肝癌活性及作用机理。本研究制备了大灰藓醇提物(Hypnum plumaeforme Wils. ethanol extract, HPEE)和桧叶金发藓醇提物(Polytrichum juniperinum Hedw. ethanol extract, PJEE),检测了HPEE和PJEE的多糖、黄酮及三萜含量,及其体外抗肝癌活性。结果表明,HPEE多糖、黄酮及三萜含量分别为34.72%、6.57%及32.26%,PJEE多糖、黄酮及三萜含量分别为52.35%、1.11%及32.78%。HPEE及PJEE均显著抑制人肝癌细胞HepG2和BEL-7404及小鼠肝癌细胞H22的增殖(p<0.05),并呈剂量依赖性,HPEE及PJEE对HepG2细胞的IC50分别为100μg/m L和260μg/mL。采用Annexin V/PI双染检测细胞凋亡,结果显示HPEE及PJEE均能够显著诱导HepG2细胞的凋亡和坏死(p<0.05),并且低剂量和高剂量PJEE能够分别将HepG2细胞阻滞在G0/G1和G2/M期。以上结果表明HPEE及PJEE通过诱导细胞凋亡及细胞周期阻滞从而抑制肝癌细胞增殖。本研究为深入研究大灰藓及桧叶金发藓成分及抗肝癌机制提供理论依据,为有效开发苔藓资源提供理论指导。 In recent years,natural drugs from plant have attracted extensive attention in cancer treatment.Moss has been recorded in Chinese herb medicines,but few study have reported its mechanisms.In this study,the ethanol extracts of Hypnum plumaeforme(HPEE)and Polytrichm juniperinum(PJEE)were prepared to analyze the contents of polysaccharides,flavonoids and triterpenes and anti-hepatocarcinoma activities.The results showed that the contents of polysaccharide,flavonoid and triterpene were 34.72%,6.57%and 32.26%in HPEE,and52.35%,1.11%and 32.78%in PJEE,respectively.Both HPEE and PJEE significantly inhibited the proliferation of human hepatocarcinoma HepG2,BEL-7404 cells and murine hepatocarcinoma H22 cells(p<0.05)in a dose-dependent manner.The IC50 values of HPEE and PJEE on HepG2 were 100μg/m L and 260μg/m L,respectively.Annexin V/PI staining was used to detect apoptosis and the result showed that both HPEE and PJEE could significantly induced apoptosis and necrosis of HepG2 cells(p<0.05).Furthermore,PJEE at low dose and high dose could arrest HepG2 cells at G0/G1 and G2/M phase,respectively.The above results indicated that HPEE and PJEE inhibited hepatocarcinoma cell proliferation by the induction of apoptosis and cell cycle arrest.This study might provide a theoretical basis for further studies on the constituents and the mechanism of anti-hepatoma of Hypnum plumaeforme and Polytrichum juniperinum,and might offer theoretical guidance for the effective development of moss resources.
