基于脂肪组织细胞因子探讨黄芪葛根配伍对糖尿病大鼠降糖降脂的作用机制

摘要 目的:考察黄芪葛根配伍对脂肪细胞因子的调控作用,明确两药配伍降低血糖血脂的作用机制。方法:雄性SD大鼠随机分为:正常对照组、模型对照组、金芪降糖片1.47 g/kg组、黄芪2.7 g/kg组、葛根1.35 g/kg组、黄芪葛根汤4.05 g/kg组。一次性腹腔注射STZ诱导糖尿病模型。造模同日给予相应药物或蒸馏水10 ml/kg,连续灌胃30天。检测血糖(Glu)、三酰甘油(TG)、胆固醇(CHO),酶联免疫吸附实验(ELISA)检测脂联素(APN)、瘦素(LEP)、肿瘤坏死因子α(TNF-α)、白介素-12(IL-12)、IL-15含量,实时荧光定量PCR法(Real-time PCR)检测趋化素(chemerin)、趋化素受体(chemR23)mRNA相对表达量。结果:与正常对照组比较,造模30天模型对照组Glu、TG、CHO含量明显升高,APN、LEP含量明显降低,TNF-α、IL-12、IL-15含量增加,chemerin、chemR23mRNA表达上调。黄芪、葛根及其配伍能明显降低模型对照组大鼠Glu、TG、CHO(黄芪葛根配伍除外)、TNF-α(葛根除外)、IL-12(葛根除外)、IL-15、下调chemerin(黄芪葛根配伍除外)、chemR23 mRNA表达,黄芪葛根配伍降低糖尿病大鼠TG、IL-2作用优于单一用药,黄芪抑制TNF-α、IL-15分泌作用优于两药合用。黄芪、黄芪葛根配伍能升高模型对照组APN、LEP含量,但两药没有交互作用。结论:黄芪葛根配伍通过促进脂肪组织APN、LEP分泌,降低TNF-α、IL-12含量、下调chemR23 mRNA表达达到降低糖尿病大鼠血糖、三酰甘油的作用。 Objective:To observe the regulatory effect of Radix Astragali-Pueraria lobate Compatibility on cytokines in adipose tissue,and its mechanism of reducing blood glucose and lipids Method:Male SD rats were divided into normal group,model group,Jinqi Jiangtang 1.47 g/kg group,Radix Astragali 2.7 g/kg group,Pueraria lobata 1.35 g/kg group,Radix Astragali-Pueraria lobata Compatibility 4.05 g/kg group.Diabetic model was induced by intraperitoneal injection of STZ one time,on the same day,rats were given corresponding drugs or distilled water(10 ml/kg)for 30 days.Glu,Triglyceride(TG)and cholesterol(CHO)in serum were detected by biochemical method.The contents of adiponectin(APN),leptin(LEP),Tumor Necrosis Factorα(TNF-α),Interleukin 12(IL-12)and IL-15 were detected by enzyme linked immunosorbent assay(ELISA).The relative expression of chemerin and chemR23 mRNA was detected by Real-time PCR.Results:Compared with the normal group,the level of blood glucose,TG and CHO were increased in model group,the expression of APN and LEP were decreased,the contents of TNF-α,IL-12,IL-15 were increased,the expressions of chemerin and chemR23 mRNA were up regulated.In Radix Astragali group,Pueraria lobata group and their compatibility group,the contents of blood glucose,TG,CHO(except Astragali-Pueraria lobata Compatibility),TNF-α(except Pueraria lobata),IL-12(except Pueraria lobata),IL-15 were significantly decreased,expressions of chemerin(except Astragali-Pueraria lobata Compatibility)and chemR23 mRNA were 10 ml/kg down regulated.The compatibility group showed better effect than single drug in reducing TG and IL-12.The inhibitory effect of Radix Astragali on the expression of TNF-αand IL-15 was better than the Compatibility group.Radix Astragali group and Radix Astragali-Pueraria lobate Compatibility group could increase APN and LEP on model rats,but there was no interaction between the two drugs.Conclusion:Radix Astragali-Pueraria lobate Compatibility can reduce blood glucose and TG in diabetic rats by promoting the expression of
