比索洛尔对压力负荷诱导的小鼠心力衰竭的影响 认领

Effects of bisoprolol on mice of heart failure induced by pressure overload in vivo

摘要 目的研究比索洛尔对压力负荷诱导的小鼠心力衰竭(CHF)的影响及作用机制。方法按照体重将雄性C57小鼠随机分为3组:假手术组、模型组、实验组,每组20只。模型组和实验组小鼠采用主动脉缩窄术构建压力负荷诱导的小鼠心衰模型。小鼠在造模第4周后,实验组开始每日灌胃2 mg·kg^-1比索洛尔3 mL,连续给药4周;假手术组和模型组给予等量0.9%NaCl。脱氧核糖核苷酸末端转移酶介导的缺口末端标记染色试剂盒(TUNEL)染色检测小鼠心肌细胞凋亡,蛋白质印迹法检测B淋巴细胞瘤-2(Bcl-2)蛋白和Bcl-2相关X蛋白(Bax)的表达。结果假手术组、模型组和实验组大鼠心脏组织心肌细胞的凋亡率分别为(2.46±0.45)%,(31.15±2.56)%和(13.36±2.14)%;这3组的Bcl-2的相对表达分别为0.97±0.03,0.35±0.05和0.65±0.09;这3组的Bax蛋白的相对表达分别为1.01±0.03,4.45±0.19和1.58±0.04。上述指标:模型组与假手术组相比或实验组与模型组相比,差异均有统计学意义(均P<0.05)。结论比索洛尔可能通过调控Bcl-2、Bax的表达,抑制心肌细胞凋亡,来发挥对心脏的保护作用。 Objective To investigate effects and mechanism of bisoprolol on chronic heart failure(CHF)induced by pressure overload in vivo in mice.Methods Male C57 mice were randomly divided into three groups:sham operation group,model group,experimental group according to weight,with 20 mice in each group.CHF mice model was constructed through transverse aortic constriction method in the model group and the experimental group.After the 4 th week of modeling,experimental group began to administer 2 mg·kg^-1 bisoprolol per day,3 mL of gastric perfusion,continuous administration for 4 weeks;the same amount of normal saline was given to sham operation group and model group.The deoxyribonucleotide terminal transferase-mediated nick labeling kit staining was used to detect the apoptosis of mouse cardiomyocytes.Western blot was used to detect the relative expression of B-cell lymphoma-2(Bcl-2)protein and Bcl-2 associated X protein(Bax).Results The apoptotic rates of cardiomyocytes in sham operation group,model group and experimental group were(2.46±0.45)%,(31.15±2.56)%and(13.36±2.14)%,respectively;the relative expression of Bcl-2 protein in the three groups were 0.97±0.03,0.35±0.05 and 0.65±0.09,respectively;the relative expressions of Bax protein in the three groups were 1.01±0.03,4.45±0.19 and 1.58±0.04,respectively.Comparison between model group and sham operation group,or between experimental group and model group,the differences of the factors were statistically significant(all P<0.05).Conclusion Bisoprolol may play a protective role in the heart by regulating the expression of Bcl-2 and Bax and inhibiting the apoptosis of cardiomyocytes.
