模拟失重48h后血管内皮细胞长链非编码RNA的差异表达分析

Differential Expression Analysis of Long Non-coding RNAs in Human Vascular Endothelial Cells after 48h Simulated Weightlessness

摘要 目的探究模拟失重48h后人脐静脉内皮细胞(HUVECs)中长链非编码RNA(lncRNAs)的表达差异。方法采用高通量测序技术检测HUVECs模拟失重48h组与水平静置对照组中lncRNAs的差异表达,对部分差异表达的lncRNAs进行PCR验证。结果差异表达lncRNAs共37个,模拟失重48h处理后,有22个lncRNAs显著上调(P<0.05),15个lncRNAs显著下调(P<0.05)。PCR结果表明,有4个lncRNAs的变化趋势与测序结果一致。结论模拟失重48h后HUVECs中lncRNAs的表达发生明显变化,lncRNAs的差异表达可为探索模拟失重下血管内皮细胞功能改变的发生机制提供依据。 Objective To explore the differential expression of long non-coding RNAs(lncRNAs)in human umbilical vein endothelial cells(HUVECs)after 48 hsimulated weightlessness.Methods High throughput sequencing technology was used to detect the differential expression of lncRNAs between48 hsimulated weightlessness groups and horizontal standing control groups.Expression levels of some differentially-expressed lncRNAs were validated by PCR.Results A total of 37 differentially-expressed lncRNAs were identified.After 48 hsimulated weightlessness,22 lncRNAs were up-regulated(P<0.05)and 15 down-regulated significantly(P<0.05).PCR analysis of four selected lncRNAs confirmed the results of RNA-sequencing.Conclusion The expression levels of lncRNAs in HUVECs are altered significantly after 48 hsimulated weightlessness.Differentially-expressed lncRNAs may provide evidence for exploring mechanisms of functional changes in human vascular endothelial cells under simulated weightlessness.
