痛经安汤对原发性痛经模型大鼠疼痛、雌二醇及孕酮影响的研究

Tongjing’an Tang Has Effect on Pain,Estradiol and Progesterone in Rats with Primary Dysmenorrhea

摘要 目的:研究痛经安汤治疗原发性痛经模型大鼠的作用机制,为临床治疗该病寻求新的实验依据。方法:将60只大鼠随机分为空白组、模型组、阳性药组、痛经安汤低、中、高剂量组,每组10只。除空白组外,其余各组大鼠每天皮下注射己烯雌酚,连续10天,第10天根据分组每天灌胃相应药物45 min,空白组和模型组每天灌胃生理盐水10 m L/kg,阳性药组灌胃田七痛经胶囊混悬液1.6 g/(kg·d),痛经安汤低、中、高剂量组分别灌胃7.2 g/(kg·d),14.4 g/(kg·d),28.8 g/(kg·d)的痛经安汤水煎液。用缩宫素制备大鼠原发性痛经模型,观察各组大鼠30 min内的扭体反应。用放射免疫法测定血清雌二醇(Serum estradiol,E2)、孕酮(Serum progesterone,P)、血浆β-内啡肽(Plasmaβ-endorphin,β-EP)含量。结果:与空白组比较,模型组平均扭体次数、E2、P、β-EP水平均偏高(P <0.05,P <0.01);与模型组比较,阳性药组、痛经安汤各剂量组平均扭体次数、E2水平均偏低(P <0.05,P <0.01),阳性药组、痛经安汤各剂量组血清P水平均升高(P <0.05,P <0.01),其中中剂量组血清P水平最高;阳性药组β-EP降低(P <0.05),痛经安汤各剂量组β-EP均升高(P <0.01)。结论:痛经安汤能调节原发性痛经模型大鼠性激素(E2、P)水平并提高血浆中β-EP水平,提示痛经安汤可通过调节机体内分泌、改善机体内环境,发挥缓解痛经的作用。 Objective:To study the mechanism of Tongjing’an tang in the treatment of primary dysmenorrhea in rats,and to seek a new experimental basis for the clinical treatment of this disease.Methods:A total of 60 rats were randomly divided into the blank group,the model group,the positive medicine group,the Tongjing’an tang groups of low-dose,mediumdose and high-dose,10 rats in each group.Except those in the blank group,the rats in the other groups were given subcutaneous injection of diethylstilbestrol for ten days.On the 10 th day,after given corresponding medicine by gavage for 45 minutes per day according to the grouping,the blank group and model group were given 10 mL/kg of saline by gavage per day;the positive medicine group was given 1.6 g/(kg·d)of suspension of Tianqi Tongjing capsules by gavage;the Tongjing’an tang groups of low-dose,medium-dose and high-dose were given 7.2 g/(kg·d),14.4 g/(kg·d)and 28.8 g/(kg·d)of water decoction of Tongjing’an tang respectively.The rat models with primary dysmenorrhea were established by oxytocin,and the writhing response in 30 minutes in each group was observed.The contents of serum estradiol(E2),serum progesterone(P)and plasmaβ-endorphin(β-EP)were measured through radioimmunoassay.Results:Compared with those in the blank group,the average writhing times as well as the levels of E2,P andβ-EP in the model group were higher(P<0.05,P<0.01);compared with those in the model group,the average writhing times and the levels of E2 were lower in the positive medicine group and the Tongjing’an tang groups of each dose respectively(P<0.05,P<0.01),and the levels of P in serum were increased in the above groups respectively(P<0.05,P<0.01),among which the one in the Tongjing’an tang group of medium-dose was the highest;β-EP in the positive medicine group was decreased(P<0.05),and that in the Tongjing’an tang groups of each dose was increased respectively(P<0.01)Conclusion:Tongjing’an tang can regulate the levels of sex hormones including E2 and P in model rats with pr
