The microbial consortium GF-20(GF-20) can efficiently decompose corn stover at low temperatures. The present study explored the key microbes of GF-20 and evaluated different culture conditions on its composition stability to promote the utilization of corn stover decomposing microbes in low temperature regions. GF-20 was subcultured to the 15 th generation under different temperatures, pHs, carbon, and nitrogen sources. Then, the dynamics of fermenting pH, cellulose enzyme activities, carbohydrate concentration, and oxidation reduction potential were determined to estimate the degradation efficiency of corn stover with GF-20. Furthermore, the structural stability and functional microbes of GF-20 were identified on the basis of PCR-denaturing gradient gel electrophoresis(DGGE) profiling and principal component analysis. The results showed that the offspring of GF-20 subcultured under different temperatures(4–30°C) and pH(6.0–9.0) conditions maintained stable growth, decomposition function, and composition structure. Furthermore, consortia GF-20 had a stable composition structure, which induced GF-20 to secrete cellulose and promote substrate decomposition as corn stover and ammonium were used as sources of carbon and nitrogen, respectively. According to the PCR-DGGE profiles, the key strains of GF-20 were determined to be Bacillus licheniformis, Cellvibrio mixtus subsp. mixtus, Bacillus tequilensis, Clostridium populeti, and Clostridium xylanolyticum. 展开更多

Six different environmental samples were applied to enrich microbial consortia for efficient degradation of corn stalk,under the thermophilic and mesophilic conditions.The consortium obtained from anaerobic digested sludge under thermophilic condition(TC-Y)had the highest lignocellulose-degrading activity.The CO2yield was 246.73 m L/g VS in23 days,meanwhile,the maximum CO2production rate was 15.48 mL/(CO2·d),which was28.75%and 52.27%higher than that under mesophilic condition,respectively.The peak value of cellulase activity reached 0.105 U/mL,which was at least 34.61%higher than the other groups.In addition,49.5%of corn stalk was degraded in 20 days,moreover,the degradation ratio of cellulose,hemicellulose and lignin can reach 52.76%,62.45%and42.23%,respectively.Microbial consortium structure analysis indicated that the TC-Y contained the phylum of Gemmatimonadetes,Acidobacteria,Chloroflexi,Planctomycetes,Firmicutes,and Proteobacteria.Furthermore,the Pseudoxanthomonas belonging to GammaProteobacteria might be the key bacterial group for the lignocellulose degradation.These results indicated the capability of degrading un-pretreated corn stalk and the potential for further investigation and application of TC-Y. 展开更多

A great deal of oil contaminated the shoreline by the Qingdao oil pipeline explosion in 2013. The four oildegrading consortia were enriched from sediment samples with crude oil as sole carbon and energy sources. The biodiversity and community analysis showed that the Luteibacter, Parvibaculum and a genus belonging to Alcanivoracaceae were found predominant bacteria in the four consortia, which belonged to Proteobacteria. Nine strains exhibiting distinct 16S rRNA gene sequences were isolated from the consortia. These strains were identified to eight genera based on 16S rRNA gene sequences. Five of the nine strains degraded more than 30% of the crude oil in two weeks by gravimetric method. From the analysis of GC-MS, most of the isolated strains tended to degrade n-alkanes rather than PAHs. Five strains showed high degrading ability of the total n-alkanes. Only Strain D2 showed great PAHs degrading ability and the degrading rates ranged from 34.9% to 77.5%. The sequencing analysis of the oil-degrading consortia confirmed that the genus of Alcanivorax was one of the dominant bacteria in Consortia A and E and Strain E4 might be one of the dominant bacteria. The strains obtained in this study demonstrated the potential for oil bioremediation in oil-contaminated beach ecosystems. 展开更多

This paper focuses on how to use consortium blockchain to improve the regulation of peer-to-peer(P2 P) lending market. The partial decentralized consortium blockchain with limited pre-set nodes can well improve transparency and security, which is suitable for financial regulation. Considering irregularities of the P2P lending market, the Hyperledger-based Peer-to-Peer Lending System(HyperP2PLS) is proposed. First elaborate the application scenario and business logic of the system, where a national P2P Lending Trading Center will be established to integrate all transactions and information of P2P lending market. Then construct the system architecture consisting of the blockchain network, HTTP server, and applications. The algorithm of implementation process and the web application for users have been well illustrated. The performance analysis shows that HyperP2PLS can guarantee the reliability, safety, transparency and efficiency. 展开更多

混合菌降解柴油效能及协同机制研究

从南京某炼油厂污泥中筛选出4株高效柴油降解菌,分别命名为KX-1、KX-3、KX-5和KX-12。其中,KX-1和KX-5为产生物表面活性剂菌株。将4株菌两两组合,结果表明第4组混合菌(克雷伯氏菌KX-3和铜绿假单胞菌KX-5)的7 d柴油降解率最高,达54.6%... 从南京某炼油厂污泥中筛选出4株高效柴油降解菌,分别命名为KX-1、KX-3、KX-5和KX-12。其中,KX-1和KX-5为产生物表面活性剂菌株。将4株菌两两组合,结果表明第4组混合菌(克雷伯氏菌KX-3和铜绿假单胞菌KX-5)的7 d柴油降解率最高,达54.6%。在单菌和混合菌的14 d降解过程中,通过测试OD600值、烃剩余量、菌落数、疏水性、乳化性及表面活性剂产量等指标并采用GC-MS分析,得出混合菌的协同降解机制为:(1)产生物表面活性剂菌KX-5所产生的表面活性剂显著提高了疏水性和乳化性,促进了细胞的生长与柴油的降解;(2)菌株KX-3和KX-5对不同碳数烷烃的降解能力表现出一定的互补性。生化试验和FTIR测试表明该表面活性剂为鼠李糖脂。 展开更多

An active mesophilic lignocellulose degrading microbial consortium,designated LZF-12,was bred from humus-rich soil by successive subcultivation under facultative aerobic static condition.Batch experiments were performed to investigate the structural and functional stability of lignocellulose degradation of rice straw of 10 g·L-1.The results showed that efficient degradation of rice straw(>70%)could be achieved and acetic acid concentration accounted for over 70%of total aqueous products from different generations by microbial consortium LZF-12 within 7 days.Denaturing gradient gel electrophoresis(DGGE)and sequencing of 16S rDNA sequences amplified from the total consortium DNA representing the presence of sequences were related to those of Clostridium,Clostridium cellulolyticum,Pseudomonas,Acetivibrio and some uncultured bacteria in LZF-12.DGGE pattern profiles from different LZF-12 generations were reproducible,suggesting the relative stabilities of the microbial community structure and succession mechanism in the established consortium. 展开更多

为明确高效纤维素降解复合菌系降解过程中微生物群落结构的变化规律及关键的降解功能菌,利用该复合菌系对滤纸和稻秆进行生物处理,通过底物降解、微生物生长量、发酵液pH的变化情况,选择不同降解时期复合菌系提取的总DNA进行细菌16S rRN... 为明确高效纤维素降解复合菌系降解过程中微生物群落结构的变化规律及关键的降解功能菌,利用该复合菌系对滤纸和稻秆进行生物处理,通过底物降解、微生物生长量、发酵液pH的变化情况,选择不同降解时期复合菌系提取的总DNA进行细菌16S rRNA基因扩增子高通量测序。通过分解特性试验确定在接种后培养第12、72、168 h分别作为降解初期、高峰期、末期。该复合菌系分别主要由1个门、2个纲、2个目、7个科、11个属组成。随着降解的进行,短芽胞杆菌属Brevibacillus、喜热菌属Caloramator的相对丰度逐渐降低;梭菌属Clostridium、芽胞杆菌属Bacillus、地芽胞杆菌属Geobacillus、柯恩氏菌属Cohnella的相对丰度逐渐升高;解脲芽胞杆菌属Ureibacillus、泰氏菌属Tissierella、刺尾鱼菌属Epulopiscium在降解高峰期时相对丰度最高;各时期类芽胞杆菌属Paenibacillus、瘤胃球菌属Ruminococcus的相对丰度无明显变化。上述11个主要菌属均属于厚壁菌门,具有嗜热、耐热、适应广泛pH、降解纤维素或半纤维素的特性。好氧型细菌是降解初期的主要优势功能菌,到中后期厌氧型细菌逐渐增多,并逐步取代好氧型细菌成为降解纤维素的主要细菌。 展开更多

To achieve higher energy utilization and lower generation cost for renewable sources(e.g., wind and solar energy), much work has been focused on demand response in smart grid(SG). Nonetheless, most existing studies consider energy trading with utility company which results in high energy loss from high voltage to low voltage and privacy leakage. Besides, there are relatively few researches devoted to electricity scheduling and price optimum among households without a third party. To cope with these issues, a novel deep deterministic policy gradient(DDPG)-based energy trading method with consortium blockchain(DETCB) is introduced. Firstly, in order to enhance system security, executing energy transaction among households is on the basis of consortium blockchain, which leads to not only anonymous trade but also public account. Moreover, the primary target from the aspect of the system is apparently the maximal social welfare, thus exploiting an iterative decision-making method combined with DDPG algorithm by non-profit controllers to obtain optimal trading prices and carry out optimal electricity allocation. To this end, security analysis demonstrates that DETCB contributes to creating a secure, stable and trustful environment. Furthermore, the excellent performance concerning social welfare, algorithm efficiency, and transaction energy sum is shown by numerical results. 展开更多
