BACKGROUND Enteropathy associated T-cell lymphoma (EATL) is a rare form of peripheral Tcell lymphoma and makes up less than 5% of gastrointestinal lymphomas. EATL can be divided into type 1 which is associated with celiac disease, and monomorphic epitheliotropic intestinal T-cell lymphoma (MEITL), formally type 2, which is not associated with celiac disease. CASE SUMMARY We present a 60-year-old African American female, without celiac disease, who presented with abdominal pain, diarrhea, and 30 lb. weight loss over a 3 month period. She was subsequently diagnosed with EATL throughout her entire gastrointestinal tract. She is currently undergoing chemotherapy with EOCH (Etoposide, Oncovin, Cyclophosphamide, and Hydroxydaunorubicin). EATL is most common in the Asian and Hispanic population yet the incidence in African Americans is uncertain and emphasizes the rarity of this case. A literature review was included to further emphasize similarities and differences between our case and previously reported cases of MEITL. CONCLUSION The patient was diagnosed with EATL, immunochemical testing was not conclusive for MEITL however was suggestive of the disease. 展开更多

Adult T-cell leukemia(ATL)is a mature T-cell malignancy caused by human T-cell leukemia virus type I infection,and 10%-25%of patients show central nervous system(CNS)involvement.CNS involvement significantly reduces survival and there are no effective treatments for CNS involvement.Therefore,an appropriate animal model is required to evaluate the inhibitory effects of novel drugs on the progression of ATL with CNS involvement.Here,we established a mouse model of ATL with CNS involvement using NOD.Cg-Prkdc scid Il2rg tm1Wjl/SzJ mice inoculated with ATL cells intramuscularly in the postauricular region,and these mice showed paraparesis.Of the 10 mice inoculated with ATL cells intramuscularly(I.M.)at 5 weeks of age,8(80%)showed paraparesis,whereas none of the 10 mice inoculated with ATL cells subcutaneously(S.C.)showed paraparesis.In the I.M.group,PCR detected HTLV-1-specific genes in the thoracic and lumbar vertebrae;however,in the S.C.group,the vertebrae were negative for HTLV-1 genes.Histological analysis revealed a particularly high incidence of tumors,characterized by accumulation of the injected cells,in the thoracic vertebrae of mice in the I.M.group.Tumor cell infiltration was relatively high in the bone marrow.Spinal cord compression caused by invasion of the tumor mass outside the pia mater was observed in the thoracic vertebrae of the spinal cord.In conclusion,we have reported a mouse model of tumor growth with paraparesis that may be used to assess novel therapeutic agents for ATL with CNS involvement. 展开更多

Synergistic effects of combined therapy of curcumin and Fructus Ligustri Lucidi for treatment of osteoporosis: cellular and molecular evidence of enhanced bone formation

OBJECTIVE: The present study explored the effects of the combined herbal therapy consisting of curcumin (CUR) and Fructus Ligustri Lucidi (FLL) on aspects of bone regeneration. METHODS: Prior to analyzing the ability of this novel combined herbal therapy to promote aspects of bone regeneration, its cytotoxicity was determined using MC3T3-E1 cells (pre-osteoblast model). Cell proliferation was evaluated using phase-contrast microscopy and cell differentiation was estimated using alkaline phosphatase activity. The effect of the combined herbal therapy (CUR + FLL) was also assessed in terms of mineralization in the extracellular matrix (ECM) of cultured cells. Further, to explore the molecular mechanisms of bone formation, time-dependent expression of bone-regulating protein biomarkers was also evaluated. RESULTS: Combined herbal therapy (CUR + FLL) significantly upregulated the viability, proliferation and differentiation of MC3T3-E1 cells compared to the monotherapy of CUR or FLL. The magnitude of ECM mineralization (calcium deposition) was also higher in MC3T3-E1 cells treated with combined therapy. The time-dependent expression of bone-forming protein biomarkers revealed that the tendency of expression of these bone-regulating proteins was remarkably higher in cells treated with combined therapy.CONCLUSION: The co-administration of CUR and FLL had superior promotion of elements of bone regeneration in cultured cells, thus could be a promising alternative herbal therapy for the management of bone erosive disorders such as osteoporosis. 展开更多

BACKGROUND Pluripotent stem cell-derived cardiomyocytes(CMs)have become one of the most attractive cellular resources for cell-based therapy to rescue damaged cardiac tissue.AIM We investigated the regenerative potential of mouse embryonic stem cell(ESC)-derived platelet-derived growth factor receptor-α(PDGFRα)^+cardiac lineagecommitted cells(CLCs),which have a proliferative capacity but are in a morphologically and functionally immature state compared with differentiated CMs.METHODS We induced mouse ESCs into PDGFRα^+CLCs andαMHC^+CMs using a combination of the small molecule cyclosporin A,the rho-associated coiled-coil kinase inhibitor Y27632,the antioxidant Trolox,and the ALK5 inhibitor EW7197.We implanted PDGFRα^+CLCs and differentiatedαMHC+^CMs into a myocardial infarction(MI)murine model and performed functional analysis using transthoracic echocardiography(TTE)and histologic analysis.RESULTS Compared with the untreated MI hearts,the anterior and septal regional wall motion and systolic functional parameters were notably and similarly improved in the MI hearts implanted with PDGFRα^+CLCs andαMHC+CMs based on TTE.In histologic analysis,the untreated MI hearts contained a thinner ventricular wall than did the controls,while the ventricular walls of MI hearts implanted with PDGFRα^+CLCs andαMHC^+CMs were similarly thicker compared with that of the untreated MI hearts.Furthermore,implanted PDGFRα+CLCs aligned and integrated with host CMs and were mostly differentiated intoα-actinin^+CMs,and they did not convert into CD31^+end othelial cells orαSMA^+mural cells.CONCLUSION PDGFRα^+CLCs from mouse ESCs exhibiting proliferative capacity showed a regenerative effect in infarcted myocardium.Therefore,mouse ESC-derived PDGFRα^+CLCs may represent a potential cellular resource for cardiac regeneration. 展开更多

Alcohol, a widely abused drug, has deleterious effects on the immature nervous system. This study investigates the effect of chronic in vitro ethanol exposure on the metabolism of immature rat cerebellar granular cells (CGCs) and on their response to oxygen-glucose deprivation (OGD). Primary CGC cultures were exposed to ethanol (100 mM in culture medium) or to control ethanol-free medium starting day one in vitro (DIV1). At DIV8, the expression of ATP synthase gene ATP5g3 was quantified using real-time PCR, then cultures were exposed to 3 hours of OGD or normoxic conditions. Subsequently, cellular metabolism was assessed by a resazurin assay and by ATP level measurement. ATP5g3 expression was reduced by 12-fold (P = 0.03) and resazurin metabolism and ATP level were decreased to 74.4±4.6% and 55.5 ±6.9%, respectively after chronic ethanol treatment compared to control values (P<0.01). Additionally, after OGD exposure of ethanol-treated cultures, resazurin metabolism and ATP level were decreased to 12.7±1.0% and 9.0±2.0% from control values (P<0.01). These results suggest that chronic ethanol exposure reduces the cellular ATP level, possibly through a gene expression down-regulation mechanism, and increases the vulnerability to oxygen-glucose deprivation. Thus, interventions which improve metabolic function and sustain ATP-levels could attenuate ethanol-induced neuronal dysfunction and should be addressed in future studies. 展开更多

Liver transplant(LT)is the primary treatment for patients with end-stage liver disease.About 25000 LTs are performed annually in the world.The potential for intraoperative bleeding is quite variable.However,massive bleeding is common and requires blood transfusion.Allogeneic blood transfusion has an immunosuppressive effect and an impact on recipient survival,in addition to the risk of transmission of viral infections and transfusion errors,among others.Techniques to prevent excessive bleeding or to use autologous blood have been proposed to minimize the negative effects of allogeneic blood transfusion.Intraoperative reinfusion of autologous blood is possible through previous selfdonation or blood collected during the operation.However,LT does not normally allow autologous transfusion by prior self-donation.Hence,using autologous blood collected intraoperatively is the most feasible option.The use of intraoperative blood salvage autotransfusion(IBSA)minimizes the perioperative use of allogeneic blood,preventing negative transfusion effects without negatively impacting other clinical outcomes.The use of IBSA in patients with cancer is still a matter of debate due to the theoretical risk of reinfusion of tumor cells.However,studies have demonstrated the safety of IBSA in several surgical procedures,including LT for hepatocellular carcinoma.Considering the literature available to date,we can state that IBSA should be routinely used in LT,both in patients with cancer and in patients with benign diseases. 展开更多

Objective:To investigate the potential antifibrotic mechanisms of Chinese medicine Ganshuang Granules (肝爽颗粒,GSG)and to provide clinical therapeutic evidence of its effects.Methods:A cirrhotic mouse model was established by intraperitoneally injecting a mixture of CCl4(40%)and oil (60%)at 0.2mL per 100g of body weight twice a week for 12 weeks.After 12-week modeling,GSG was intragastric administrated to the mice for 2 weeks,and the mice were divided into low-,medium-and high-dose groups at doses of 1,2 and 4g/(kg·day), respectively.Liver morphology changes were observed using Masson's trichrome staining and B-ultrasound. The levels of alanine aminotransferase (ALT),aspartate aminotransferase (AST)and hyaluronic acid (HA)in serum were detected using an automatic biochemistry analyzer.The expressions of desmin,smooth muscle actin (SMA)and Foxp3 in liver were detected by immunofluorescence.The regulatory T cell (Treg)frequency was determined through flow cytometry analysis.Collagen-Ⅰ,SMA,IL-6,tumor necrosis factor α (TNF-α), interleukin (IL)-1β and transforming growth factor β1(TGF-β1)expression levels were measured using quantitative polymerase chain reaction (qPCR).Results:Masson's staining result showed fewer pseudolobule structures and fibrous connective tissue in the GSG-treatment groups than in the spontaneous recovery group. Ultrasonography showed that GSG treatment reduced the number of punctate hyperechoic lesions in mice cirrhotic livers.The serum ALT,AST,HA levels were significantly ameliorated by GSG treatment (ALT:F=8.104, P=0.000;AST:F=7.078,P=0.002;and HA:F=7.621,P=0.001).The expression levels of collagen-Ⅰ and SMA in the cirrhotic livers were also attenuated by GSG treatment (collagen-Ⅰ :F=3.938,P=0.011;SMA:F=4.115, P=0.009).Tregs,which were elevated in the fibrotic livers,were suppressed by GSG treatment (F=8.268, P=0.001).The expressions of IL-6,TNF-α and IL-1β increased,and TGF-β levels decreased in the cirrhotic livers after GSG treatment (IL-6:F=5.457,P=0.004;TNF-α:F=6.023 展开更多

AIM:To assess helper T(Th) lymphocyte subset balance in patients with Vogt-Koyanagi-Harada(VKH) disease. METHODS:Sixty-eight active VKH patients and seventytwo inactive VKH patients were included in this study. One hundred healthy individuals served as controls. Peripheral blood was obtained from VKH patients and healthy controls. Th lymphocyte subsets were analyzed by flow cytometry. Plasma concentration of interleukin(IL)-17, IL-10, transforming growth factor(TGF)-β, IL-23 and IL-6 was examined by enzyme-linked immunosorbent assay(ELISA). RESULTS:VKH patients with active uveitis had significantly higher percentages of both Th1 and Th17 cells and lower percentages of regulatory T(Treg) cells as compared with inactive VKH patients and healthy controls. Th1/Th2 and Th17/Treg ratios were also significantly elevated in active VKH patients. The percentages of Th1, Th17 and Treg cells and the Th1/Th2, Th17/Treg ratio did not differ between inactive VKH patients and healthy controls. There was no difference concerning the percentage of Th2 cells among all the groups. VKH patients with active uveitis showed an elevated level of peripheral Th17 related cytokines levels(TGF-β, IL-6, IL-23, and IL-17) and a decreased level of Treg related cytokines(IL-10) compared with inactive VKH patients and healthy controls. Inactive VKH patients showed no differences in peripheral Th17 related cytokines(TGF-β, IL-6, IL-23, and IL-17) and Treg related cytokines(IL-10) levels compared with healthy controls. CONCLUSION:Th1 and Th17 cells are significantly increased and Treg cells significantly decreased in active VKH compared with inactive VKH or healthy controls. Therefore, Th lymphocyte subset analysis may serve as a disease biomarker for VKH. 展开更多

Bisperoxo(1,10-phenanthroline)oxovanadate(BpV)can reportedly block the cell cycle.The present study examined whether BpV alters gene expression by affecting DNA methyltransferases(DNMTs),which would impact the cell cycle.Immortalized mouse hippocampal neuronal precursor cells(HT22)were treated with 0.3 or 3μM BpV.Proliferation,morphology,and viability of HT22 cells were detected with an IncuCyte real-time video imaging system or inverted microscope and 3-(4,5-dimethylthiazol-2-yl)-5(3-carboxymethonyphenol)-2-(4-sulfophenyl)-2H-tetrazolium,respectively.mRNA and protein expression of DNMTs and p21 in HT22 cells was detected by real-time polymerase chain reaction and immunoblotting,respectively.In addition,DNMT activity was measured with an enzyme-linked immunosorbent assay.Effects of BpV on the cell cycle were analyzed using flow cytometry.Results demonstrated that treatment with 0.3μM BpV did not affect cell proliferation,morphology,or viability;however,treatment with 3μM BpV decreased cell viability,increased expression of both DNMT3B mRNA and protein,and inhibited the proliferation of HT22 cells;and 3μM BpV also blocked the cell cycle and increased expression of the regulatory factor p21 by increasing DNMT expression in mouse hippocampal neurons. 展开更多

BACKGROUND An in vitro injury model mimicking a corneal surface injury was optimised using human corneal epithelial cells (hCEC). AIM To investigate whether corneal-stroma derived stem cells (CSSC) seeded on an amniotic membrane (AM) construct manifests an anti-inflammatory, healing response. METHODS Treatment of hCEC with ethanol and pro-inflammatory cytokines were compared in terms of viability loss, cytotoxicity, and pro-inflammatory cytokine release, in order to generate the in vitro injury. This resulted in an optimal injury of 20%(v/v) ethanol for 30 s with 1 ng/mL interleukin-1 (IL-1) beta. Co-culture experiments were performed with CSSC alone and with CSSC-AM constructs. The effect of injury and co-culture on viability, cytotoxicity, IL-6 and IL-8 production, and IL1B, TNF, IL6, and CXCL8 mRNA expression were assessed. RESULTS Co-culture with CSSC inhibited loss of hCEC viability caused by injury. Enzyme linked immunosorbent assay and polymerase chain reaction showed a significant reduction in the production of IL-6 and IL-8 pro-inflammatory cytokines, and reduction in pro-inflammatory cytokine mRNA expression during co-culture with CSSC alone and with the AM construct. These results confirmed the therapeutic potential of the CSSC and the possible use of AM as a cell carrier for application to the ocular surface.CONCLUSION CSSC were shown to have a potentially therapeutic anti-inflammatory effect when treating injured hCEC, demonstrating an important role in corneal regeneration and wound healing, leading to an improved knowledge of their potential use for research and therapeutic purposes. 展开更多

The evolution of the natural and pristine Mediterranean coastline Port aux Princes-Sidi Daoued(Gulf of Tunis,NE-Tunisia) is studied during the period of 1887–2010 on the basis of an ancient minute of bathymetry(1887) and aerial photographs treated by numerical photogrammetric methods. Morphological changes of the coastline shows a general retreat despite the absence of the various anthropogenic actions. Adding to the drift currents and the currents of high energy that are generated by the N-W dominant waves along steeply sub-marine funds, the erosion is mainly due to the sea level rise which increased since the beginning of the 2000 s. The Port aux PrincesSidi Daoued coastline works as a single littoral cell limited by Jbel Korbous to the SW and the fishing harbor of Sidi Daoued to the N-E. 展开更多

The most common organ where follicular dendritic cell sarcoma (FDCS) occurs is in cervical lymph nodes, while few cases are found in extranodal organs such as liver, spleen, and soft tissue. This is a case report that FDCS occurs in the hepatogastric ligament. To our knowledge, there is no such case that has been reported previously. A 47-year-old male patient was found to have an intraabdominal mass during an annual physical examination. Computed tomography showed a 4.2 cm × 4.1 cm mass located at the lesser curvature of the stomach, above the pancreas. During operation, a tumor mass was found in the hepatogastric ligament and a radical resection was performed. The tumor was diagnosed as FDCS by pathology and immunohistochemical testing. The patient had a favorable recovery, and no obvious abnormality was found 3 months postoperation. 展开更多

Clinical value of serum pepsinogen levels for the diagnosis of esophageal squamous cell carcinoma 预览

Objective Pepsinogens have been previously studied as markers of gastric atrophy. The objective of this study was to investigate the clinical significance of the serum levels of pepsinogen (PG) I and II, as well as the pepsinogen I/II ratio (PGR) in the diagnosis of esophageal squamous cell carcinoma. Methods A retrospective data analysis of patients who underwent gastroscopy and PG examination in Renmin Hospital was performed. The subjects were grouped into cancer and healthy control groups, and the differences in the serum levels of PGI and PGII, as well as the PGRs were compared. The receiver operating curve and the area under the curve (AUC) were also compared between the groups. Results A total of 351 Chinese patients were enrolled in the study, 209 with esophageal squamous cell carcinoma and 142 healthy controls. Overall, the levels of PGI (P < 0.0001) and PGII (P = 0.0007), as well as the PGR (P = 0.007) of the cancer group were lower than those of the control group. Male subjects in the cancer group had lower PGI (P < 0.0001), PGII (P < 0.0001), and (P = 0.0138). The subjects < 65 years old in the cancer group showed lower PGI (P < 0.0001), PGII (P = 0.001), and PGR (P = 0.0087).Overall, these results show that the levels of PGI (AUC 0.64) and PGII (AUC 0.60) have a predictive ability for discriminating esophageal carcinoma. Moreover, in males < 65 years old, PGI (AUC 0.73) and PGII (AUC 0.69) also showed to have a predictive ability for discriminating esophageal carcinoma. Conclusion Serum PG levels in patients with esophageal squamous cell carcinoma, especially in males aged < 65 years old, are lower than those in healthy people. PGI and PGII are useful for screening esophageal squamous cell carcinoma. 展开更多

We summarized relevant clinical studies to elaborate the epidemiology, pathological and clinical characteristics and the treatment status of small cell lung cancer. This paper first described the epidemiology and the pathological and clinical characteristics of SCLC and the systematic treatment of extensive-stage SCLC and then introduced the current targeted therapy and immunotherapy for SCLC to provide clinicians and patients with a more systematic, comprehensive, and beneficial treatment regimen. We expect that these studies can provide clinicians with a clear direction in molecularly targeted therapy or immunotherapy, so that a treatment approach with better antitumor effects and longer-lasting clinical benefits can be provided to the patients. 展开更多

A commercial solid oxide fuel cell with a Ni/YSZ anode was characterized under a pure methane atmosphere. The amount of deposited carbon increased with an increase in temperature but decreased when the temperature exceeded 700℃. The reactivity of carbon decreased with increasing deposition temperature. Filamentous carbon was deposited from 400 to 600℃, whereas flake carbon was deposited at 700 and 800℃. With increasing temperature, the intensity ratio of the D band over the sum of the G and D bands was constant at the beginning and then decreased with the transformation of the carbon morphology. The crystallite size increased from 2.9 to 13 nm with increasing temperature. The results also indicated that the structure of the deposited carbon was better ordered with increasing deposition temperature. In comparison with pure Ni powders, the interaction between the YSZ substrate and Ni particles could not only modify the carbon deposition kinetics but also reduce the temperature effect on the structure and reactivity variation of carbon. 展开更多
