In the present study, we investigated anti-inflammatory effect of Cardamine komarovii flower(CKF) on lipopolysaccharide(LPS)-induced acute lung injury(ALI). We determined the effect of CKF methanolic extracts on LPS-induced pro-inflammatory mediators NO and prostaglandin E2(PGE2), production of pro-inflammatory cytokines(IL-1β, TNF-α, and IL-6), and related protein expression levels of MyD88/TRIF signaling pathways in peritoneal macrophages(PMs). Nuclear translocation of NF-κB-p65 was analyzed by immunofluorescence. For the in vivo experiments, an ALI model was established to detect the number of inflammatory cells and inflammatory factors(IL-1β, TNF-α, and IL-6) in bronchoalveolar lavage fluid(BALF) of mice. The pathological damage in lung tissues was evaluated through H&E staining. Our results showed that CKF can decrease the production of inflammatory mediators, such as NO and PGE2, by inhibiting their synthesis-related enzymes i NOS and COX-2 in LPS-induced PMs. In addition, CKF can downregulate the mRNA levels of IL-1β, TNF-α, and IL-6 to inhibit the production of inflammatory factors. Mechanism studies indicated that CKF possesses a fine anti-inflammatory effect by regulating My D88/TRIF dependent signaling pathways. Immunocytochemistry staining showed that the CKF extract attenuates the LPS-induced translocation of NF-kB p65 subunit in the nucleus from the cytoplasm. In vivo experiments revealed that the number of inflammatory cells and IL-1β in BALF of mice decrease after CKF treatment. Histopathological observation of lung tissues showed that CKF can remarkably improve alveolar clearance and infiltration of interstitial and alveolar cells after LPS stimulation. In conclusion, our results suggest that CKF inhibits LPS-induced inflammatory response by inhibiting the MyD88/TRIF signaling pathways, thereby protecting mice from LPS-induced ALI. 展开更多

麻疯树BRI1基因的鉴定及其在不同发育时期花蕾中的表达分析 预览

[目的]研究油菜素内酯受体BRI1的同源基因(JcBRI1)在麻疯树花发育过程中的作用。[方法]利用RT-PCR技术克隆JcBRI1基因的CDS,以pET-30a(+)质粒为框架构建原核表达载体转化至大肠杆菌进行诱导表达,随后利用LC-MS/MS对表达产物进行质谱鉴定,并以氨基酸序列为基础分析JcBRI1蛋白质结构。利用荧光定量PCR分析麻疯树JcBRI1基因在雌雄花发育的关键时期的表达水平,初步确定其在麻疯树花发育过程中的表达模式。[结果]克隆获得了JcBRI1基因的CDS,长度为3 591 bp。SDS-PAGE检测结果表明:JcBRI1基因在大肠杆菌BL21(DE3)和Rosetta(DE3)中均能表达,但在低温环境的表达量更高。原核表达产物的LC-MS/MS鉴定结果表明:JcBRI1氨基酸序列与预期的一致,JcBRI1基因的表达产物为麻疯树BRI1蛋白。对JcBRI1在麻疯树花器官不同发育时期的表达水平进行检测分析,结果表明:JcBRI1基因的表达量在雌花发育的第一个时期即大孢子母细胞时期就达到了最高值,在随后几个时期持续下调;然而,其在雄花各个时期的表达量并无明显差异,表明其很有可能参与了麻疯树雌花大孢子母细胞发育过程。[结论]麻疯树JcBRI1基因为LRR-RLKs家族成员BRI1的同源基因,很有可能参与麻疯树雌花大孢子母细胞的发育过程,至于JcBRI1在麻疯树大孢子母细胞发育过程中的具体作用机制还需进一步研究。 展开更多

Transcription factors (TFs) are key cellular components that control gene expression. They recognize specific DNA sequences, the TF binding sites (TFBSs), and thus are targeted to specific regions of the genome where they can recruit transcriptional co-factors and/or chromatin regulators to fine-tune spatiotemporal gene regulation. Therefore, the identification of TFBSs in genomic sequences and their subsequent quantitative modeling is of crucial importance for understanding and predicting gene expression. Here, we review how TFBSs can be determined experimentally, how the TFBS models can be constructed in silico, and how they can be optimized by taking into account features such as position interdependence within TFBSs, DNA shape, and/or by introducing state-of-the-art computational algorithms such as deep learning methods. In addition, we discuss the integration of context variables into the TFBS modeling, including nucleosome positioning, chromatin states, methylation patterns, 3D genome architectures, and TF cooperative binding, in order to better predict TF binding under cellular contexts. Finally, we explore the possibilities of combining the optimized TFBS model with technological advances, such as targeted TFBS perturbation by CRISPR, to better understand gene regulation, evolution, and plant diversity. 展开更多

Background:Eucommia ulmoides Oliv. is a medicinal plant native to China, with its bark (Eucommiae Cortex) traditionally being used for medicinal purposes. Previous research has shown that Eucommia male flowers can exert anti-inflammatory, analgesic, antibacterial, and other pharmacological effects, including immune regulation. This study explored the anti-inflammatory effects of the 70% ethanol extract of male flowers (EF) of E. ulmoides in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells and LPS-administered mice.Methods:Cytotoxicity of EF for RAW 264.7 cells was investigated using Cell Counting Kit-8. The production of proinflammatory mediators, nitric oxide (NO), tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6 was determined using enzyme-linked immunosorbent assays. IL-17, IL-23, and IL-10 mRNA levels were determined using quantitative real-time polymerase chain reaction. Activation of the nuclear factor (NF)-κB pathway in RAW 264.7 cells was investigated via Western blotting. In vivo antiinflammatory effects of EF were studied in an LPS-induced acute inflammation mouse model by analyzing lung tissue histopathology, serum TNF-α and IL-6 levels, and myeloperoxidase (MPO) activity in lung tissue.Results:EF showed no significant cytotoxicity at concentrations from 10 to 60 μg/mL (cell viability > 80%) in the CCK-8 cell viability assay. EF inhibited the RAW 264.7 cell proliferation (EF 60 μg/mL, 120 μg/mL, and 250 μg/mL vs. negative control: 87.31±2.39% vs. 100.00±2.50%, P=0.001;79.01±2.56 vs. 100.00±2.50%, P<0.001;and 64.83±2.50 vs. 100.00±2.50%, P<0.001), suppressed NO (EF 20 μg/mL and 30 μg/mL vs. LPS only, 288.81±38.01 vs. 447.68±19.07 μmol/L, P=0.004;and 158.80±45.14 vs. 447.68±19.07 μmol/L, P<0.001), TNF-α (LPS+EF vs. LPS only, 210.20±13.85 vs. 577.70±5.35 pg/mL, P<0.001), IL-1β (LPS+EF vs. LPS only, 193.30±10.80 vs. 411.03±42.28 pg/mL, P<0.001), and IL-6 (LPS+EF vs. LPS only, 149.67±11.60 vs. 524.80±6.24 pg/mL, P<0.001) secretion, and downregulated the mRNA expr 展开更多

以地被菊俏粉阁为试材,分期短日照处理40d(当日17:00至次日8:00),设置5个处理(以不遮光为对照),测定植株生长发育的相关指标并记录开花进程,探求营养生长时间对菊花生长开花的影响。结果表明,光周期处理前,T4处理(营养生长40d)俏粉阁根... 以地被菊俏粉阁为试材,分期短日照处理40d(当日17:00至次日8:00),设置5个处理(以不遮光为对照),测定植株生长发育的相关指标并记录开花进程,探求营养生长时间对菊花生长开花的影响。结果表明,光周期处理前,T4处理(营养生长40d)俏粉阁根系长度、鲜质量及干质量显著高于其他处理;俏粉阁株高、冠幅随着营养生长时间的增加逐渐增加,处理22~28d,T2,T3,T4处理株高、冠幅生长量显著高于其他处理,处理36~42d,对照处理株高、冠幅生长量最大;俏粉阁花期随着营养生长时间的增加出现延迟,T4处理植株到达花期各阶段的间隔时间最短,开花最晚,但各处理植株开花总时长差异不显著;T4处理植株开花量、舌状花长度、花径、重瓣性及花粉活力显著高于其他处理。随着营养生长时间的增加,俏粉阁株高、冠幅、根系长度等逐渐增加,盛花期花朵品质提高,但开花出现延迟。当营养生长20d时进行短日诱导,生长速度最快,开花最早,花朵品质较好。 展开更多

白及花与块茎石油醚部位成分分析及其生物活性的比较研究

目的分析白及花和块茎石油醚部位成分及生物活性。方法以石油醚(60~90℃)为提取溶剂,采用索氏提取法提取白及花和块茎脂溶性成分,采用GC-MS分析其化学成分,通过滤纸片扩散法、CCK8法和分光光度法分别对其进行抑菌、抗肿瘤和α-淀粉酶抑... 目的分析白及花和块茎石油醚部位成分及生物活性。方法以石油醚(60~90℃)为提取溶剂,采用索氏提取法提取白及花和块茎脂溶性成分,采用GC-MS分析其化学成分,通过滤纸片扩散法、CCK8法和分光光度法分别对其进行抑菌、抗肿瘤和α-淀粉酶抑制作用研究,通过检测DPPH自由基、ABTS自由基和羟自由基的清除率考察其体外抗氧化活性。结果白及花和块茎中共鉴定出20种石油醚部位成分,其中花11种,块茎19种,共有成分10种,α-乙酰基-γ-丁内酯(15.20%,21.84%)和苄醇(12.07%,24.10%)为主要成分。白及花中石油醚部位成分的抑菌活性高于块茎,对枯草芽孢杆菌效果最好,最小抑菌浓度和最小杀菌浓度分别为0.41,0.51 g·L-1和0.53,0.66 g·L-1。当质量浓度为5.0 mg·mL-1时,白及块茎石油醚部位成分对A549细胞的抑制率为29.451%,高于花中石油醚部位成分27.621%的抑制率,前者对α-淀粉酶的IC50为1.819 mg·mL-1,后者的IC50为2.028mg·mL-1。两者对DPPH自由基、ABTS自由基和羟自由基的清除率均随质量浓度的增大而增大,当浓度达到1.0mg·mL-1时,前者对ABTS自由基和羟自由基的清除率分别为99.67%和92.22%,高于后者90.07%和38.11%的清除率,后者对DPPH自由基的清除率为31.23%,高于前者18.94%的清除率,两者均低于Vc的清除率。结论白及花和块茎中石油醚部位成分主要为酯类和醇类,2种石油醚部位成分均具有一定的生物活性,具有潜在的药用价值,并可为白及花的资源开发提供参考。 展开更多

Objective: Flower herbs are an important category of traditional Chinese medicinal materials, some of which are used as healthcare tea in folk. However, the increasing adulteration of medicinal herbs is threatening consumer safety. The adulteration of flower herbs and their healthcare tea products in the market were investigated.Methods: A total of 33 flower herb samples from several retail pharmacies in China were randomly collected and 27 flower healthcare tea samples were purchased online. They were identified using ITS2-based Traditional Chinese Medicine Database(TCMD). Additionally, Lonicerae Japonicae Flos and the adulterants were compared in the ITS2 secondary structures.Results: There were one adulterant(Inulae Flos) in flower herb materials and eight adulterants in healthcare tea samples. Inula linariifolia was an adulterate species of Inulae Flos, Robinia pseudoacacia was of Sophorae Flos, and Lonicera macranthoides was of Lonicerae Japonicae Flos. Sophorae Flos and Lonicerae Japonicae Flos were two healthcare tea products with high adulteration rates.Conclusion: The TCMD is powerful tool to identify flower herbs and the adulterants that frequently occurred in the flower herb market, especially online shops. 展开更多

In order to understand the mechanism of spathe color variation in Anthurium andraeanum at the protein level, the leaves, inflorescences and spathes of the wild type and two mutants of A. andraeanum ‘Madural’ were used as research objects in which the differential expression of proteins related to flower color mutants was analyzed by one-dimensional electrophoresis and mass spectrometry (1-DE/MS). The 1-DE patterns showed that the protein components expressed highly in spathes were mainly concentrated in the molecular weight range of 20-42 kD, and differential bands were detected between the wild type and the mutant, while no significant differences were detected in the leaf and inflorescence proteins. According to the results of mass spectrometry analysis of the differential bands, 21 known functional proteins involved in life processes such as glucose metabolism, resistance, cytoskeleton, gene regulation and signal transduction were identified. It showed that in addition to the influences from anthocyanidins, the spathe color variation of A. andraeanum ‘Madural’ is also regulated by a variety of metabolic pathway-related proteins. 展开更多
