浅部真菌病荧光染色显微成像及计算机辅助诊断系统的初步临床应用

目的评估荧光染色显微成像及计算机辅助诊断系统(AFMICADS)在临床上辅助诊断浅部真菌感染的准确性。方法2018年7-9月选择华中科技大学同济医学院附属协和医院各临床科室门诊及住院疑似浅部真菌感染患者106例,送检标本126份,其中皮肤鳞... 目的评估荧光染色显微成像及计算机辅助诊断系统(AFMICADS)在临床上辅助诊断浅部真菌感染的准确性。方法2018年7-9月选择华中科技大学同济医学院附属协和医院各临床科室门诊及住院疑似浅部真菌感染患者106例,送检标本126份,其中皮肤鳞屑83份,甲屑43份,分别进行常规镜检、改良沙氏培养基培养及荧光染色镜检(人工荧光镜检和AFMICADS荧光镜检)。以常规镜检和/或真菌培养阳性(仅满足任意1项即可为阳性)为标准,计算3种镜检方法的准确率、敏感性和特异性。统计分析采用SPSS10.0软件,McNemar检验和Kappa检验分析3种镜检方法与常规镜检和/或真菌培养结果的差异及一致性,诊断符合率检验分析3种镜检方法两两之间一致性的差异。结果126份标本中,人工荧光镜检阳性124份(98.4%),AFMICADS荧光镜检阳性123份(97.6%),阳性率均高于常规镜检和/或真菌培养法[阳性98份(77.8%),均P<0.001]。AFMICADS荧光镜检的敏感性为100%,特异性为10.7%,一致性为80.2%;人工荧光镜检的敏感性100%,特异性7.1%,一致性79.4%,两种方法的一致性差异无统计学意义(P>0.05)。结论AFMICADS荧光镜检浅部真菌感染标本的一致性与人工荧光镜检相当。 展开更多

涂片真菌荧光染色法对真菌性角膜炎诊断价值的研究

目的分析真菌荧光染色技术在真菌性角膜炎诊断中的敏感性及特异性,探讨其临床应用价值。方法前瞻性病例对照研究。连续收集2017年8月至2018年4月就诊于首都医科大学附属北京同仁医院确诊为感染性角膜炎的患者105例(105只眼),其中男性69... 目的分析真菌荧光染色技术在真菌性角膜炎诊断中的敏感性及特异性,探讨其临床应用价值。方法前瞻性病例对照研究。连续收集2017年8月至2018年4月就诊于首都医科大学附属北京同仁医院确诊为感染性角膜炎的患者105例(105只眼),其中男性69例,女性36例,年龄(45.1±20.1)岁。根据裂隙灯显微镜、角膜激光共聚焦显微镜(IVCM)检查,以及病灶处角膜刮片吉姆萨染色、荧光染色和病原学培养的结果,将感染性角膜炎患者分为真菌性角膜炎(FK)组以及非真菌性角膜炎(NFK)组,分析上述检查方法对真菌性角膜炎诊断的敏感性和特异性,并通过受试者工作特征(ROC)曲线计算曲线下面积(AUC),明确真菌荧光染色方法对真菌性角膜炎的诊断价值。结果105例感染性角膜炎患者中,真菌性角膜炎66例,非真菌性角膜炎39例(其中细菌性角膜炎29例,阿米巴角膜炎10例)。FK组真菌培养阳性者46例(69.7%),其中以镰刀菌感染为主(20/46,43.5%),其次为链隔孢属(10/46,21.7%)以及曲霉属(9/46,19.6%)等。真菌涂片荧光染色后,组织背景呈均质的黑色或弱蓝色荧光,真菌细胞壁成分呈亮蓝紫色或蓝色的明亮荧光,形态、结构、菌丝密度等均容易识别。不同检查方法对角膜真菌感染诊断的敏感性分别为涂片荧光染色(97.0%)、IVCM(87.9%)、吉姆萨染色(86.7%)、真菌培养(69.7%);特异性方面,真菌培养最高(100.0%),其次为IVCM和吉姆萨染色(94.9%)和荧光染色(87.2%)。AUC值由低至高依次为真菌培养(0.848),涂片吉姆萨染色(0.906),IVCM(0.914)和涂片荧光染色(0.921)。结论真菌荧光染色是一种快速、敏感的真菌镜检筛查手段,对真菌性角膜炎辅助诊断的敏感性及特异性较高,尤其适用于菌量少及抗真菌治疗后患者的诊断。 展开更多

It has been challenging to develop deep blue organic molecular fluorescent emitters with CIE y(y≤0.08)based on triplet-triplet annihilation(TTA). Here, we report facilely available dianthracenylphenylenebased emitters, which have a 3,5-di(4-t-butylphenyl)phenyl moiety at the one end and 4-cyanophenyl or 3-pyridyl at the other end, respectively. Both fluorophores show a high glass transition temperature of over 220℃ with a thermal decomposition temperature of over 430℃ at an initial weight loss of1%. The preliminary characterizations of the organic light-emitting diodes(OLEDs) that utilized these nondoped emitters provided high EQEs of 4.6%à5.9% with CIE coordinates(0.15, 0.07–0.08). The analysis of the EL transient decay revealed that TTA contributed to the observed performance. The results show that the new emitters are attractive as a potential TTA-based host to afford stable deep blue fluorescent OLEDs. 展开更多

生活污水处理过程荧光光谱及紫外光谱特征分析

应用平行因子分析,并结合污水理化指标的相关性分析,对污水处理厂进出水以及各个处理单元中的污水的三维荧光光谱及紫外光谱特征进行了研究。对生活污水来源进行了解析并与同期工业污水进行对比。结果表明,污水处理厂中进出水以及各个... 应用平行因子分析,并结合污水理化指标的相关性分析,对污水处理厂进出水以及各个处理单元中的污水的三维荧光光谱及紫外光谱特征进行了研究。对生活污水来源进行了解析并与同期工业污水进行对比。结果表明,污水处理厂中进出水以及各个处理单元的污水均含有3种主要的荧光组分,分别是UVA类腐殖质、UVC类腐殖质和类色氨酸。根据得分值可以得出处理后类色氨酸在污水的荧光组分中降低了94%,而类腐殖质趋于稳定。生活污水水质主要受类色氨酸影响,水质比工业废水较稳定。相关性分析表明,类色氨酸与传统指标具有较强的相关性,类腐殖质与UV254之间相关性较好。 展开更多

Two crystals,[Sm(Hcpna)(cpna)·3 H2O]n(1) and [Eu(Hcpna)(cpna)·2 H2O]n(2),have been synthesized by solvothermal method based on Sm3+ and Eu3+ with multiple acid ligands5-(4-carboxyphenoxy)nicotinic acid(H2cpna), and the new complexes have been characterized by single-crystal X-ray diffraction, elemental analysis, IR spectroscopy, thermogravimetric analysis and fluorescence analysis. Complex 1 exhibits a 1 D chain structure. It crystallizes in the triclinic system, space group P1 with a = 5.9738(11), b = 12.528(2), c = 18.021(4)?,α= 93.156(3)o,β=98.067(3)o,γ= 92.692(3)o, V = 1331.3(4) A3, Z = 2, S = 1.069, F(000)= 702, R = 0.0277 and wR =0.0688(I > 2σ(I)). Complex 2 shows a two-dimensional network structure, which crystallizes in monoclinic system, space group C2/c with a = 13.6733(19), b = 18.422(3), c = 10.4307(15) A,β=93.881(2)o, V= 2621.4(7) A3, Z = 4, S = 1.110, F(000)= 1372, R = 0.0302 and wR = 0.0811(I >2σ(I)). The solid-state fluorescent test results show tcomplex 1 has weaker fluorescence intensity than the ligand. However, complex 2 has excellent fluorescence properties, and can emit bright red light under the irradiation of ultraviolet lamp. 展开更多

吲哚菁绿荧光显像技术在肝母细胞瘤精准切除手术中的应用 预览

目的探讨吲哚菁绿(indocyanine green,ICG)介导的近红外光在肝母细胞瘤中的显像特点,以及该技术对术中微小病灶识别、肿瘤切缘界定和术中导航的价值。方法2018年3月至2018年6月由复旦大学附属儿科医院收治的拟切除肝母细胞瘤的患儿8例,... 目的探讨吲哚菁绿(indocyanine green,ICG)介导的近红外光在肝母细胞瘤中的显像特点,以及该技术对术中微小病灶识别、肿瘤切缘界定和术中导航的价值。方法2018年3月至2018年6月由复旦大学附属儿科医院收治的拟切除肝母细胞瘤的患儿8例,术前至少24 h予以静脉注射ICG,术中切肝前进行肝脏微小病灶检测、肿瘤切缘界定和手术导航,切肝后进行切缘病灶检测和肿瘤荧光分型,最终与病理结果对比。结果8例肝母细胞瘤中,男童和女童各4例,年龄5~132个月,平均33个月。术后复发再次手术1例,术前活检化疗后再手术1例,术前经验性化疗后再手术4例,2例一期行肝肿瘤切除术。术中8例患儿肿瘤都呈现明亮荧光显影,与正常肝脏组织边界清晰;1例复发为微小病灶(1.0 cm),但由于病灶位置较深(距离肝脏表面 >1.5 cm),肉眼观察、扪诊和ICG荧光显像未能发现,规则切除肝叶后,ICG识别出复发病灶。根据荧光显影分型,全荧光4例,其中2例肝母细胞瘤一期行肿瘤切除术;1例术前有活检和化疗史的病例;1例为复发病例,其有肝炎和肝轻度硬化病史,同时伴有肝脏弥漫性小结节样影。部分荧光4例,均为术前有经验性化疗的病例。结合肝母细胞瘤的病理分类,胎儿型7例,其中荧光显像为全荧光型4例,部分荧光型3例;混合性胎儿上皮和胚胎上皮型1例,其荧光显像为部分荧光型。所有病灶经ICG分子荧光边界定界,切除肿瘤后,经荧光再测和病理检查,均获得了肿瘤R0切除。结论ICG荧光显影技术在肝母细胞瘤中有较好的显影效果,显影类型与术前有无化疗有一定关系。ICG荧光显像在确定肿瘤边界和保证肿瘤R0切除具有一定优势和重要价值。但ICG荧光显像对深部微小肿瘤灶的识别和在肝硬化病例中的运用存在一定的局限性。 展开更多

High sensitive,accurate detection for tumor-associated overexpressed enzyme activity is highly significant for further understanding enzyme function,discovering potential drugs,and early diagnosis and prevention of diseases.In this work,we developed a facile,direct and single-step detection platform for primary ovarian cancers related glycosidase activity based on the inner filter effect(IFE)between glycosidase catalytic product and black phosphorus quantum dots(BPQDs).Highly fluorescent BPQDs were successfully synthesized from bulk black phosphorus by a simple liquid exfoliation method.Under the catalysis ofβ-galactosidase,p-nitrophenyl-β-D-galactopyranoside(PNPG)was transformed into pnitrophenol(PNP)andβ-D-galactopyranoside.Meanwhile,the absorption of catalytic product PNP greatly overlapped with the excitation and emission spectra of fluorescent BPQDs,leading to the fluorescence quenching of BPQDs with a high quenching efficiency.The proposed sensing strategy provided a low detection limit of 0.76 U/L,which was 1-2 orders of magnitude lower than most unmodified sensing platforms.D-Galactal was selected as the inhibitor forβ-galactosidase to further assess the feasibility of screening potential inhibitors.The fluorescence recovery of BPQDs suggests that the unmodified sensing platform is feasible to discover potential drugs ofβ-galactosidase.Our work paves a general way in the detection of glycosidase activity with fluorescent BPQDs,which can be promising for glycosidase-related disease diagnosis and pathophysiology elucidation. 展开更多

A new kind of bifunctional magnetic-fluorescent nanoparticles(NPs)with abundant carboxyls on the surface has been prepared by covalently combining glutathiose(GSH)-modified CdTe quantum dots(QDs)with Fe3O4@SiO2NPs.The silica-coated Fe3O4NPs were functionalized with amino groups by(3-aminopropyl)triethoxysilane to provide Fe3O4@SiO2-NH2NPs,which was then chemically conjugated with GSH-modified CdTe QDs to form bifunctional magnetic-fluorescent NPs,Fe3O4@SiO2-NH-CO-CdTe-QDs NPs.The properties and morphologies of Fe3O4@SiO2-NH-CO-CdTe-QDs NPs were investigated by FTIR,X-ray diffraction,transmission electron microscopy(TEM),absorption and fluorescence spectroscopy and vibrating sample magnetometry.TEM images display that Fe3O4@SiO2-NH-CO-CdTe-QDsNPs possess spherical core-shell structure with a uniform size about 50 nm.The bifunctional NPs were found to exhibit good magnetic and strong fluorescent properties favorable for their application in the detection of latent fingerprints.Furthermore,the carboxyls on the surface of NPs have good absorptions with water in air and the residues of fingerprints so as to not only avoid dust flying to protect the health of operators,but also improve the efficiency of detection. 展开更多

Amine groups can interact with the cell membrane which results in cell death. These limitations of polymeric amines stimulated an investigation of new structures for gene delivery. Imidazole-containing polymers have attracted attention as lesser basic substances, while they are able to interact with polymeric acids. Further development of imidazole-based gene delivery agents requires knowledge about some fundamental aspects of interaction between nucleic acids, and polymeric imidazoles. In this work, we studied the complexation of poly(1-vinylimidazole) and oligomeric DNA. We found that the number of active sites capable of binding with negatively charged phosphate groups is comparable with the number of protonated imidazole units in the case of high molecular weight polymer. The increase in polymer charge by 1-bromopropane quaternizating 1%?5% imidazole units or by decreasing the pH to 6.5?7 considerably increased the ability of poly(1-vinylimidazole) to interact with oligonucleotides. The pH sensitivity of this interaction is interesting for cancer gene therapy because the tumours have a lowered intercellular pH (stable oligonucleotide complex) and a higher extracellular pH which can lead to complex dissociation. Minimal critical length for complexation of quaternized poly(1-vinylimidazole) and DNA is below eight units which corresponds to polymers with amine groups. Fluorescence-tagged poly(1-vinylimidazole) samples were obtained and their potential for monitoring the polymer and polymer-oligonucleotide complex internalization into living cells was demonstrated. 展开更多

Enzyme-based anticancer therapy is more attractive for the less side effect than conventional chemotherapy.However,the poor stability and low membrane permeability of enzymes during the intracellular delivery are constraints for its practical applications.In this work,we synthesized novel near-infrared (NIR)-responsive core-shell-structured Prussian blue@fibrous SiO2 (PBFS) nanoparticles as the carrier of superoxide dismutase (SOD) and a glutathione (GSH)-activated Fenton reagent (DiFe).The PBFS nanoparticles are further modified with aGSH-responsive cationic polymer (poly(2-(acryloyloxy)-N,N-dimethyl-N-(4-(((2-((2-(((4-methyl-2-oxo-2H-chromen-7-yl)carbamoyl)oxy)ethyl)disulfaneyl)ethoxy)carbonyl)amino)benzyl)ethan-1-aminium,PSS) containing disulfide bonds and fluorophores.After SOD and DiFe are loaded on the PBFS-PSS nanoparticles,dual chemodynamic/photothermal therapeutic nanoparticulate systems (PBFS-PSS/DiFe/SOD) are obtained.In vitro experiments show that PBFS-PSS/DiFe/SOD nanoparticles have good biocompatibility and can be tracked under fluorescence microscope during the intracellular delivery process in MCF-7 tumor cells due to the GSH-activated release of fluorophores.They also exhibit high efficiency in NIR photothermal conversion and GSH-activated Fenton reaction in tumor cells,thus achieving high-efficient killing effect of tumor cells based on the combination of photothermal and chemodynamic therapeutic performance (PTT and CDT).This work offers a novel pathway to construct a visual multifunctional nanomedicine platform for future cancer therapy. 展开更多

A novel functional fiber(PAN-CDs) loaded with carbon dots(CDs) with excellent photoreduction and adsorption properties for Cr(VI) was prepared via an amidization reaction between the CDs’ carboxyl groups and amine groups on polyacrylonitrile(PAN)-based ion exchange fibers, which could completely preserve the fluorescence properties of the CDs. The photoluminescence(PL), photocatalysis and adsorption properties of PAN-CDs were characterized and analyzed. The PAN-CDs possess high adsorption capacity(297.6 mg/g) and excellent kinetic behavior(attaining adsorption equilibrium in 30 min)for Cr(VI) adsorption. Furthermore, the residual Cr(VI)(approximately 3 mg/L) after adsorption could be removed completely by subsequent photoreduction by the PAN-CDs.The Cr-saturated PAN-CDs could be easily separated by filtering and regenerated, with no observable decay of removal efficiency after five regeneration cycles. In addition, due to the PL quenching action of Cr(VI), the PAN-CDs can also be used as sensor for quantitative detection of trace Cr(VI) in aqueous solution. 展开更多

In this work, we designed a facile and promising strategy for fluorescence determination of S1 endonuclease activity and inhibition based on fluorescence resonance energy transfer(FRET) between positively Ag nanorods(AgNRs) and negatively-charged ROX-labeled sing-stranded DNA(ROX-ssDNA). In the absence of Sl endonuclease, the fluorescence signal of the ROX-ssDNA was efficiently quenched when the ROX-ssDNA was adsorbed on the surface of AgNRs via strong electrostatic interaction. However, upon addition of different concentration of Sl endonuclease, the fluorescence was gradually restored owing to the reduction of FRET efficiency caused by S1 endonuclease specific cleavage ROX-ssDNA into short fragments, which reduced the electrostatic interaction between AgNRs and short oligonucleotide fragment. This assay strategy exhibited a high sensitivity and excellent specificity for S1 endonuclease with a detection limit of 0.004 U/mL and a dynamic concentration range from 0.01 U/mL to 5.0 U/mL. In addition, the capabilities for screening of S1 endonuclease inhibitors and S1 endonuclease detection from complex biological matrixes were also verified. 展开更多

目的探讨空芯针穿刺活检术(core needle biopsy, CNB)与微创旋切活检术(vacuum-assisted biopsy, VAB)对有活检指征患者的临床诊断价值,比较两种方法在乳腺疾病诊断、免疫组织化学判断及荧光原位杂交(fluorescence in situ hybridizatio... 目的探讨空芯针穿刺活检术(core needle biopsy, CNB)与微创旋切活检术(vacuum-assisted biopsy, VAB)对有活检指征患者的临床诊断价值,比较两种方法在乳腺疾病诊断、免疫组织化学判断及荧光原位杂交(fluorescence in situ hybridization, FISH)检验等方面的差异。方法选取150例行CNB的患者,同时选择150例年龄、肿块大小和乳腺影像报告和数据系统(breast imaging reporting and data system, BI-RADS)分类情况相似的行VAB的病例进行配对分析,统计两种活检方式诊断的准确率及术前和术后免疫组织化学、FISH检验的符合率,同时比较两种活检方式术中和术后并发症情况。结果 VAB组与CNB组术前、术后病理诊断一致率分别为98.0%和96.7%(P=0.395)。VAB组65例导管内原位癌(ductal carcinoma in situ, DCIS)中5例为浸润性导管癌(invasive ductal carcinoma, IDC),CNB组64例DCIS中22例为IDC(P=0.034)。VAB组未发现非典型性导管增生(atypical ductal hyperplasia, ADH)病例的鉴定误差,CNB组2例ADH中1例为IDC(P=0.021)。VAB比CNB需要更长的操作时间(P<0.01),疼痛评分报告VAB的疼痛程度与CNB相似(P=0.962)。结论空芯针穿刺活检术是一种简便易行、安全且并发症少的可靠诊断技术,值得临床推广应用。而微创旋切活检术针体更粗、取材更充分,可能在免疫组织化学的判断上更具优势,病理符合率也更加明显,特别是对于IDC低估率明显降低。 展开更多

We report a facile, effective and rapid screening method for the determination of total fluoroquinolones(FQs) in milk using lanthanide coordination polymer nanomaterials as the sensing platform. The nonluminescent coordination polymer nanosheets(CPNSs) used in this work are composed of adenosine monophosphate(AMP) and terbium(Tb3+) ions(AMP/Tb CPNSs). The as-prepared CPNSs were characterized by transmission electron microscopy, Fourier transform infrared spectroscopy and fluorescence techniques. Tb3+ acts as the center ion of the coordination polymer, with AMP assembled through phosphate and amine groups. However, the reaction between FQs and AMP/Tb CPNSs noticeably turns on the fluorescence due to the strong coordination interaction between FQs and Tb3+. Under optimized conditions, the fluorescence intensity of AMP/Tb at 545 nm shows a linear relationship with FQ concentrations between 0.04 μmol/L and 5.0 μmol/L. The relative standard deviation(RSD) of the intraday precision is below 1%, and detection limits are as low as 0.01 μmol/L. The proposed method was successfully applied to the determination of FQs in milk samples. 展开更多
