AIM: To determine the effect of pirfenidone on the activated human Müller cells by platelet-derived growth factor-BB(PDGF-BB). METHODS: The primary human Müller cells were separated from retinal tissues and established the pathogenic model by stimulated with PDGF-BB. The Müller cells behaviour of normal group and the model group was measured by MTT assay, Trypan blue assay, cell migration assay, and collagen contraction assay. The expression of transforming growth factor(TGF)-β1,-β2, and pigment epithelium-derived factor(PEDF) was estimated with realtime polymerase chain reaction(PCR), Western blot and immunofluorescence analyses. RESULTS: A pathogenic/proliferative model of Müller cells was established by stimulating normal cultured Müller cells with 10 ng/mL PDGF-BB for 48 h. After treated with 0.2 and 0.3 mg/mL pirfenidone, the proliferation, migration and collagen contraction was statistically significantly depressed in the model group compared with the normal groups. The expression levels of TGF-β1 and TGF-β2 were significantly down-regulated, while the PEDF expression was significantly up-regulated after treated with 0.2 and 0.3 mg/mL pirfenidone in the model group. CONCLUSION: Pirfenidone effectively suppress the proliferation, migration and collagen contraction of the human Müller cells stimulated with PDGF-BB through down-regulation of TGF-β1/TGF-β2 and up-regulation of PEDF. 展开更多

The role of vascular endothelial growth factor A in platelet adhesion in cerebral microvessels in the early stage of subarachnoid hemorrhage remains unclear.In this study,the endovascular puncture method was used to produce a rat model of subarachnoid hemorrhage.Then,30 minutes later,vascular endothelial growth factor A antagonist anti-vascular endothelial growth factor receptor 2 antibody,10μg,was injected into the right ventricle.Immunohistochemistry and western blot assay were used to assess expression of vascular endothelial growth factor A,occludin and claudin-5.Immunohistochemical double labeling was conducted to examine co-expression of GP Ⅰa-Ⅱ integrin and type Ⅳ collagen.TUNEL was used to detect apoptosis in the hippocampus.Neurological score was used to assess behavioral performance.After subarachnoid hemorrhage,the expression of vascular endothelial growth factor A increased in the hippocampus,while occludin and claudin-5 expression levels decreased.Co-expression of GP Ⅰa-Ⅱ integrin and type Ⅳ collagen and the number of apoptotic cells increased,whereas behavioral performance was markedly impaired.After treatment with anti-vascular endothelial growth factor receptor 2 antibody,occludin and claudin-5 expression recovered,while co-expression of GP Ⅰa-Ⅱ integrin and type Ⅳ collagen and the number of apoptotic cells decreased.Furthermore,behavioral performance improved notably.Our findings suggest that increased vascular endothelial growth factor A levels promote platelet adhesion and contribute to early brain injury after subarachnoid hemorrhage.This study was approved by the Biomedical Ethics Committee,Medical College of Xi’an Jiaotong University,China in December 2015. 展开更多

Metal halide perovskites are emerging as new generation optoelectronic materials due to their high carrier mobility, long carrier diffusion length and large light absorption coefficient, which have broad applications in solar cell, light-emitting diode, laser, photodetector and transistors. Perovskite single crystal is an ideal platform for discerning the intrinsic properties of these materials. In some cases, perovskite single crystals are better candidates to gain high performance optoelectronics. However, the growth of perovskite single crystals is time and cost consuming, which has an obvious disadvantage for device exploration. Therefore, fast growth technique is highly desirable in not only promoting the use of perovskites in commercial applications but also facilitating deep physical investigation of the materials. In this review, we summarize thoroughly the development of fast growth of the halide perovskites single crystal. Specifically, we highlight the progress of rapid growth techniques with emphasis on the optimization control. 展开更多

[目的]研究不同覆盖处理对油茶细根生长、死亡的影响,为油茶水分管理提供依据。[方法]采用微根管动态监测技术,以“长林4号”油茶林为试材,设置了黑地膜、油茶壳、花生秆+稻草、生态膜以及对照(无覆盖)5种处理,对林地040cm土壤剖面的油茶细根进行了为期1年的观测。[结果]表明:花生秆+稻草处理和黑地膜处理显著增大了油茶活细根的总根长、总根尖数以及总表面积,其中以花生秆+稻草处理最优;但对活细根平均直径的促进作用却以黑地膜最为显著。4种覆盖处理死细根的总根长、总根尖数以及总表面积均大于对照;与对照处理相比,4种覆盖处理都有助于减小死细根的直径,且差异显著。油茶活细根的总根长、总根尖数以及总表面积在3月和6—7月出现峰值;而死细根的总根长、总根尖数以及总表面积在春季变化较平缓,而后缓慢上升。油茶细根的净生长速率呈先上升后下降趋势,在5—6月出现峰值。花生秆+稻草处理和黑地膜处理显著提高了油茶细根的年净(根长)生产力,其中花生秆+稻草处理作用最为明显。[结论]夏季为油茶细根生长和死亡的高峰期,花生秆+稻草处理对促进油茶活细根根量和年净(根长)生产力的作用最显著,黑地膜处理对促进油茶活细根直径的增大效果最明显。 展开更多

Background:Chondrocytes in the growth plate(GP)undergo increases in volume during different cascades of cell differentiation during longitudinal bone growth.The volume increase is reported to be the most significant variable in understanding the mechanism of long bone growth.Methods:Forty‐five postnatal Sprague‐Dawley rat pups,7‐15 days old were divided into nine age groups(P7‐P15).Five pups were allocated to each group.The rats were sacrificed and tibia and metatarsal bones were harvested.Bone lengths were measured after 0,24,48,and 72 hours of ex vivo incubation.Histology of bones was carried out,and GP lengths and chondrocyte densities were determined.Results:There were significant differences in bone length among the age groups after 0 and 72 hours of incubation.Histological sectioning was possible in metatarsal bone from all age groups,and in tibia from 7‐to 13‐day‐old rats.No significant differences in tibia and metatarsal GP lengths were seen among different age groups at 0 and 72 hours of incubation.Significant differences in chondrocyte densities along the epiphyseal GP of the bones between 0 and 72 hours of incubation were observed in most of the age groups.Conclusion:Ex vivo growth of tibia and metatarsal bones of rats aged 7‐15 days old is possible,with percentage growth rates of 23.87±0.80%and 40.38±0.95%measured in tibia and metatarsal bone,respectively.Histological sectioning of bones was carried out without the need for decalcification in P7‐P13 tibia and P7‐P15 metatarsal bone.Increases in chondrocyte density along the GP influence overall bone elongation. 展开更多

Growth structures and growth strata of the Qianjiadian Basin in the western Yanshan fold and thrust belt, North China

The northwestward subduction of the Izanagi Plate beneath the eastern Eurasian Plate during the Late Mesozoic caused a series of compressional deformation events in the Yanshan fold and thrust belt(YFTB), but the tectonic deformation timing, tectonic properties and relationship between tectonic uplift and sediment accumulation in the intermontane basins continue to be debated. For this reason, the sedimentology, sediment provenance, and basin structure of the Qianjiadian Basin(QJB) in the northern Beijing region during the Late Jurassic and Early Cretaceous were studied in detail. The results suggest that a fault-propagation fold-type(FPFT) growth structure and growth strata developed on the western edge of the QJB and that the top part of Member 2 and Member 3 of the Tuchengzi Formation are growth strata controlled by the limb rotation mechanism. In two small thrust faults in the QJB, the Mesoproterozoic Xiamaling Formation is thrust over the Tuchengzi Formation, and these faults may have controlled the development of the fault-bend fold-type growth strata. An analysis of the "source-to-sink" process suggests that the hanging wall succession of the Shangyi-Pingquan fault(SPF) was the main source area of the Late Jurassic to early Early Cretaceous strata in the QJB. A zircon206 Pb/238 U age of 140.8±2.4 Ma for the volcanic rocks at the bottom of the FPFT growth strata represents the timing of the initiation of FPFT growth structure development. The discovery of the FPFT growth structure and growth strata in the QJB indicate that the QJB was an intermontane flexural basin controlled by fold and thrust structures during the early Early Cretaceous. Near the E-W-trending SPF, the NE-SW-trending Qianjiadian thrust fault(QJTF) and two small intrabasinal thrust faults may constitute a unified right-lateral strike-slip system that formed in response to the northwestward flat subduction of the Izanagi Plate beneath the East Asian continent during the Jurassic-Early Cretaceous. 展开更多

Modeling water and heat transfer in soil-plant-atmosphere continuum applied to maize growth under plastic film mulching

Based on our previous work modeling crop growth (CropSPAC) and water and heat transfer in the soilplant- atmosphere continuum (SPAC), the model was improved by considering the effect of plastic film mulching applied to field-grown maize in North-west China. In CropSPAC, a single layer canopy model and a multi-layer soil model were adopted to simulate the energy partition between the canopy and water and heat transfer in the soil, respectively. The maize growth module included photosynthesis, growth stage calculation, biomass accumutation, and participation. The CropSPAC model coupled the maize growth module and SPAC water and heat transfer module through leaf area index (LAI), plant height and soil moisture condition in the root zone. The LAI and plant height were calculated from the maize growth module and used as input for the SPAC water and heat transfer module, and the SPAC module output for soil water stress conditions used as an input for maize growth module. We used γS, the representation of evaporation resistance, instead of the commonly used evaporation resistance γS0 to reflect the change of latent heat flux of soil evaporation under film mulching as well as the induced change in energy partition. The model was tested in a maize field at Yingke irrigation area in North-west China. Results showed reasonable agreement between the simulations and measurements of LAI, above-ground biomass and soil water content. Compared with the original model, the modified model was more reliable for maize growth simulation under film mulching and showed better accuracy for the LAI (with the coefficient of determination R^2= 0.92, the root mean square of error RMSE = 1.23, and the Nush-Suttclife efficiency Ens = 0.87), the above-ground biomass (with R^2 = 0.96, RMSE= 7.17 t·ha^-1 and Ens = 0.95) and the soil water content in 0-1 m soil layer (with R^2 =0.7& RMSE = 49.44 mm and Ens = 0.26). EGFR mutations of plasmafree tumor DNA were detected by ADx-ARMS and ADxsuperamplification refractory mutation system (ADx-SuperARMS)at the same time. Patients with EGFR-mutantin tumor tissues and receiving EGFR-TKIs were finallyenrolled. Plasma mutation-positive patients with bothmethods were high abundance group. A method for determining vegetation growth process using remote sensing data: A case study in the Three-River Headwaters Region, China

Accurate measurements of the associated vegetation phenological dynamics are crucial for understanding the relationship between climate change and terrestrial ecosystems. However, at present, most vegetation phenological calculations are based on a single algorithm or method. Because of the spatial, temporal, and ecological complexity of the vegetation growth processes, a single algorithm or method for monitoring all these processes has been indicated to be elusive. Therefore, in this study, from the perspective of plant growth characteristics, we established a method to remotely determine the start of the growth season(SOG) and the end of the growth season(EOG), in which the maximum relative change rate of the normalized difference vegetation index(NDVI) corresponds to the SOG, and the next minimum absolute change rate of the NDVI corresponds to the EOG. Taking the Three-River Headwaters Region in 2000–2013 as an example, we ascertained the spatiotemporal and vertical characteristics of its vegetation phenological changes. Then, in contrast to the actual air temperature data, observed data and other related studies, we found that the SOG and EOG calculated by the proposed method is closer to the time corresponding to the air temperature, and the trends of the SOG and EOG calculated by the proposed method are in good agreement with other relevant studies. Meantime, the error of the SOG between the calculated and observed in this study is smaller than that in other studies. 展开更多

Recently, it is noted that the effects of FGF19 and FGF21 on metabolism suggest that endocrine fibroblast growth factors (FGFs) and their derivatives carry great potential as novel therapeutic agents for metabolic conditions. The pharmacological application of FGF21/19 holds great promise as an effective therapeutic means for treating obesity and diabetes. OBJECTIVE:To determine the therapeutic effect and potential mechanism of Huatan Tongluo decoction on rats with collagen-induced arthritis.METHODS:Forty specific pathogen-free Wistar rats were selected,and 10 were randomly selected as the control(group 1).The remaining rats were injected intradermally with emulsified type II bovine collagen at the tail base and back,followed by a booster 7 d post first immunization.After establishing collagen-induced arthritis(CIA),rats were randomly divided into three groups(n=10).The rats were treated orally for 30 d as follows:group 1,saline;group 2,model(saline);group 3,tripterygium polyglycoside(TP;7.81 mg/kg,positive control);group 4,Huatan Tongluo decoction(HTTL;7.5 g/kg).Body weight,ankle swelling and arthritis index were measured over the course of the study.The rats were sacrificed 30 d after treatment.Morphological changes in the synovium were observed by hematoxylin and eosin staining.Pannus formation and synovial thickness in the left ankle were observed by color Doppler ultrasoundVascular endothelial growth factor(VEGF)and VEGFR2 protein levels were measured by immunohistochemistry.VEGF/VEGFR2 mRNA levels were measured by real-time quantitative polymerase chain reaction.RESULTS:Compared with the model group,a significantly lower arthritis index was observed in the positive control group(P<0.05)and HTTL group(P<0.01),after treatment.Both positive control and HTTL reduced intra-articular pannus formation and synovial thickening.Furthermore,VEGF mRNA,and VEGFR2 protein and mRNA levels were significantly downregulated(P<0.05)in the treatment groups.CONCLUSION:Inhibition of the expression of VEGF and VEGFR2 in synovial tissues and the formation of pannus and synovial hyperplasia may be part of the mechanism of HTTL for relieving the symptoms of rheumatoid arthritis in CIA rats. 展开更多

Objective Epidermal growth factor receptor(EGFR) and vascular endothelial growth factor(VEGF) inhibitors are two targeted therapies for metastatic colorectal cancer(mCRC). However, few studies have focused on the safety and efficacy of combined targeted therapy against those of a single inhibition therapy of EFGR or VEGF. This meta-analysis aimed to compare the anti-tumor activity of the combined inhibition therapy and single inhibition therapy in patients with mCRC. Methods We searched PubMed, Medline, the Cochrane library, Embase, and annual meeting proceedings for relevant clinical trials. Objective response rate(ORR), progression-free survival(PFS), overall survival(OS), and adverse events were extracted and calculated.Results Nine trials comprising 3977 patients were selected for the analysis. The combined inhibition therapy showed a 3.7% improvement in ORR compared with single inhibition, and this difference was statistically significant [hazard ratio(HR)= 1.33;95% confidence interval(CI), 1.01–1.74;P = 0.04]. Subgroup analysis showed that the combined EGFR and VEGF inhibitor therapy had an 11.65% improvement in ORR compared with VEGF inhibitor therapy(OR = 2.14;95% CI, 1.34–3.40;P = 0.001). EGFR and VEGF inhibitor therapy and chemotherapy had an 18.08% improvement in ORR compared with chemotherapy(OR = 2.21;95% CI, 1.05–4.64;P = 0.04). Moreover, EGFR and VEGF inhibitor therapy significantly improved PFS compared with VEGF inhibitor therapy(OR = 0.82;95% CI, 0.69–0.97;P = 0.02). VEGF inhibitor therapy and chemotherapy significantly improved PFS compared with EGFR and VEGF inhibitor therapy and chemotherapy(OR = 1.20;95% CI, 1.11–1.30;P = 0.00). In addition, EGFR and VEGF inhibitor therapy showed improved OS compared with VEGF inhibitor therapy(HR = 0.78, 95% CI: 0.65–0.94;P = 0.008). In situ measurements of austenite grain growth were made at various temperatures in the range of 1273-1473 K with subsequent isothermal holding time of 3600 s for the Nb-Ti-bearing and Nb-Ti-free high carb on steel by using a confocal laser scanning microscope.The solid solute behavior of Nb-Ti carbides during austenitizing process was analyzed.The experimental results indicate that the austenite grains of both steels grow up gradually with increasing the heating temperature and holding time;the size and growth rate of austenite grain of Nb-Ti-bearing high carbon steel are much lower than those of Nb-Ti-free high carbon steel.A large amount of(Nb,Ti)(C,N)nanoparticles are observed in Nb-Tibearing steel,which retain the strong pinning effect on austenite grain boundary.The kinetics model of austenite grain growth of Nb-Ti-bearing steel during isothermal heat treatment is obtained and the predicted values calculated by using the model meet the experimental values very well. 展开更多

A CMOS compatible process is prese nted in order to grow self-catalyzed InAs nano wires on silic on by molecular beam epitaxy. The crucial step of this process is a new in-situ surface preparation under hydrogen (gas or plasma) during the substrate degassing combined with an in-situ arsenic ann eali ng prior to growth. Morphological and structural characterizati ons of the InAs nano wires are prese nted and growth mecha nisms are discussed in detail. The major in flue nee of surface termi nation is exposed both experime ntally and theoretically using statistics on ensemble of nanowires and density functional theory (DFT) calculations. The differences observed between Molecular Beam Epitaxy (MBE) and Metal Organic Vapor Phase Epitaxy (MOVPE) growth of I nAs nano wires can be explai ned by these differe nt surfaces termi nations. The transition between a vapor solid (VS) and a vapor liquid solid (VLS) growth mechanism is presented. Optimized growth conditions lead to very high aspect ratio nano wires (up to 50 nm in diameter and 3 micron in len gth) without passi ng the 410℃ thermal limit, which makes the whole process CMOS compatible. Overall, our results suggest a new method for surface preparation and a possible tuning of the growth mechanism using different surface termi nations. 展开更多
