Thinopyrum intermedium and barley are two close relatives of wheat and carry many genes that are potentially valuable for the improvement of various wheat traits.In this study we created wheat double substitution lines by hybridizing different wheat–Th.intermedium and wheat-barley disomic alien substitution lines,with the aim of using genes in Th.intermediumand barley for wheat breeding and investigating the genetic behavior of alien chromosomes and their wheat homoeologs.As expected,we obtained two types of wheat double substitution lines,2D2Ai#2(2B)2H(2A) and 2A2Ai#2(2B)2H(2D),in which different group 2 wheat chromosomes were replaced by barley chromosome 2H and Th.intermedium chromosome 2Ai#2.The new materialswere characterized usingmolecular markers,genomic in situ hybridization (GISH),and fluorescent in situ hybridization (FISH).GISH and FISH experiments revealed that the double substitution lines harbor 42 chromosomes including 38 wheat chromosomes,a pair of barley chromosomes,and a pair of Th.intermedium chromosomes.Analysis using specific DNAmarkers showed that two pairs of wheat homoeologous group 2 chromosomes in the new lines were substituted by a pair of 2H and a pair of 2Ai#2 chromosomes.Chromosome 2H showed a higher transmission rate than 2Ai#2,and both chromosomes were preferentially transmitted between generations via female gametes.Evaluation of botanic and agronomic traits demonstrated that,compared with their parents,the new lines showed similar growth habits and plant type but differences in plant height,flowering date,and self-fertility.Cytological observations using different probes suggested that the double substitution lines showed nearly normal genetic behavior before and during meiosis.The novel substitution lines can potentially be used in wheat meiosis research and breeding programs. 展开更多

Promising Peanut Lines in Northeastern Philippines 预览

Peanut or ground nut (Arachis hypogea L.) is one of the food legumes with commercial importance in the Philippines grown by farmers (with small landholdings) in most parts of the country. However, one of the major constraints to peanut is the lack of supply of good stocks coupled with modern production technology. To alleviate this problem especially in those areas planted to this crop is the development of improved lines, their evaluation to different locations and their maintenance for sustained production. Thus, the project aimed to adapt, evaluate, select and identify high yielding, disease-resistant and other desirable characters of peanut in northeastern Philippines particularly at Mariano Marcos State University (MMSU), Dingras, Ilocos Norte, conducted from July 2014 to May 2015. The different lines of peanut were arranged in a randomized complete block design (RCBD) replicated four times. Optimum cultural requirements from planting to harvesting were followed to permit expressions of the genetic potential of the different lines of the crop. During wet and dry season trials, results revealed significant differences in all treatments relative to pod and seed yield, seed size, shelling percentage and plant height except number of pods per plant, and days to flower and mature. The lines ICGV 00337, ICGV 95111, and ICGV 98371 out yielded the remaining seven peanut lines registering a pod yield of 1,860 to 1,020 kg/ha and had a yield advantage of 73% to 58.9%, respectively over the check variety Ilocos Red. Growing peanut in northeastern Philippines can approximately give an income ranging from Php87,120 to Php93,960 per hectare with a benefit/cost ratio of 2.02 to 2.19. 展开更多

The economically valuable oil crop Brassica napus(AACC,2 n=38),which arose from interspecific hybridization between the diploid ancestors Brassica rapa(AA,2 n=20) and Brassica oleracea(CC,2 n=18),has a complex genome.More than 10% of the assembled sequences,most of which belong to the C subgenome,have not been anchored to the corresponding chromosome.Previously,a complete set of monosomic alien addition lines(MAALs,C1–C9) with each of the nine C-subgenome chromosomes added to the extracted A subgenome was obtained from the allotetraploid B.napus donor Oro,after the ancestral B.rapa(RBR Oro) genome was restored.These MAALs effectively reduced the complexity of the B.napus genome.Here,we determined the expression values of genes on unanchored scaffolds in the MAALs and RBR Oro.Then,multiple comparisons of these gene expression values were used to determine the affiliations of the nonanchored scaffolds on which the genes were located.In total,54.68%(44.11 Mb) of the 80.67 Mb of non-anchored scaffolds belonging to the C subgenome were assigned to corresponding C chromosomes.This work highlights the potential value of these MAALs in improving the genome quality of B.napus. 展开更多

A Liutex based definition and identification of vortex core center lines 预览

Six core issues for vortex definition and identification concern with (1) the absolute strength,(2) the relative strength,(3) the rotational axis,(4) the vortex core center,(5) the vortex core size, and (6) the vortex boundary (Liu C. 2019). However, most of the currently popular vortex identification methods, including the Q criterion, the criterion and the Acj criterion etc., are Eulerian local region-type vortex identification criteria and can only approximately identify the vortex boundary by somewhat arbitrary threshold. On the other hand, the existing Eulerian local line-type methods, which seek to extract line-type features such as vortex core line, are not entirely satisfactory since most of these methods are based on vorticity or pressure minimum that will fail in many cases. The key issue is the lack of a reasonable mathematical definition for vortex core center. To address this issue, a Liutex (previously named Rortex) based definition of vortex core center is proposed in this paper. The vortex core center, also called vortex rotation axis line here, is defined as a line where the Liutex magnitude gradient vector is aligned with the Liutex vector, which mathematically implies that the cross product of the Liutex magnitude gradient vector and the Liutex vector on the line is equal to zero. Based on this definition, a novel three-step method for extracting vortex rotation axis lines is presented. Two test cases, namely the Burgers vortex and hairpin vortices, are examined to justify the proposed method. The results demonstrate that the proposed method can successfully identify vortex rotation axis lines without any user-specified threshold, so that the proposed method is very straightforward, robust and efficient. 展开更多

Drag reduction phenomenon in pipelines has received lots of attention during the past decades due to its potential engineering applications, especially in fluid transporting industries. Various methods to enhance drag reduction have been developed throughout the years and divided into two categories;non-additives method and additives method. Both categories have different types of methods, with different formulations and applications which will generally be discussed in this review. Among all the methods discussed, drag reduction using polymer additive is as one of the most enticing and desirable methods. It has been the subject of research in this field and has been studied extensively for quite some time. It is due to its ability to reduce drag up to 80% when added in minute concentrations. Reducing drag in the pipe will require less pumping power thus offering economic relieves to the industries. So, this paper will be focusing more on the use of polymer additives as drag reducing agent, the general formulations of the additives, major issues involving the use of drag reducing polymers, and the potential applications of it. However, despite the extensive works of drag reduction polymer, there are still no models that accurately explain the mechanism of drag reduction. More studies needed to be done to have a better understanding of the phenomenon. Therefore, future research areas and potential approaches are proposed for future work. 展开更多

Atacama Large Millimetre/sub-millimetre Array(ALMA)observations of CO(1–0)and CO(2–1)emissions from the circumstellar envelope of the asymptotic giant branch(AGB)star EP Aqr have been made with four times better spatial resolution than previously available.They are analysed with emphasis on the de-projection in space of the effective emissivity and flux of matter using as input a prescribed configuration of the velocity field,assumed to be radial.The data are found to display an intrinsic axisymmetry with respect to an axis making a small angle with respect to the line of sight.A broad range of wind configurations,from prolate(bipolar)to oblate(equatorial)has been studied and found to be accompanied by significant equatorial emission.Qualitatively,the effective emissivity is enhanced near the equator to produce the central narrow component observed in the Doppler velocity spectra and its dependence on star latitude generally follows that of the wind velocity with the exception of an omni-present depression near the poles.In particular,large equatorial expansion velocities produce a flared disc or a ring of effective emissivity and mass loss.The effect on the determination of the orientation of the star axis of radial velocity gradients,and possibly competing rotation and expansion in the equatorial disc,is discussed.In general,the flux of matter is found to reach a broad maximum at distances of the order of 500 AU from the star.Arguments are given that may be used to favour one wind velocity distribution over another.As a result of the improved quality of the data,a deeper understanding of the constraints imposed on morphology and kinematics has been obtained. 展开更多

Unusual infrared emission toward Sgr B2: possible planar C24

Interstellar graphene could be present in the interstellar medium(ISM), resulting from the photochemical processing of polycyclic aromatic hydrocarbon(PAH) molecules and/or collisional fragmentation of graphitic particles. Indeed, by comparing the observed ultraviolet(UV) extinction and infrared(IR) emission of the diffuse ISM with that predicted for graphene, as much as ~ 2% of total interstellar carbon could be locked up in graphene without violating the observational constraints. While the possible detection of planar C24, a small piece of a graphene sheet, has been reported towards several Galactic and extragalactic planetary nebulae, graphene has not yet been detected in interstellar environments. In this work, we search for the characteristic IR features of C24 at ~ 6.6, 9.8 and 20 μm toward Sgr B2, a high-mass star formation region, and find a candidate target toward R.A.(J2000)= 267.05855?and Decl.(J2000)=-28.01479?in Sgr B2 whose Spitzer/IRS spectra exhibit three bands peaking at ~ 6.637, 9.853 and 20.050 μm which appear to be coincident with those of C24. Possible features of C60 are also seen in this region. The candidate region is a warm dust environment heated by massive stars or star clusters, associated with a WISE spot(a tracer of star formation activity), close to the HII region candidate IRAS 17450–2759, and is surrounded by seven young stellar object candidates within ~ 5′, suggesting that the creation and/or excitation of C24 could be related to star formation activities. 展开更多

The Five-hundred-meter Aperture Spherical radio Telescope(FAST) is expected to complete its commissioning in 2019. FAST will soon begin the Commensal Radio Astronomy FasT Survey(CRAFTS), a novel and unprecedented commensal drift scan survey of the entire sky visible from FAST. The goal of CRAFTS is to cover more than 20000 deg2 and reach redshift up to about 0.35. We provide empirical measurements of the beam size and sensitivity of FAST across the 1.05 to 1.45 GHz frequency range of the FAST L-band array of 19-beams(FLAN). Using a simulated HI-galaxy catalogue based on the HI Mass Function(HIMF), we estimate the number of galaxies that CRAFTS may detect. At redshifts below 0.35, over 6 × 105 HI galaxies may be detected. Below the redshift of 0.07, the CRAFTS HIMF will be complete above a mass threshold of 109.5 M⊙. FAST will be able to investigate the environmental and redshift dependence of the HIMF to an unprecedented depth, shedding light onto the missing baryon and missing satellite problems. 展开更多

Laser-induced breakdown spectroscopy(LIBS)is regarded as a promising technique for real-time sorting of scrap metals due to its capability of fast multi-elemental and in-air analysis.This work reports a method for signal processing which ensures high accuracy and high speed during similar metal sorting by LIBS.Similar metals such as aluminum alloys or stainless steel are characterized by nearly the same constituent elements with slight variations in elemental concentration depending on metal type.In the proposed method,the original data matrix is substantially reduced for fast processing by selecting new input variables(spectral lines)using the information for the constituent elements of similar metals.Specifically,principal component analysis(PCA)of full-spectra LIBS data was performed and then,based on the loading plots,the input variables of greater significance were selected in the order of higher weights for each constituent element.The results for the classification test with aluminum alloy,copper alloy,stainless steel and cast steel showed that the classification accuracy of the proposed method was nearly the same as that of full-spectra PCA,but the computation time was reduced by a factor of 20 or more.The results demonstrated that incorporating the information for constituent elements can significantly accelerate classification speed without loss of accuracy. 展开更多

Pure lines derived from multiple parents provide abundant variation for genetic study.However,efficient genetic analysis methods and user-friendly software are still lacking.In this study, we developed linkage analysis methods and integrated analysis software for pure-line populations derived from four-way and eight-way crosses.First, polymorphic markers are classified into different categories according to the number of identifiable alleles in the inbred parents.Expected genotypic probability is then derived for each pair of complete markers, and based on them a maximum likelihood estimate(MLE) of recombination frequency is calculated.An EM algorithm is proposed for calculating recombination frequencies in scenarios that at least one marker is incomplete.A linkage map can thus be constructed using estimated recombination frequencies.We describe a software package called GAPL for recombination frequency estimation and linkage map construction in multi-parental pure-line populations.Both simulation studies and results from a reported four-way cross recombinant inbred line population demonstrate that the proposed method and software can build more accurate linkage maps in shorter times than other published software packages.The GAPL software is freely available from www.isbreeding.net and can also be used for QTL mapping in multi-parental populations. 展开更多

We have theoretically designed and experimentally observed free-space propagation of topological singular lines of cylindrical vector optical fields with non-integer topological charges. The polarization singular lines are due to the orientation uncertainty of the polarization states, caused by non-integer topological charges. The results reveal that during propagation, evolution of the polarization singular lines results in the special intensity pattern,distribution of polarization states, and chains of polarization singularities. We have also proposed a method to generate triple straight and spiral singular lines, which may contribute to the research of complex optical fields. 展开更多

Abstract Six new cell lines were established from embryonic tissues of the diamondback moth, Plutella xylostella (L.). The cell lines showed differential characteristics, including growth in attachment or in suspension, susceptibility to a baculovirus infection and expression of genes involved in the glucosinolate detoxification pathway in R xylostella larvae. Five of the cell lines grew attached to the culture flask and one cell line grew unattached as a suspension cell line. The cell lines had population doubling times ranging from IS to 23 h. Among five of the P. xylostella cell lines examined for infection of a nucleopolyhe. drovirus from Autographa californica, AcMNPV four cell lines were highly susceptible to AcMNPV infection, but one was only semi-permissive to AcMNPV infection. The production of two recombinant proteins, a β-galactosidase of bacterial origin and a secreted alkaline phosphatase of eukaryotic origin, in the R xylostella cell lines was examined in comparison with that in the cell line Sf9 which is commonly used for recombinant protein production. In the P. xylostella cell lines, expression of three important midgut genes involved in the glucosinolate detoxification pathway, including the glucosinolate sulfatase genes GSS1 and GSS2 and the sulfatase modifying factor gene SUMF1、was detected. The R xylostella cell lines developed in this study could be useful in in vitro research systems for studying insec-virus interactions and complex molecular mechanisms in glucosinolate detoxification and insect-plant interactions. 展开更多
