利用G-四链体DNA(5′-CTGGGAGGGAGGGAGGGA-3′)与氯化血红素结合形成G-四链体-HeminDNA酶,其能高效催化H2O2氧化反应底物由无色变为绿色,当溶液中有Ag^+或Hg^2+存在时会阻碍该DNA酶的形成,导致绿色溶液变浅。基于此,建立了比色法测定Ag^... 利用G-四链体DNA(5′-CTGGGAGGGAGGGAGGGA-3′)与氯化血红素结合形成G-四链体-HeminDNA酶,其能高效催化H2O2氧化反应底物由无色变为绿色,当溶液中有Ag^+或Hg^2+存在时会阻碍该DNA酶的形成,导致绿色溶液变浅。基于此,建立了比色法测定Ag^+和Hg^2+的传感器。在最佳实验条件下,溶液的吸光度与Ag^+和Hg^2+浓度分别在100.0~1000.0nmol/L和80.0~800.0nmol/L范围内具有良好的线性关系,检出限(3δ/Slope)分别为55.9nmol/L和64.3nmol/L。该方法具有较好的选择性,采用该方法对实际样品进行测试,结果满意。 展开更多

In this work,a metal-organic frameworks material MIL-88 was prepared easily using solvent-thermal method,and was first found to have catalytic activities similar to those of biological enzymes such as catalase and peroxidase.The material was characterized by XRD,SEM,TEM,EDX,FT-IR techniques and an N_2 adsorption method.It exhibited peroxidase-like activity through catalytic oxidation of the peroxidase substrate 3,3',5,5'-tetramethylbenzidine(TMB) in the presence of H_2O_2,producing a blue-colored solution.Under optimal conditions,the absorbance at 652 nm is linearly correlated with the concentration of H_2O_2 from 2.0×10~(-6) mol/L to 2.03×10~(-5) mol/L(R~2=0.981) with a detection limit of 5.62×10~(-7) mol/L(S/N=3).More importantly,a sensitive and selective method for ascorbic acid detection was developed using this material as a catalyst.The analytical method for ascorbic acid detection was observed to have a linear range from 2.57×10~(-6) mol/L to 1.01×10~(-5) mol/L(R~2=0.989) with a detection limit of 1.03×10~(-6) mol/L(S/N=3).This work suggests MOFs have advantages of preparing biomimetic catalysts and extends applications of the functional MOFs in the field of biosensor. 展开更多

OBJECTIVE:Tephrosia purpurea(Linn.) Pers.is widely used in traditional medicine to treat liver disorders,febrile attacks,enlargement and obstruction of liver,spleen,and kidney.In the present study,investigations were carried out to determine the seasonal impact on the content of flavonoid glycosides and on antioxidant activities so as to identify the optimal time of harvesting.METHODS:The plant materials were collected in different seasons during 2013-2014.Air-dried,powdered plant materials were extracted with 95%ethanol and ethanokwater(1:1) by ultrasound-assisted extraction process.Their chemical composition in terms of total polyphenol and flavonoid contents(TPCs and TFCs) was determined using modified colorimetric Folin-Ciocalteu method and aluminum chloride colorimetric assay respectively.To determine the in vitro antioxidant activity,diphenyl-picryl hydrazyl(DPPH) radical-scavenging assay and total antioxidant capacity by phosphomolybdate antioxidant assay were carried out.High-performance liquid chromatography(HPLC)/photo-diode array(PDA) analysis was used to quantify the flavonoid glycosides in the samples collected in different seasons.Correlation studies were also carried out between antioxidant activities and TPCs.RESULTS:The highest TPC and TFC were found to be in the 95%ethanolic extract of the August sample and the lowest in the 50%hydro-alcoholic extract of the plant sample collected in winter season.It was observed that in both the assays used to determine the antioxidant activity,the 95%ethanolic extracts in all the seasons showed a higher activity than their respective 50%hydroalcoholic extracts with an increase in activity as we go from cold to hot to rainy seasons.Based on correlation analysis,DPPH radical-scavenging activities as well as the spectrophotometrically measured phosphomolybdenum complex were also strongly correlated with TPC of the extracts.The most abundant flavonoid glycoside was quercetin-3-O-rhamnoglucoside in all the seasons.The content of all flavonoids was observed highest 展开更多

The fresh weight(FW) of total... Onion(Allium cepa L.) is a very important vegetable crop widely cultivated throughout the world. In this study, we measured total anthocyanins and flavonols of onions of three colors. The fresh weight(FW) of total anthocyanins were(29.99±1.19),(9.64±1.30) and(0.75±0.40) mg 100 g–1 fruit in red, yellow and white onions, respectively. Likewise, the FW of total flavonoids were(111.10±5.98),(36.64±3.59), and 0 mg 100 g–1 in red, yellow and white, respectively. Four types of anthocyanins(delphinidin 3,5-diglycosides, cyanidin 3,5-diglycosides, cyanidin 3-glycosides and cyanidin 3-(6′′-malonyl)-glucopyranoside) and two kinds of flavonoids(quercetin and quercetin 3-glycosides) were identified in two varieties(red and yellow bulb) of onions by HPLC/DAD-ESI/MS. The total polyphenol contents were also measured by means of Folin-Ciocalteu method. Moreover, all the concentrations of anthocyanins, flavonoids and polyphenols showed significantly positive correlations with antioxidant activities measured by DPPH˙, ABTS˙+ and FRAP assays. OBJECTIVE:Moxibustion is an important traditional Chinese medicine therapy using heat from ignited moxa fl oss for disease treatment.The purpose of the present study is to establish a reproducible method to assess the color of moxa fl oss,discriminate the samples based on chromatic coordinates and explore the relationship between chromatic coordinates and total fl avonoid content(TFC).METHODS:Moxa fl oss samples of different storage years and production ratios were obtained from a moxa production factory in Henan Province,China.Chromatic coordinates(L*,a* and b*)were analyzed with an ultraviolet-visible spectrophotometer and the chroma(C*)and hue angle(h°)values were calculated.TFC was determined by a colorimetric method.Data were analyzed with correlation,principal component analysis(PCA).RESULTS:Signifi cant differences in the chromatic values and TFC were observed among samples of different storage years and production ratios.Samples of higher production ratio displayed higher chromatic characteristics and lower TFC.Samples of longer storage years contained higher TFC.Preliminary separation of moxa fl oss production ratio was obtained by means of color feature maps developed using L*-a* or L*-b* as coordinates.PCA allowed the separation of the samples from their storage years and production ratios based on their chromatic characteristics and TFC.CONCLUSION:The use of a colorimetric technique and CIELAB coordinates coupled with chemometrics can be practical and objective for discriminating moxa fl oss of different storage years and production ratios.The development of color feature maps could be used as a model for classifying the color grading of moxa fl oss. 展开更多

Heparin has been considered to be a potentially useful ligand for low-density lipoprotein(LDL) detection and analysis in a clinical context.In order to construct an affinity surface for preferential adsorption of LDL,heparin-modified gold surface(GS-Hep) was fabricated by a self-assembling method and hydrophobic-modified gold surfaces(GS-Hydro) was used as a control.The morphologies of the modified gold surfaces were investigated by atomic force microscopy(AFM) and the quantity of heparin bound to gold surface was assayed by the toluidine blue(TB) colorimetric method.Water contact angles were determined to investigate wettability on GS-Hep and GS-Hydro.Surface plasmon resonance(SPR) technique was used subsequently to detect the selective binding of LDL with heparin.And the investigation on the effect of pH on LDL adsorption suggests that lower pH lead to higher quantities of LDL adsorption on GS-Hep.Compared with GS-Hydro,GS-Hep is selective for LDL from both single and binary protein solutions.Moreover,adsorbed LDL on GS-Hep could be washed off by injecting elution solution,such as NaCl solution,for the purpose of the regeneration of GS-Hep for further LDL adsorption. 展开更多

The interactions of antibiotic with living cells were studied by lectin conjugated gold nanoparticles(GNPs) based colorimetric assay. Because of the high affinity of lectin for saccharides, the lectin conjugated GNPs are able to employ as indicators for monitoring the antibiotic induced changes of glycosyl complexes. The interactions of a well known antibiotic, tunicamycin, with two different cell lines, HeLa and SHG-44, were selected to establish this assay. In the presence of tunicamycin, the dose- and time-dependence on the decreasing of binding affinity of lectin conjugated GNPs with living cells were demonstrated by conventional microscopic and UV-Vis spectroscopic studies. The experimental result demonstrates that our approach can be used to identify antibiotic induced expression difference of glycosyl complexes on different cellular surfaces and determine drug activity quantitatively. For further confirming the capability of the GNP-based assay, the system was also studied by confocal laser scanning microscopy(CLSM) and classic flow cytometry(FCM) assay, and satisfactory results were obtained. 展开更多
