The long-range interaction between Rydberg-excited atoms endows a medium with large optical nonlinearity.Here,we demonstrate an optical switch to operate on a single photon from an entangled photon pair under a Rydberg electromagnetically induced transparency configuration.With the presence of the Rydberg blockade effect,we switch on a gate field to make the atomic medium nontransparent thereby absorbing the single photon emitted from another atomic ensemble via the spontaneous fourwave mixing process.In contrast to the case without a gate field,more than 50%of the photons sent to the switch are blocked,and finally achieve an effective single-photon switch.There are on average 1-2 gate photons per effective blockade sphere in one gate pulse.This switching effect on a single entangled photon depends on the principal quantum number and the photon number of the gate field.Our experimental progress is significant in the quantum information process especially in controlling the interaction between Rydberg atoms and entangled photon pairs. 展开更多

BACKGROUND Diabetic nephropathy(DN)is the main cause of chronic kidney disease and endstage renal disease worldwide.Although available clinical trials have shown that endothelin receptor(ER)antagonists may be a novel and beneficial drug for DN,no consistent conclusions regarding their sufficient effectiveness and safety for patients with DN have been presented.AIM To assess the effectiveness and safety of ER antagonists among patients with DN.METHODS The EMBASE,PubMed,MEDLINE,Cochrane,and ClinicalTrials.gov databases were searched without any language restrictions.Relative risks with 95%confidence intervals(CIs)for dichotomous data and mean differences or standardized mean difference with 95%CIs for continuous data were calculated using Review Manager 5.3 software.Publication bias was assessed using Egger’s test with Stata/SE software.RESULTS We enrolled seven studies with six data sets and 5271 participants.The ER antagonists group showed a significantly greater reduction in albuminuria and more patients with 40%reduction in urinary albumin-to-creatinine ratio than the control group(P<0.0001 and P=0.02,respectively).Subgroup analysis for reductions in estimated glomerular filtration rate(eGFR)showed that for the middle-dosage subgroup,the ER antagonists group exhibited lower eGFR reduction than the control group(P<0.00001;mean difference,0.7095%CI:0.66,0.74).Moreover,significant reductions in systolic and diastolic blood pressure were observed in the invention group.CONCLUSION ER blockades combined with angiotensin converting enzyme inhibitor/angiotensin II type 1 receptor blockers may be an effective treatment to lower blood pressure and reduce proteinuria in DN with declined eGFR.However,attention should be given to adverse events,including cardiac failure,anemia,and hypoglycemia,as well as serious adverse events. 展开更多

Classical Hodgkin lymphoma(cHL) has been identified with universal genetic alterations of chromosome 9p24.1,which contains PD-L1/PD-L2 genes.The amplification of 9p24.1 is associated with the increased expression of PDL1 and PD-L2 on RS cells, which promotes their immune evasion, and subsequently makes cHL sensitive to PD-1 blockade.Several PD-1 inhibitors have shown significant efficacies with overall response rate(ORR) of 70%-90% in relapse/refractory(r/r) cHL and have acquired the approvals for this indication.Recently, more and more studies are conducted to investigate PD-1 blockade in earlier disease course and in combination with neo-agents or chemotherapy.Unlike cHL, non-Hodgkin lymphoma(NHL) consists of numerous subtypes harboring highly biological heterogeneity.Only a few subtypes have been shown to have genetic alteration of 9p24.1 including primary mediastinal B cell lymphoma(PMBL), gray zone lymphoma(GZL) with features intermediate between diffuse large B cell lymphoma(DLBCL) and cHL, primary central nervous system lymphoma(PCNSL) and primary testicular lymphoma(PTL).Epstein-Barr virus(EBV)-associated lymphomas have a virally mediated overexpression of PD-L1, also making them sensitive to PD-1 blockade.Therefore, PD-1 inhibitors are less effective in most r/r NHL than in r/r cHL.Further understanding of the biological features of NHL and immune checkpoint inhibitors(ICPi) combined therapy is the research focus in the future.In this review, we outlined the recent progress of ICPi in lymphoma originating from clinical studies. 展开更多

BACKGROUND Immunotherapy targeting programmed death-1(PD-1)or programmed deathligand-1(PD-L1)has been shown to be effective in a variety of malignancies but has poor efficacy in pancreatic ductal adenocarcinoma(PDAC).Studies have shown that PD-L1 expression in tumors is an important indicator of the efficacy of immunotherapy.Tumor cells usually evade chemotherapy and host immune surveillance by epigenetic changes.Protein arginine methylation is a common posttranslational modification.Protein arginine methyltransferase(PRMT)1 is deregulated in a wide variety of cancer types,whose biological role in tumor immunity is undefined.AIM To investigate the combined effects and underlying mechanisms of anti-PD-L1 and type I PRMT inhibitor in pancreatic cancer in vivo.METHODS PT1001B is a novel type I PRMT inhibitor with strong activity and good selectivity.A mouse model of subcutaneous Panc02-derived tumors was used to evaluate drug efficacy,toxic and side effects,and tumor growth in vivo.By flow cytometry,we determined the expression of key immune checkpoint proteins,detected the apoptosis in tumor tissues,and analyzed the immune cells.Immunohistochemistry staining for cellular proliferation-associated nuclear protein Ki67,TUNEL assay,and PRMT1/PD-L1 immunofluorescence were used to elucidate the underlying molecular mechanism of the antitumor effect.RESULTS Cultured Panc02 cells did not express PD-L1 in vitro,but tumor cells derived from Panc02 transplanted tumors expressed PD-L1.The therapeutic efficacy of anti-PD-L1 mAb was significantly enhanced by the addition of PT1001B as measured by tumor volume(1054.00±61.37 mm3 vs 555.80±74.42 mm3,P<0.01)and tumor weight(0.83±0.06 g vs 0.38±0.02 g,P<0.05).PT1001B improved antitumor immunity by inhibiting PD-L1 expression on tumor cells(32.74%±5.89%vs 17.95%±1.92%,P<0.05).The combination therapy upregulated tumorinfiltrating CD8+T lymphocytes(23.75%±3.20%vs 73.34%±4.35%,P<0.01)and decreased PD-1+leukocytes(35.77%±3.30%vs 6.48%±1.08%,P<0.001)in tumor tissue compared to th 展开更多

Immunotherapy strategies targeting the programmed cell death ligand 1(PD-L1)/programmed cell death 1(PD-1)pathway in clinical treatments have achieved remarkable success in treating multiple types of cancer.However,owing to the heterogeneity of tumors and individual immune systems,PD-L1/PD-1 blockade still shows slow response rates in controlling malignancies in many patients.Accumulating evidence has shown that an effective response to anti-PD-Ll/anti-PD-1 therapy requires establishing an integrated immune cycle.Damage in any step of the immune cycle is one of the most important causes of immunotherapy failure.Impairments in the immune cycle can be restored by epigenetic modification,including reprogramming the environment of tumor-associated immunity,eliciting an immune response by increasing the presentation of tumor antigens,and by regulating T cell trafficking and reactivation.Thus,a rational combination of PD-L1/PD-1 blockade and epigenetic agents may offer great potential to retrain the immune system and to improve clinical outcomes of checkpoint blockade therapy. 展开更多

PD-1 and CTLA-4 antibodies offer great hope for cancer immunotherapy.However,many patients are incapable of responding to PD-1 and CTLA-4 blockade and show low response rates due to insufficient immune activation.The combination of checkpoint blockers has been proposed to increase the response rates.Besides,antibody drugs have disadvantages such as inclined to cause immune-related adverse events and infiltration problems.In this study,we developed a cyclic peptide C25 by using Ph.D.-C7C phage display technology targeting LAG-3.As a result,C25 showed a relative high affinity with human LAG-3 protein and could effectively interfere the binding between LAG-3 and HLA-DR(MHC-II).Additionally,C25 could significantly stimulate CD8~+T cell activation in human PBMCs.The results also demonstrated that C25 could inhibit tumor growth of CT26,B16 and B 16-OVA bearing mice,and the infiltration of CD8~+T cells was significantly increased while FOXP3~+Tregs significantly decreased in the tumor site.Furthermore,the secretion of IFN-γby CD8~+T cells in spleen,draining lymph nodes and especially in the tumors was promoted.Simultaneously,we exploited T cells depletion models to study the anti-tumor mechanisms for C25 peptide,and the results combined with MTT assay confirmed that C25 exerted anti-tumor effects via CD8+T cells but not direct killing.In conclusion,cyclic peptide C25 provides a rationale for targeting the immune checkpoint,by blockade of LAG-3/HLA-DR interaction in order to enhance anti-tumor immunity,and C25 may provide an alternative for cancer immunotherapy besides antibody drugs. 展开更多

Diluted magnetic semiconductors(DMSs)have traditionally been employed to implement spin-based quantum computing and quantum information processing.However,their low Curie temperature is a major hurdle in their use in this field,which creates the necessity for wide bandgap DMSs operating at room temperature.In view of this,a single-electron transistor(SET)with a global back-gate was built using a wide bandgap ZnO nanobelt(NB).Clear Coulomb oscillations were observed at 4.2 K.The periodicity of the Coulomb diamonds indicates that the Coulomb oscillations arise from single quantum dots of uniform size,whereas quasi-periodic Coulomb diamonds correspond to the contribution of multi-dots present in the ZnO NB.By applying an AC signal to the global back-gate across a Coulomb peak with varying frequencies,single-electron pumping was observed;the increase in current was equal to the production of electron charge and frequency.The current accuracy of about 1%for both single-and double-electron pumping was achieved at a high frequency of 25 MHz.This accurate single-electron pumping makes the ZnO NB SET suitable for single-spin injection and detection,which has great potential for applications in quantum information technology. 展开更多

Checkpoint blockade based immune therapy has shown to be effective but benefit only the minority of patients whose tumors have been pre-infiltrated by T cells. To overcome this obstacles, a PEG-modified Bi2Se3 nanocage (NC) loaded with imiquimod (R848), which could efficiently destroy the tumors thus producing enough tumor-associated antigens (TAA) and with the existence of R848, a toll-like-receptor-7 agonist, could generate strong anti-cancer immune responses is reported in this study. Moreover, immunogenic Bi2Se3 NC-PEG/R848 mediated photothermal therapy (PTT) sensitizes tumors to checkpoint inhibition mediated by a PD-L1 antibody, not only ablating cancer cells upon NIR laser but also causing strong anti-cancer immunity to suppress distant tumor growth post PTT. Both in vitro and in vivo experiments demonstrate that the Bi2Se3 NC-PEG/R848 could effectively activate a PTT-induced immune response as well as silence immune resistance based on PD-L1 checkpoint blockade to ablate the primary tumor and further inhibit the tumor metastasis. Bi2Se3 NC reported here exhibits high photothermal conversion efficiency and stability, as well as competent drug loading capacity with large hollow structures and high surface area. Our study not only provides a facial way to synthesize Bi2Se3 NC, but also offers an alternative strategy for tumor metastasis. 展开更多

The pretreatment serum albumin/globulin ratio (AGR)has been used as a prognostic biomarker for various cancer types.However, the prognostic value of the AGR for prostate cancer,especially for metastatic prostate cancer (mPCa)after maximal androgen blockade (MAB),remains unclear.The aim of this study was to evaluate the prognostic value of the pretreatment serum AGR for mPCa treated with MAB.This retrospective study included 214 mPCa patients receiving MAB from October 2007 to March 2017. The correlation of the AGR with survival was estimated using Kaplan-Meier analysis and Cox proportional hazards models.The cutoff value of the AGR was 1.45 according to the receiver operating characteristic curve.Kaplan-Meier analysis demonstrated that patients with a low AGR (<1.45)had poor outcomes in terms of progression-free survival (PFS)and cancer-specific survival (CSS). Multivariate Cox analyses showed that the AGR was an independent predictor of PFS (hazard ratio [HR]=0.642;95%confidence interval [CI]:0.430-0.957;P=0.030)and CSS (HR =0.412;95%CI:0.259-0.654;P<0.001).Furthermore,in a subset of 79 patients with normal serum albumin levels (≥40.0 g l^-1),the serum AGR remained an independent predictor of CSS (P=0.009). The pretreatment AGR was an independent prognostic biomarker for PFS and CSS in patients with mPCa receiving MAB.In addition,the AGR remained effective for the prediction of CSS in patients with normal albumin levels (≥40 g l^-1).However,further prospective studies are needed to confirm our conclusions. 展开更多

Motivated by experimental advances that the collective excitation of two Rydberg atoms was observed, we provide an elaborate theoretical study for the dynamical behavior of two-atom Rabi oscillations. In the large-intermediate-detuning case, the two-photon Rabi oscillation is found to be significantly affected by the strength of the interatomic van der Waals interaction. With a careful comparison of the exact numbers and values of the oscillation frequency, we propose a new way to determine the strength of excitation blockade, agreeing well with the previous universal criterion for full, partial, and no blockade regions. In the small-intermediate-detuning case, we find a blockade-like effect, but the collective enhancement factor is smaller than 1/2 due to the quantum interference of double optical transitions involving the intermediate state.Moreover, a fast two-photon Rabi oscillation in ns timescale is manifested by employing intense lasers with an intensity of MW/cm2, offering a possibility of ultrafast control of quantum dynamics with Rydberg atoms. 展开更多

目的观察古溪针刀配合拔罐治疗腰背肌筋膜炎的临床效果。方法选取2013年10月~2014年3月于广州中医药大学附属中山医院脊柱科门诊就诊的腰背肌筋膜炎患者,共82例。采用随机数字表法将其分为治疗组(41例)和对照组[39例(脱落2例)],治疗组... 目的观察古溪针刀配合拔罐治疗腰背肌筋膜炎的临床效果。方法选取2013年10月~2014年3月于广州中医药大学附属中山医院脊柱科门诊就诊的腰背肌筋膜炎患者,共82例。采用随机数字表法将其分为治疗组(41例)和对照组[39例(脱落2例)],治疗组采用古溪针刀疗法配合拔罐治疗,对照组采用局部封闭注射加小针刀治疗。比较两组患者的平均治疗次数、临床疗效,并均于治疗前、治疗后1个月记录两组患者的视觉模拟评分(VAS)及Oswestry功能障碍指数(ODI)。结果治疗组的平均治疗次数较对照组少(P<0.05);治疗前两组患者VAS评分、ODI比较差异无统计学意义(P>0.05),治疗后两组患者VAS评分、ODI较治疗前降低(P<0.05),但两组患者治疗后组间VAS评分和ODI比较差异无统计学意义(P>0.05)。两组患者总有效率比较,差异无统计学意义(P>0.05)。结论两种疗法均可以缓解腰背肌筋膜炎患者的疼痛及功能障碍,但与局部封闭注射加小针刀治疗比较,古溪针刀疗法配合拔罐可在较少的治疗次数下改善患者症状,并取得很好的临床效果。 展开更多

The response rate depends on the immune microenvironment, tumor mutational burden (TMB), expression level of immune checkpoint proteins, and molecular subtypes of cancers. Along with the Cancer Genome Project, various open access databases, including The Cancer Genome Atlas and Gene Expression Omnibus, provide large volumes of data, which allow researchers to explore responsive or resistant biomarkers of immune checkpoint inhibitors? In this review, we introduced some methodologies on database selection, biomarker screening, current progress of immune checkpoint blockade in solid tumor treatment, possible mechanisms of drug resistance, strategies of overcoming resistance, and indications for immune checkpoint inhibitor therapy. 展开更多

Cervical cancer (CC) is the fourth most commonly diagnosed female malignancy and a leading cause of cancer-related mortality worldwide, especially in developing countries. Despite the use of advanced screening and preventive vaccines, more than half of all CC cases are diagnosed at advanced stages, when therapeutic options are extremely limited and side effects are severe. Given these circumstances, new and effective treatments are needed. In recent years, exciting progress has been made in immunotherapies, including the rapid development of immune checkpoint inhibitors. Checkpoint blockades targeting the PD-1/PD-L1 axis have achieved effective clinical responses with acceptable toxicity by suppressing tumor progression and improving survival in several tumor types. In this review, we summarize recent advances in our understanding of the PD-1/PD-L1 signaling pathway, including the expression patterns of PD-1/PD-L1 and potential PD-l/PD-Ll-related therapeutic strategies for CC. 展开更多

We propose how to achieve quantum nonreciprocity via unconventional photon blockade(UPB) in a compound device consisting of an optical harmonic resonator and a spinning optomechanical resonator. We show that, even with very weak single-photon nonlinearity, nonreciprocal UPB can emerge in this system, i.e., strong photon antibunching can emerge only by driving the device from one side but not from the other side. This nonreciprocity results from the Fizeau drag, leading to different splitting of the resonance frequencies for the optical counter-circulating modes. Such quantum nonreciprocal devices can be particularly useful in achieving backaction-free quantum sensing or chiral photonic communications. 展开更多

Immunotherapy has become a highly promising paradigm for cancer treatment. Herein, a chemo-immunotherapy was developed by encapsulating chemotherapeutic drug doxorubicin(DOX) and Toll-like receptor 7 agonist imiquimod(IMQ) in low molecular weight heparin(LMWH)-D-α-tocopheryl succinate(TOS) micelles(LT). In this process, LMWH and TOS were conjugated by ester bond and they were not only served as the hydrophilic and hydrophobic segments of the carrier, but also exhibited strong anti-metastasis effect. The direct killing of tumor cells mediated by DOX-loaded micelles(LT-DOX)generated tumor-associated antigens, initiating tumor-specific immune responses in combination with IMQ-loaded micelles(LT-IMQ). Furthermore, the blockade of immune checkpoint with programmed cell death ligand 1(PD-L1) antibody further elevated the immune responses by up-regulating the maturation of DCs as well as the ratios of CD8+ CTLs/Treg and CD4+ Teff/Treg. Therefore, such a multifunctional strategy exhibited great potential for inhibiting the growth of orthotopic and metastatic breast cancer. 展开更多
