为了研究色氨酸2,3-双加氧酶(TDO2)在梅花鹿茸角再生中的作用,本试验通过原位杂交方法检测TDO2mRNA在梅花鹿茸角中的表达。在体外分离培养的鹿茸软骨细胞中添加TDO2抑制剂680C91,诱导24h后,应用Real-time PCR方法检测肥大软骨细胞分化... 为了研究色氨酸2,3-双加氧酶(TDO2)在梅花鹿茸角再生中的作用,本试验通过原位杂交方法检测TDO2mRNA在梅花鹿茸角中的表达。在体外分离培养的鹿茸软骨细胞中添加TDO2抑制剂680C91,诱导24h后,应用Real-time PCR方法检测肥大软骨细胞分化标志分子Ⅹ型胶原(COLⅩ)及RUNT相关转录因子3(RUNX3)mRNA表达的变化,进一步研究TDO2对鹿茸软骨细胞分化的调节机理。结果发现,TDO2在鹿茸真皮层、间充质层及软骨层中均有表达。680C91诱导24h后,鹿茸软骨细胞中COLⅩ的表达量显著降低,同时RUNX3的表达水平也显著降低。以上结果表明,TDO2可能通过RUNX3来调节梅花鹿鹿茸软骨细胞的分化。 展开更多

目的探讨抑制自噬对根尖乳头干细胞成骨分化水平的影响。方法5、10ng/mL肿瘤坏死因子-α(TNF-α)分别处理根尖乳头干细胞(SCAPs),对照组不做处理,检测自噬相关蛋白LC3-II表达水平,GFP-LC3质粒转染并检测细胞内GFP-LC3数目,吖啶橙染色检... 目的探讨抑制自噬对根尖乳头干细胞成骨分化水平的影响。方法5、10ng/mL肿瘤坏死因子-α(TNF-α)分别处理根尖乳头干细胞(SCAPs),对照组不做处理,检测自噬相关蛋白LC3-II表达水平,GFP-LC3质粒转染并检测细胞内GFP-LC3数目,吖啶橙染色检测酸性囊泡情况。TNF-α,TNF-α+3-MA分别处理SCAPs,检测LC3-II的表达水平,GFP-LC3质粒转染并检测GFP-LC3数目,CCK-8法检测细胞活力,流式细胞仪检测细胞凋亡。TNF-α、TNF-α+3-MA分别处理SCAPs,对照组不做处理,诱导成骨向分化,qRT-PCR检测分化第3、7、14天时成骨相关基因碱性磷酸酶(ALP)、骨涎蛋白(BSP)、骨钙素(OCN)的表达。结果TNF-α可诱导SCAPs自噬活化:TNF-α组LC3-II/β-actin水平较对照组升高且具有浓度依赖性(P<0.05),TNF-α组GFP-LC3数目较对照组升高且具有浓度依赖性(P<0.05),TNF-α组酸性囊泡较对照组增多。3-MA可抑制TNF-α诱导的SCAPs自噬活化:TNF-α+3-MA组LC3-II/β-actin水平及细胞内GFP-LC3数目较TNF-α组显著降低(P<0.05),并下调细胞活力(P<0.05),上调细胞凋亡水平(P<0.05)。抑制自噬导致SCAPs成骨分化的抑制:TNF-α+3-MA组ALP、BSP表达量在分化第3、7、14天均较TNF-α组降低(P<0.05),OCN的表达量在第3、7天较TNF-α组降低(P<0.05)。结论TNF-α可诱导SCAPs自噬水平的活化;自噬可能对TNF-α作用下的SCAPs起细胞保护作用,对抗细胞凋亡;自噬的抑制将下调TNF-α作用下SCAPs的成骨向分化水平,提示自噬在TNF-α作用下SCAPs的成骨分化过程中具有重要作用。 展开更多

Stem cell transplantation has brought new hope for the treatment of neurological diseases.The key to stem cell therapy lies in inducing the specific differentiation of stem cells into nerve cells.Because the differentiation of stem cells in vitro and in vivo is affected by multiple factors,the final differentiation outcome is strongly associated with the microenvironment in which the stem cells are located.Accordingly,the optimal microenvironment for inducing stem cell differentiation is a hot topic.EGb761 is extracted from the leaves of the Ginkgo biloba tree.It is used worldwide and is becoming one of the focuses of stem cell research.Studies have shown that EGb761 can antagonize oxygen free radicals,stabilize cell membranes,promote neurogenesis and synaptogenesis,increase the level of brain-derived neurotrophic factors,and replicate the environment required during the differentiation of stem cells into nerve cells.This offers the possibility of using EGb761 to induce the differentiation of stem cells,facilitating stem cell transplantation.To provide a comprehensive reference for the future application of EGb761 in stem cell therapy,we reviewed studies investigating the influence of EGb761 on stem cells.These started with the composition and neuropharmacology of EGb761,and eventually led to the finding that EGb761 and some of its important components play important roles in the differentiation of stem cells and the protection of a beneficial microenvironment for stem cell transplantation. 展开更多

目的探讨瘦素(leptin)对人根尖乳头干细胞(human stem cells from the apical papilla,hSCAPs)增殖及成骨/成牙本质相关基因表达的影响,为临床年轻恒牙根尖持续发育的研究提供实验基础。方法采用人根尖乳头组织块培养法获得hSCAPs,以免... 目的探讨瘦素(leptin)对人根尖乳头干细胞(human stem cells from the apical papilla,hSCAPs)增殖及成骨/成牙本质相关基因表达的影响,为临床年轻恒牙根尖持续发育的研究提供实验基础。方法采用人根尖乳头组织块培养法获得hSCAPs,以免疫荧光染色法、Western blot和qRT-PCR分别检测hSCAPs中leptin及其受体OBRb蛋白及基因的表达。实验组用质量分数为0.1μg/mL的leptin(0.1μg/mL组)和1.5μg/mL的leptin(1.5μg/mL组)分别刺激hSCAPs,对照组仅加入α-MEM培养基,CCK-8、流式细胞仪法、qRT-PCR分别检测各组hSCAPs增殖情况及成骨/成牙本质相关基因:碱性磷酸酶(alkaline phosphatase,ALP)、牙本质基质蛋白-1(dentin matrix protein-1,DMP-1)和牙本质涎磷蛋白(dentin sialophosphoprotein,DSPP)、骨钙素(osteocalcin,OCN)mRNA的表达。结果hSCAPs表达leptin及其受体OBRb的蛋白及基因。与对照组相比,实验组细胞增殖能力及S期细胞比例均高于对照组(P<0.05),且其中1.5μg/mL组高于0.1μg/mL组(P<0.05)。3 d和7 d时,0.1μg/mL组、1.5μg/mL组hSCAPs的ALP、DMP-1、DSPP mRNA的表达量高于对照组,且1.5μg/mL组高于0.1μg/mL组,差异有统计学意义(P<0.05),14 d时0.1μg/mL组、1.5μg/mL组hSCAPs的ALP、DMP-1、DSPP mRNA的表达量明显低于对照组(P<0.05);0.1μg/mL组、1.5μg/mL组OCN mRNA的表达量于7 d、14 d时高于对照组,且1.5μg/mL组均高于0.1μg/mL组,差异有统计学意义(P<0.05),于14 d达到峰值。结论Leptin可促进hSCAPs增殖,并上调hSCAPs成骨/成牙本质相关基因的表达。 展开更多

This study aims to contribute to marketing theory by applying monopolistic competition theory to investigate how differentiation affects online retailers7 pricing power. We examine the intermediary effect of customer loyalty on the relationship between differentiation and pricing power of online retailers. We assume that customers have different preferences for the differentiated characteristics of online retailers, and thus different degrees of willingness to pay premiums, which gives online retailers pricing power. Structural equation modeling (SEM) is employed to test the hypothesized relati on ships between differentiation, customer loyalty, and pricing power. The statistical results of the empirical data indicate that some of the differentiation characteristics of online retailers can cultivate their pricing power indirectly. Logistics quality followed by commodity assortment and transaction security produces profound effects on the buildup of pricing power with customer loyalty as an intermediary factor. Results further reveal that commodity quality is positively related to pricing power. This study expands the definition of differentiation by combining corporate strategic positioning with commodity pricing. Implications for practice and di recti ons for future research are provided. 展开更多

It is universally recognized that the occurrence of hepatolithiasis is closely related to cholangitis, cholestasis, abnormal bile metabolism, anatomical abnormality of bile ducts, malnutrition and low socio-economic status. Surgery is the primary treatment method for hepatolithiasis in clinic, but some patients may still suffer from recurrence or complications after surgery. Traditional Chinese medicine(TCM) has its own unique advantages in preventing and treating recurrence of calculi. miR-199b促进慢性创面血管化的作用及机制研究 预览

目的:探索miR-199b对HaCaT细胞增殖、迁移及对VEGFA、JAG1、SET信号通路的影响。方法:转染miR-199b上调与下调慢病毒到人永生化表皮细胞(HaCaT)。分为miR-199b上调(up)、miR-199b下调(dowm)、空载对照(NC组)三个组,进行CCK8、凋亡、周... 目的:探索miR-199b对HaCaT细胞增殖、迁移及对VEGFA、JAG1、SET信号通路的影响。方法:转染miR-199b上调与下调慢病毒到人永生化表皮细胞(HaCaT)。分为miR-199b上调(up)、miR-199b下调(dowm)、空载对照(NC组)三个组,进行CCK8、凋亡、周期试验,并采用RT-PCR技术检测VEGFA、JAG1、SET基因的表达,采用WB技术检测miR-199b对下游蛋白变化(如VEGFA、JAG1等分子)的影响。结果:miR-199b up组OD450/fold值显著高于NC组,差异有统计学意义(P<0.05)、miR-199b down组OD450/fold值略低于NC组,差异无统计学意义(P>0.05)。与NC组相比,G1期miR-199b up组与miR-199bdown组的细胞增多(P<0.05);在S期,miR-199b up组及miR-199b down组的细胞减少(P<0.05);在G2/M期,miR-199bup组及miR-199b down组的细胞减少,差异均具有统计学意义(P<0.05)。与NC组比较,miR-199b up组凋亡细胞百分比下降(P<0.05)、miR-199b down组凋亡细胞百分比上升(P<0.05);SET在miR-199b up组的表达明显低于NC组,在miR-199bdown组表达明显高于NC组。从定量PCR结果可以看出,HaCaT细胞中,miR-199b up组JAG1表达丰度略高于NC组,差异无统计学意义(P>0.05),miR-199b down组JAG1表达丰度明显高于NC组(P<0.05);miR-199b up组VEGFA表达丰度明显低于NC组(P<0.05)、miR-199b down组VEGFA表达丰度明显高于NC组(P<0.05);miR-199b up组SET表达丰度小于NC组(P>0.05)、miR-199b down组SET表达丰度明显大于NC组(P<0.05)。结论:miR-199b可抑制VEGF、JAG1、SET蛋白的表达,为其靶基因,可促进细胞的增殖,延长细胞周期,并抑制细胞凋亡。 展开更多

miR-21在鸭胚胎期胸肌发育中的功能研究

x旨在通过研究鸭miR-21的分子特征、时空表达模式以及对鸭骨骼肌卫星细胞的增殖与分化的影响,为探索骨骼肌发育机制提供支撑。本研究选取180枚(98±5)g鸭蛋,相同条件孵化。从孵化第11天到第27天每天取3枚鸭蛋(分别表示为:E11、E12、... x旨在通过研究鸭miR-21的分子特征、时空表达模式以及对鸭骨骼肌卫星细胞的增殖与分化的影响,为探索骨骼肌发育机制提供支撑。本研究选取180枚(98±5)g鸭蛋,相同条件孵化。从孵化第11天到第27天每天取3枚鸭蛋(分别表示为:E11、E12、E13……E26、E27),无菌状态下逐只采集鸭胚胸肌样品。E27时另取3只鸭胚逐只采集腿肌、心、肝、肾、肌胃、小肠、腹脂和皮脂。混合酶消化法和差速贴壁法分离和纯化骨骼肌卫星细胞,骨骼肌卫星细胞增殖、分化时期分别过表达和干扰miR-21表达,利用荧光定量、蛋白杂交、EdU检测及流式细胞术等检测miR-21对鸭骨骼肌卫星细胞增殖和分化的影响。qRT-PCR结果显示,miR-21的表达随着孵化时间的增加而增加,E19天时到达最高,然后逐渐降低;E27时,miR-21在肝中的表达量最高。在增殖的鸭骨骼肌卫星细胞中过表达miR-21,荧光定量PCR检测CDK2和CyclinD1基因的mRNA表达量分别显著(P﹤0.05)和极显著增加(P﹤0.01),EdU检测阳性细胞数目显著增加(P﹤0.05),流式细胞仪检测发现G2期细胞数目极显著降低(P﹤0.01)、S期细胞数量显著增加(P﹤0.05)。在增殖的骨骼肌卫星细胞中,干扰miR-21表达,所得结果相反。在分化的鸭骨骼肌卫星细胞中过表达miR-21,qRT-PCR检测MyoG和MyHC基因的mRNA表达量均极显著增加(P﹤0.01);蛋白杂交(WB)检测结果显示,MyoG蛋白表达显著增加(P﹤0.05),MyHC蛋白表达极显著增加(P﹤0.01)。免疫荧光检测发现,MyHC阳性细胞数和10核以上的肌管数都极显著增加(P﹤0.01)。在分化的鸭骨骼肌卫星细胞中,干扰miR-21的表达,所得结果相反。结果表明,miR-21具有促进成肌细胞鸭骨骼肌卫星细胞增殖和分化的作用。 展开更多

成年火龙果植株茎段愈伤组织诱导、增殖及分化研究 预览

【目的】为建立和优化火龙果的再生体系。【方法】以成年植株茎段作为外植体,研究了不同表面灭菌方法、不同取样时间及取样部位对外植体污染、褐化的影响,以及不同植物生长调节剂浓度及配比、培养条件等对茎段愈伤组织诱导、增殖及分化... 【目的】为建立和优化火龙果的再生体系。【方法】以成年植株茎段作为外植体,研究了不同表面灭菌方法、不同取样时间及取样部位对外植体污染、褐化的影响,以及不同植物生长调节剂浓度及配比、培养条件等对茎段愈伤组织诱导、增殖及分化的影响。【结果】适宜幼嫩茎段表面灭菌的方式为75%酒精消毒30s+0.1%HgCl2消毒6~8min,适宜成熟茎段表面灭菌的方式为75%酒精消毒30s+0.1%HgCl2消毒12~15min;以春季萌发的10~20cm枝条诱导愈伤组织效果较好,最适愈伤组织诱导的培养基为MS+TDZ0.4mg/L+KT0.8mg/L,暗培养7d,再转入光照培养,愈伤组织为黄绿色、致密状,外表有颗粒状突起。愈伤组织增殖培养基以MS+TDZ0.4mg/L+KT(或ZT)0.8mg/L效果较好,增殖系数在8倍以上。在愈伤组织分化过程中,添加20%~30%的椰子水对不定芽的形成具有一定促进作用,但分化率较低。【结论】以成年火龙果植株茎段建立的再生体系,愈伤组织分化率总体表现较低,最高分化率仅13%,因此还有待进一步加强和完善。 展开更多

OBJECTIVE: The present study explored the effects of the combined herbal therapy consisting of curcumin (CUR) and Fructus Ligustri Lucidi (FLL) on aspects of bone regeneration. METHODS: Prior to analyzing the ability of this novel combined herbal therapy to promote aspects of bone regeneration, its cytotoxicity was determined using MC3T3-E1 cells (pre-osteoblast model). Cell proliferation was evaluated using phase-contrast microscopy and cell differentiation was estimated using alkaline phosphatase activity. The effect of the combined herbal therapy (CUR + FLL) was also assessed in terms of mineralization in the extracellular matrix (ECM) of cultured cells. Further, to explore the molecular mechanisms of bone formation, time-dependent expression of bone-regulating protein biomarkers was also evaluated. RESULTS: Combined herbal therapy (CUR + FLL) significantly upregulated the viability, proliferation and differentiation of MC3T3-E1 cells compared to the monotherapy of CUR or FLL. The magnitude of ECM mineralization (calcium deposition) was also higher in MC3T3-E1 cells treated with combined therapy. The time-dependent expression of bone-forming protein biomarkers revealed that the tendency of expression of these bone-regulating proteins was remarkably higher in cells treated with combined therapy.CONCLUSION: The co-administration of CUR and FLL had superior promotion of elements of bone regeneration in cultured cells, thus could be a promising alternative herbal therapy for the management of bone erosive disorders such as osteoporosis. 展开更多

目的研究碱性成纤维细胞因子(bFGF)对大鼠表皮干细胞分化为神经细胞的影响。方法分离获得饲养了1~3天的新生SD大鼠表皮基底层组织,利用10^min快速贴壁法消化、分离获得表皮干细胞,于倒置显微镜下观察表皮干细胞的形态。培养表皮干细胞... 目的研究碱性成纤维细胞因子(bFGF)对大鼠表皮干细胞分化为神经细胞的影响。方法分离获得饲养了1~3天的新生SD大鼠表皮基底层组织,利用10^min快速贴壁法消化、分离获得表皮干细胞,于倒置显微镜下观察表皮干细胞的形态。培养表皮干细胞的培养基为K-SFM,然后按照不同密度表皮干细胞进行分组处理(0.1×10^7/mL,0.3×10^7/mL,0.5×10^7/mL,0.1×10^6/mL),每组加入20ng/mL的bFGF,利用细胞免疫组织化学法检测细胞标志物Nestin和NSE的变化,以及观察细胞形态发生的变化。结果成功分离得到SD大鼠表皮干细胞;bFGF诱导后,0.3×10^7/mL组和0.1×10^7/mL组细胞第3天即可见细胞开始伸展生长,在约1周时细胞开始分化成神经细胞;且在细胞形态发生双极化改变的数目趋势上也具有一致性;Nestin和NSE检测均呈阳性表达。结论bFGF有助于诱导表皮干细胞向神经细胞的分化,而且该诱导作用具有一定的细胞密度依赖性。 展开更多

Although the remaining nerve tissue can regenerate and partly restore erectile function when the cavernous nerve is compressed/severed and function lost,the limited regenerative ability of these nerve tissues often fails to meet clinical needs.Adipose-derived stem cells are easy to obtain and culture,and can differentiate into neural cells.Their proliferation rate is easy to control and they may be used to help restore injured cavernous nerve function.Sprague-Dawley male rats(n=45)were equally randomized into three groups:fifteen rats as a sham-operated group,fifteen rats as a bilateral nerve crush(BINC)group(with no further intervention),fifteen rats as a BINC with intracavernous injection of one million neural-like cells from adipose-derived stem cells(NAS)(BINC+NAS)group.After 4 weeks,erectile function was assessed by stimulating the cavernous body.The number of myelinated axons in the dorsal cavernous nerve was determined by toluidine blue staining.The area of neuronal nitric oxide synthase-positive fibers in the dorsal penile nerve was measured by immunohistochemical staining.Masson staining was used to analyze the ratio of smooth muscle to collagen in penile tissue.The results demonstrate that maximal intracavernous pressure,the ratio of maximal intracavernous pressure to mean arterial pressure,the numbers of myelinated axons and neuronal nitric oxide synthase-positive fibers in the dorsal penile nerve,and the ratio of smooth muscle to collagen could be increased after cell transplantation.These findings indicate that neural-like cells from adipose-derived stem cells can effectively alleviate cavernous nerve injury and improve erectile function.All animal experiments were approved by the Animal Ethics Committee of Huazhong University of Science and Technology,China(approval No.2017-1925)on September 15,2017. 展开更多

Cypripedium tibeticum is a threatened orchid which efficient conservation requires knowledge of its extent and structure of genetic variation. Using two chloroplast DNA fragments(rps16 and trnL-F), we analyzed 157 individuals from 9 populations representing the species range in China. Seven haplotypes were identified. C. tibeticum had high total genetic diversity(H_T=0.80) with major contribution to this diversity made by among-population component(GST=0.64,ФST= 0.86). However, despite high population differentiation there was no clear phylogeographic structure. The populations CY and DC made the greatest contribution to the total gene diversity as well as allelic richness. The possible mechanisms and implications of these findings for conservation of the species are discussed. 展开更多

Behcet’s disease is a condition with a complicated and unclear etiology that comprises multi-systemic, chronic, inflammatory vasculitis. Behcet’s disease can affect every tissue and organ in the body,and is characterized by recurrent oral and genital ulcers, ocular inflammation, skin lesions, and other manifestations. The incidence of Behcet’s disease has a distinct regional specificity, and is most prevalent along the Silk Road, a route that stretched between the Mediterranean, Middle East and Far East.This article reviews the recent literature to evaluate the prevalence, clinical manifestations, pathogenesis and mechanism, and current treatments of Behcet’s disease. Furthermore, the etiology of Behcet’s disease will be evaluated from the aspect of Traditional Chinese Medicine(TCM) syndrome differentiation. As Behcet’s disease is complex and intractable, its treatment warrants further research.Traditionally, Behcet’s disease is treated with Western Medicine(WM) via medications that act locally and systemically;this WM treatment protocol usually has a good effect, but relapse can occur after reducing the dosage. Thus, it may be ideal to treat Behcet’s disease via a combination of WM and TCM.Recent studies have indicated that such a combination of Chinese and Western treatments has a better effect than either treatment alone. The aim of the present review is to describe the clinical features of Behcet’s disease, and to outline its possible pathogenesis in terms of both TCM and WM. Based on these findings, the present review proposes a Behcet’s disease treatment protocol composed of a combination of Chinese and WM that can effectively improve the occurrence of relapse caused by the reduction of the dosage of Western medication. 展开更多

Plant trichomes originate from epidermal cells.In this work,we demonstrated that a homeodomain-leucine zipper(HD-Zip)gene,GhA06G1283(Gh HD-1A),was related to the leaf trichome trait in allotetraploid cotton and could be a candidate gene for the T1 locus.The ortholog of GhHD-1A in the hairless accession Gossypium barbadense cv.Hai7124 was interrupted by a long terminal repeat(LTR)retrotransposon,while GhHD-1A worked well in the hairy accession Gossypium hirsutum acc.T586.Sequence and phylogenetic analysis showed that GhHD-1A belonged to the HD-Zip IV gene family,which mainly regulated epidermis hair development in plants.Silencing of GhHD-1A and its homoeologs GhHD-1D in allotetraploid T586and Hai7124 could significantly reduce the density of leaf hairs and affect the expression levels of other genes related to leaf trichome formation.Further analysis found that GhHD-1A mainly regulated trichome initiation on the upper epidermal hairs of leaves in cotton,while the up-regulated expression of GhHD-1A in different organs/tissues also altered epidermal trichome development.This study not only helps to unravel the important roles of GhHD-1A in regulating trichome initiation in cotton,but also provides a reference for exploring the different forms of trichome development in plants. 展开更多
