腐植酸对蛋鸡产蛋后期产蛋性能、抗氧化及促产蛋激素分泌的影响 预览

试验旨在探究天然腐植酸和合成腐植酸对蛋鸡产蛋后期生产性能、蛋品质、血清生化指标、抗氧化指标和激素水平的影响。选取52周龄体重、产蛋率相近的海兰白蛋鸡405羽,随机分成3个组,每组9个重复,每个重复15羽。对照组饲喂基础饲粮(CON组)... 试验旨在探究天然腐植酸和合成腐植酸对蛋鸡产蛋后期生产性能、蛋品质、血清生化指标、抗氧化指标和激素水平的影响。选取52周龄体重、产蛋率相近的海兰白蛋鸡405羽,随机分成3个组,每组9个重复,每个重复15羽。对照组饲喂基础饲粮(CON组),试验组在基础饲粮中分别添加1000 mg/kg的天然腐植酸A(HAA组)和合成腐植酸B(HAB组),预试期2周,正试期8周。结果表明:与CON组相比,添加腐植酸均可提高平均蛋重(P<0.05),降低不合格蛋率(P<0.05)和料蛋比(P<0.01),其中HAA组产蛋率极显著提高,HAA组的料蛋比低于HAB组(P<0.01);饲粮添加腐植酸对蛋品质无显著影响;与CON组相比,HAA组血清中白蛋白含量降低(P<0.05),HAB组乳酸脱氢酶活性提高(P<0.05),HAA组与HAB组血清中甘油三酯含量均降低(P<0.01);相比于CON组,添加HAB可提高谷胱甘肽过氧化物酶活性(P<0.05);与CON组相比,饲粮中添加腐植酸提高了血清中促黄体素水平(P<0.01),对促卵泡素水平有升高趋势(P=0.060),且HAA组中促卵泡素水平高于HAB组(P<0.01)。结果提示,腐植酸可促进机体促性腺激素分泌,改善机体抗氧化水平,显著提高产蛋后期蛋鸡的生产性能,且天然提取的腐植酸作用效果更佳。 展开更多

试验旨在探讨稳定可靠的贮精腺上皮细胞分离及原代培养方法,为研究鸡贮精机理提供细胞模型。以鸡输卵管的子宫阴道交接部组织样为材料,采用酶消化法和组织块培养法分离培养母鸡贮精腺上皮细胞,观察母鸡贮精腺上皮细胞的培养情况,比较不... 试验旨在探讨稳定可靠的贮精腺上皮细胞分离及原代培养方法,为研究鸡贮精机理提供细胞模型。以鸡输卵管的子宫阴道交接部组织样为材料,采用酶消化法和组织块培养法分离培养母鸡贮精腺上皮细胞,观察母鸡贮精腺上皮细胞的培养情况,比较不同细胞培养方法获得贮精腺上皮细胞的生长情况。结果表明,用胶原酶或胰酶单独消化母鸡子宫阴道交接部组织,经100目过滤后获得的贮精腺上皮细胞24 h后可贴壁,但48~72 h后细胞死亡;用胶原酶Ⅺ(0.01 g/mL)与胰酶(0.25%)先后消化母鸡子宫阴道交接部组织后再经100目过滤获得的贮精腺上皮细胞贴壁性良好,24~48 h细胞出现明显增殖,72 h后细胞增殖速度减慢,开始死亡;用组织块培养法7 d可获得鸡贮精腺上皮原代细胞,该细胞可传2~3代;用组织块培养法获得的细胞进行免疫组化试验,发现细胞表达贮精腺差异表达基因编码的NXPH1蛋白,该蛋白在培养细胞内的表达符合其分泌蛋白特性,表明组织块培养法所获细胞可用于后续研究。综上,用组织块培养法获得的鸡贮精腺上皮细胞可为研究母鸡贮精腺机制提供细胞模型。 展开更多

Maize grain is primarily used as an energy source for poultry and other animals.Maize has relatively high phytate-P content and very low intrinsic phytase activity.Given that feed phosphates are produced from finite rock phosphate resources,a reduction in the use of feed phosphates in maize-based diets by increasing the utilization of plant P sources by animals is necessary to make poultry meat and egg production more sustainable.The utilization of P by poultry is affected by two intrinsic characteristics of maize:the concentration of inositol phosphates and the activity of the intrinsic phytase of the grain in the digestive tract.The objective of this review is to present data on the variation that exists in composition of maize relevant for P use and to address factors that influence P utilization in maize-based diets of poultry.Broiler chickens and laying hens have the potential to degrade phytate in the gastrointestinal tract,but this is depressed by high dietary Ca and P concentrations.Published values of phytate degradation in broilers are overall higher than those in laying hens.Differences also exist between broiler chickens and growing turkeys and Pekin ducks.The exogenous supplementation of microbial phytases and the introduction of transgenic high phytase maize in poultry diets are efficient not only for the improvement of phytate-P digestibility,production performance,egg quality and bone mineralization,but also for the reduction of P excreta to control environmental impact. 展开更多

[Objectives]This study was conducted to investigate the effects of probiotics and enzyme preparation on cholesterol,triglyceride and serum alanine aminotransferase (ALT) and peroxidase (POD) in egg yolks.[Methods]Forty eight 450-day-old Hyline brown hens were randomly divided into 4 groups with 3 replicates in each group and 4 individuals in each replicate.Group 1 as the control check (CK) group was fed the basal diet;group 2 was fed the basal diet with the addition of 150 g/t of probiotics;group 3 was the basal diet +200 g/t lipase group;and group 4 was fed the basal diet with the addition of 150 g/t of probiotics and 200 g/t of lipase.The preliminary trial period was 10 d,and the trial period was 30 d.[Results]The contents of cholesterol and triglyceride in the egg yolks of the test groups supplemented with the probiotics and enzyme preparation were higher than those of the CK.On the 14^th day of treatment,the cholesterol contents in the egg yolks of group 2 and group 4 were significantly higher than that of group 1 ( P <0.05).On the 28^th day of treatment,the contents of cholesterol and triglyceride in the egg yolks of the three test groups were significantly higher than those in group 1 ( P <0.05).There were no significant differences in serum ALT and POD between various groups ( P >0.05).[Conclusions]The addition of probiotics and enzyme preparations to the feed of laying hens can increase the contents of cholesterol and triglyceride in egg yolks,without significantly affecting serum ALT and POD. 展开更多

This study was performed to evaluate the effect of folic acid(FA) on performance, egg quality and yolk5-methyltetrahydrofolate(5-MTHF) content. A total of 384 Hy-line W36 strain hens from 52 to 58 weeks of age were randomly assigned to 4 groups, and each group received one of following dietary treatments:0, 5.10 and 15 mg FA/kg diet. A completely randomized design was used. Egg production percentage, egg mass and egg weight were increased significantly(P < 0.05) and feed conversion ratio(FCR) was reduced significantly(P < 0.05) by increasing FA content in diets. No significant differences were detected among treatments on egg quality except for shell thickness. The dietary supplementations of laying hens diets with FA significantly increased yolk 5-MTHF content(P = 0.02). Overall, these data demonstrate that dietary supplementation with FA raised 5-MTHF content of yolk and productivity of egg production in laying hens. 展开更多

Background: Laying hens over 75 weeks of age commonly show great declines in immunity and production performance.It is unclear whether these declines can be relieved by supplementing with ascorbic acid(AA) in feed.Two trials were conducted to investigate the synthesis and metabolism of AA in layers of different ages and the effects of dietary supplemental AA on the performance and the immune and antioxidant statuses of 78 weeks old hens.Methods: In Exp.1,equal numbers(24 hens) of 35 weeks old(Young) and 75 weeks old(Old) layers were fed the same diet without AA supplementation for 4 weeks.In Exp.2,360 healthy 78 weeks old laying hens were randomly assigned to 4 treatments(basal diet supplemented with 0,0.25,0.5,or 1 g AA/kg diet) in an 8-week feeding trial.Results: The old hens tended to have decreased L-gulonolactone oxidase(GLO) synthase activity in the kidney and liver than that of the young hens(P = 0.07 and P = 0.05,respectively).Compared with the young hens,the old hens had lower hepatic antioxidant capacity allowing for the lower thioredoxin(TXN),thioredoxin reductase(TXNR) and cytochrome b5 reductase(CYB5 R) gene expression(P < 0.05),whereas increased sodium-dependent vitamin C transporter(SVCT) 1 expression levels in the ileum and kidney and enhanced splenic and hepatic AA concentrations(P < 0.05).Dietary supplementation with AA significantly decreased GLO enzyme activity but increased splenic AA concentration and anti-bovine serum albumin IgG levels(P < 0.05) and tended to increase CD4+T lymphocyte numbers(P = 0.06) in serum.Supplementation of 0.25 g AA/kg diet significantly increased hepatic total antioxidant capacity(T-AOC,P < 0.05) relative to the control group.Conclusions: Laying hens could synthesize AA in both the kidney and the liver,though the GLO enzyme activities were 100 times greater in kidneys than in livers.The old laying hens had greater absorption and reabsorption capacity and higher AA retention in some tissues that did the young hens.Dietary supplementation of AA can improve the health 展开更多

本试验旨在探讨微生态制剂对蛋鸡生产性能、免疫功能、蛋品质及粪便排放的影响。选用3 000只320日龄海兰褐蛋鸡,随机分为2组,每组3个重复,每个重复500只,两组正常饲喂基础饲粮,试验组饮水中添加微生态制剂,对照组正常饮水。试验预试期6d... 本试验旨在探讨微生态制剂对蛋鸡生产性能、免疫功能、蛋品质及粪便排放的影响。选用3 000只320日龄海兰褐蛋鸡,随机分为2组,每组3个重复,每个重复500只,两组正常饲喂基础饲粮,试验组饮水中添加微生态制剂,对照组正常饮水。试验预试期6d,正试期为60d。结果显示,试验组蛋鸡的生产性能、产蛋率和平均蛋重显著提高,料蛋比和破蛋率明显下降;试验组的血清总蛋白、血糖含量与对照组相比明显下降,而血清无机磷和钙水平明显提高;微生态制剂明显改善了试验组的蛋品质,哈夫单位提高了10%;两组每日粪便排放量和粪便总排放量均无显著影响(P>0.05),但试验组单位增重粪便排放量有所降低。该试验表明益生素饮水剂能够提高蛋鸡生产性能和鸡蛋品质、增强蛋鸡免疫功能、降低单位增重粪便排放量,是一种良好的绿色饲料添加剂。 展开更多

Effect of supplementing layer hen diet with phytogenic feed additives on laying performance, egg quality, egg lipid peroxidation and blood biochemical constituents

This study was conducted to evaluate the effects of supplementing laying hen diet with phytogenic additives on laying performance, egg quality, blood constituents and egg lipid peroxidation. Two hundred Lohmann Brown Lite laying hens were randomly allotted to 4 dietary treatments: control(without phytogenic additive), fennel seeds(5 g/kg), black cumin seeds(5 g/kg) and hot red pepper(5 g/kg).Each of the 4 diets was fed to 5 replicates of 10 hens for 8 weeks. No significant differences were observed in body weight or feed intake between the groups. Dietary inclusion of fennel followed by red pepper improved(P < 0.05) egg weight, egg production, egg mass and feed conversion ratio compared with control. Higher yolk shape index, shell and albumen weight percentages and Haugh unit(P < 0.05)were recorded in the fennel supplemented group compared with control. The egg yolk color score increased by the addition of fennel or hot red pepper in laying hen diets compared with control. The inclusion of black cumin or hot red pepper decreased serum and egg yolk cholesterol and malondialdehyde concentrations(P < 0.05) compared with control. Serum aspartate aminotransferase concentration was lower in black cumin group(P < 0.05) than in other treatments. In conclusion, the best laying performance and egg quality were obtained by dietary inclusion of fennel, followed by hot red pepper and black cumin. Dietary supplementation of black cumin or red pepper may lead to the development of low-cholesterol concentration and better antioxidant capacity of eggs. 展开更多

Background:Vitellogenin(VTG)is a precursor of egg yolk proteins synthesized within the liver of oviparous vertebrates.Betaine is an important methyl donor that is reported to improve egg production of laying hens with an unclear mechanism.In the present study,we fed betaine-supplemented diet(0.5%)to laying hens for 4 wk and investigated its effect on VTGII expression in the liver.Results:Betaine did not affect chicken weight,but significantly(P<0.05)increased egg laying rate accompanied with a significant(P<0.05)increase in hepatic concentration and plasma level of VTGII.Plasma estrogen level did not change,but the hepatic expression of estrogen receptorα(ERα)mRNA was significantly(P<0.05)up-regulated.Betaine did not affect the protein content of ERα,but significantly(P<0.05)increased hepatic expression of glucocorticoid receptor(GR)at both mRNA and protein levels.Also,ERα/GR interaction tended to be enhanced in the liver nuclear lysates of betainesupplemented hens as determined by co-immunoprecipitation.Furthermore,dietary betaine supplementation significantly increased(P<0.05)the hepatic expression of methyl-transfer enzymes,such as BHMT,GNMT,and DNMT1,which was associated with higher SAM/SAH ratio and hypomethylation of GR promoter regions.Conclusions:Betaine activates hepatic VTGII expression in association with modified DNA methylation of GR gene promoter,GR expression and ERα/GR interaction.Activation of hepatic VTGII expression may contribute,at least partly,to improved egg production in betaine-supplemented hens. 展开更多

Background:Phosphorus is an essential nutrient to maintain poultry health and performance.The objective of this study was to evaluate the effect of dietary phosphorus levels on egg production,egg quality,bone health,immune responses of laying hens challenged with Escherichia coli lipopolysaccharide.Methods:Three hundred laying hens at 28 wk were randomly divided into 2 dietary treatments with 10 replicates of 15 birds.The wheat-soybean based diets contained either 0.12%or 0.4%non-phytate phosphorus(NPP).At 32 wk of age,all the birds of each dietary treatment were injected into the abdomen with 1.5 mg/kg body weight(BW)of either LPS or saline once a day at 24-h intervals for continuous 9 d.The performance of laying hens was evaluated for 9 d.The eggs after the fifth injection were collected to value the egg quality.Three hours after the first injection,blood was collected to measure serum metabolite and immune response associated parameters.Three hours after the fifth injection,the hens were euthanized to obtain tibia,cecal tonsils and jejunum.Results:Compared with saline-injected hens,LPS-injected hens had lower feed intake and egg production(P<0.05).Eggshell thickness,strength,albumin height and Haugh unit were significantly increased in LPS-injected hens compared with saline-injected hens(P<0.05).Furthermore,laying hens challenged with LPS had lower villious height/crypt depth ration than those received saline.Serum calcium,phosphorus and SOD activities significantly decreased in the LPS-injected hens compared with the control(P<0.05).LPS up-regulated expression of IL-1β,IL-6 and IL-10 in cecum,and serum concentration of MDA,IL-1βand IL-6(P<0.05),whereas 0.40%dietary non-phytate phosphorus supplementation significantly increased(P<0.05)villi height/crypt depth ratio,decreased(P<0.05)serum MDA and IFN-γconcentration compared with the 0.12%non-phytate phosphorus group.Conclusion:In summary,this study demonstrates that 0.40%dietary non-phytate phosphorus supplementation significantly increased calcium and phosp 展开更多

本文旨在研究复合抗氧化剂对蛋鸡生产性能、蛋品质和蛋黄抗氧化力的影响。实验统用39周龄海蓝灰蛋鸡共计180羽,将蛋鸡随机进行分组,共分4组,设3个重复,每个重复15羽。对照组饲喂基础日粮,试验组(1、2、3组)分别在基础日粮基础上... 本文旨在研究复合抗氧化剂对蛋鸡生产性能、蛋品质和蛋黄抗氧化力的影响。实验统用39周龄海蓝灰蛋鸡共计180羽,将蛋鸡随机进行分组,共分4组,设3个重复,每个重复15羽。对照组饲喂基础日粮,试验组(1、2、3组)分别在基础日粮基础上添加5.0、7.5及10.0g/kg的复合抗氧化剂。试验周期为63d,预试期7d,正试期56d。结果表明,与对照组相比,添加7.5和10.0g/妇复合抗氧化剂产蛋率提高7.29%,哈氏单位、蛋壳厚度显著提高(P〈0.05)。添加5.0~10.0g/kg复合抗氧化剂对采食量和蛋重产蛋率、料蛋比、蛋黄颜色、蛋形指数无显著的影响(P〉0.05);添加复合抗氧化剂的试验组均能显著提高血清的超氧化物歧化酶、谷胱甘肽过氧化物酶和总抗氧化能力的活性(P〈0.05),降低了血清丙二醛(MDA)含量(P〈0.01);试验28d起,添加复合抗氧化剂极显著提高蛋黄T-AOC、降低MDA含量(P〈0.01);添加10.0g/kg复合抗氧化剂蛋黄T-AOC提高了137%~168%,MDA下降99.27%~166.97%。由此可知,复合抗氧化剂能够提高蛋鸡的抗氧化能力、产蛋率和哈氏单位,添加10.0g/kg复合抗氧化剂可显著提高鸡蛋的抗氧化力。 展开更多

为研究螺旋藻粉、地黄多糖和益母草提取物对蛋鸡蛋品质和部分生化指标的影响。试验选取210日龄海兰灰蛋鸡495只,采用三因素一次回归正交设计,随机分为11组,即1个对照组,10个试验组,分别在饲粮中添加螺旋藻粉5、10、15g/kg,地黄... 为研究螺旋藻粉、地黄多糖和益母草提取物对蛋鸡蛋品质和部分生化指标的影响。试验选取210日龄海兰灰蛋鸡495只,采用三因素一次回归正交设计,随机分为11组,即1个对照组,10个试验组,分别在饲粮中添加螺旋藻粉5、10、15g/kg,地黄多糖提取物2.5、3.0、3.5g/kg,益母草提取物0.7、0.9、1Ag/kg。结果表明:①饲粮添加螺旋藻粉、地黄多糖和益母草提取物,使平均蛋重、蛋黄颜色、哈夫单位明显增加,胆固醇、甘油三酯含量显著降低(P〈0.05)。添加螺旋藻粉10g/kg、地黄多糖3.0g/ks和益母草提取物0.9g/kg,哈氏单位比对照组增加了9.04%(P〈0.05)。胆固醇、甘油三酯均以添加螺旋藻粉15g/ks、地黄多糖提取物3.5g/kg和益母草提取物1.1g/kg为最佳。②随螺旋藻粉添加量的增加,蛋黄颜色、铁含量显著增加,胆固醇、甘油三酯含量显著降低(P〈0.05)。随地黄多糖提取物添加量的增加,蛋壳强度极显著增加(P〈0.01),蛋黄颜色有增加的趋势,甘油三酯含量有降低的趋势(P〈0.1)。随益母草提取物添加量的增加,蛋黄颜色显著增加(P〈0.05),蛋壳强度极显著增加(P〈0.01)。③螺旋藻粉和地黄多糖提取物对平均蛋重、蛋壳强度的拮抗作用极显著(P〈0.01),益母草提取物和地黄多糖提取物对蛋壳强度、胆固醇含量的拮抗作用显著(P〈0.05)。考虑相互作用和经济效益,添加螺旋藻粉10g/kg、地黄多糖提取物3.0g/kg和益母草提取物0.9g/kg的效果最好。 展开更多
