Immune checkpoint inhibitors (ICIs) are monoclonal antibodies that target downregulators of the anti-cancer immune response: Cytotoxic T-lymphocyte antigen- 4, programmed cell death protein-1, and its ligand programmed death-ligand 1. ICIs have revolutionized the treatment of a variety of malignancies. However, many immune-related adverse events have also been described which mainly occurs as the immune system becomes less suppressed, affecting various organs including the gastrointestinal tract and causing diarrhea and colitis. The incidence of immune-mediated colitis (IMC) ranges from 1%-25% depending on the type of ICI and if used in combination. Endoscopically and histologically there is a significant overlap between IMC and inflammatory bowel disease, however more neutrophilic inflammation without chronic inflammation is usually present in IMC. Corticosteroids are recommended for grade 2 or more severe colitis while holding the immunotherapy. About one third to two thirds of patients are steroid refractory and benefit from infliximab. Recently vedolizumab has been found to be efficacious in steroid and infliximab refractory cases. While in grade 4 colitis, the immunotherapy is permanently discontinued, the decision is controversial in grade 3 colitis. 展开更多

Immunotherapy has become the fourth cancer therapy after surgery, chemotherapy, and radiotherapy. In particular, immune checkpoint inhibitors are proved to be unprecedentedly in increasing the overall survival rates of patients with refractory cancers, such as advanced melanoma, non-small cell lung cancer, and renal cell carcinoma. However, inhibitor therapies are only effective in a small proportion of patients with problems, such as side effects and high costs. Therefore, doctors urgently need reliable predictive biomarkers for checkpoint inhibitor therapies to choose the optimal therapies. Here, we review the biomarkers that can serve as potential predictors of the outcomes of immune checkpoint inhibitor treatment, including tumor-specific profiles and tumor microenvironment evaluation and other factors. 展开更多

Objective: To explore the effects of Yinzhihuang oral liquid and albumin on bilirubin, inflammatory factors, immune indexes and related factors in neonatal pathologic jaundice. Methods: A total of 134 neonates with pathologic jaundice admitted to our hospital from May 2017 to April 2018 were randomly selected as the control group (n=67) and the observation group (n=67), the control group was treated with albumin, the observation group was treated with Yinzhihuang Oral Liquid on the basis of the control group. The bilirubin, inflammatory factors, immune indicators, alpha-fetoprotein (AFP) and transferrin (TRF) were compared and analyzed before and after treatment. Results: After treatment, the levels of TBIL, DBIL and IBIL in both groups were significantly lower than those before treatment (P<0.05), and the levels of TBIL, DBIL and IBIL [(118.60±10.85) μmol/L, (6.95±1.52) μmol/L, (115.30±14.20)μmol/L] in observation group were significantly lower than those in control group;the levels of CRP and IL-6 in both groups were significantly lower than those before treatment (P<0.05), and the levels of CRP and IL-6 [(8.26±2.07) mg/L, (12.69±2.15) pg/mL] in observation group were significantly lower than those in control group (P<0.05);the levels of CD4+, CD4+/CD8+ in both groups were significantly higher than those before treatment (P<0.05), while the levels of CD8+ was significantly lower than that before treatment, the levels of CD4+, CD4+/CD8+ [(47.08±5.70)%, (2.08±0.41)] in observation group were significantly higher than those in control group (P<0.05), and the level of CD8+ [(22.90±2.05)%] was significantly lower than that in control group (P<0.05);the levels of AFP in significantly higher than before treatment (P<0.05), and the levels of AFP [(12.69±3.04)mg/L] in observation group were significantly lower than those in control group (P<0.05), the levels of TRF [(2.02±0.35) g/L] were significantly higher than those in control group (P<0.05). Comparative susceptibility and immune responses of Asian and European honey bees to the American foulbrood pathogen, Paenibacillus larvae

Currently, this pathogen is widespread in the European honey bee— Apis mellifera. However, little is known about infectivity and pathogenicity of P. lan'ae in the Asiatic cavity-nesting honey bees, Apis cerana. Moreover, comparative knowledge of P. larvae infectivity and pathogenicity between both honey bee species is scarce. In this study, we examined susceptibility, larval mortality, survival rate and expression of genes encoding antimicrobial peptides (AMPs) including defensin, apidaecin, abaecin, and hymenoptaecin in A. mellifera and A. cerana when infected with P. larvae. Our results showed similar effects of P. larvae on the survival rate and patterns of AMP gene expression in both honey bee species when bee larvae are infected with spores at the median lethal concentration (LC5 0 ) for A. mellifera. All AMPs of infected bee larvae showed significant upregulation compared with noninfected bee larvae in both honey bee species. However, larvae of A. cerana were more susceptible than A. mellifera when the same larval ages and spore concentration of P. larvae were used. It also appears that A. cerana showed higher levels of AMP expression than A. mellifera. This research provides the first evidence of survival rate, LC50 and immune response profiles of Asian honey bees, A. cerana, when infected by P. larvae in comparison with the European honey bee, A. mellifera. 展开更多

Introducing chimeric antigen receptor into immune cells against malignancies has contributed to a revolutionary innovation in cancer immunotherapy. As an important type of adaptive immune cells, T cells first caught researchers’ attention and became great success in chimeric antigen receptor-based immunotherapy. However, engineered T cells seem to hit their bottleneck when resistance of cancerous cells, less encouraging responses in solid tumors and unwanted toxicities to the host remain to be solved.Meanwhile, innate immune cells get to join the race. Representatives such as natural killer cells, natural killer T cells, γδT cells and macrophages also prove to be well redirected with chimeric antigen receptors. Compared to chimeric antigen receptor engineered T cells, these engineered innate immune cells may possess multiple targeting and killing mechanisms, have the potential to crack the barrier of solid tumors and have less side effects in the host. Besides, possible universal access to cell resources and improvements in expansion and transduction techniques make these cells promising candidates with huge potential in translational medicine. Therefore, innate immune cells claim a brand-new dimension and are likely to supplement T cells greatly in the field of chimeric antigen receptor-based immunotherapy. 展开更多

目的探究经颈静脉肝内门体分流术(TIPS)联合部分性脾栓塞术(PSE)对肝硬化伴脾亢患者外周血细胞水平、免疫球蛋白、血氨及安全性的影响。方法选择74例肝硬化患者,依据治疗方法分为观察组(n=37)和对照组(n=37),对照组仅行TIPS治疗,观察组... 目的探究经颈静脉肝内门体分流术(TIPS)联合部分性脾栓塞术(PSE)对肝硬化伴脾亢患者外周血细胞水平、免疫球蛋白、血氨及安全性的影响。方法选择74例肝硬化患者,依据治疗方法分为观察组(n=37)和对照组(n=37),对照组仅行TIPS治疗,观察组行TIPS与PSE联合治疗。分析比较两组外周血细胞水平、免疫球蛋白、血氨及不良反应的变化。结果术后7d及30d,观察组外周血细胞血小板(PLT)、白细胞(WBC)水平明显高于术前及对照组(t分别=6.25、4.25、6.30、4.63、7.34、4.71、6.25、4.69,P均<0.05);术后30d,观察组IgA、IgG及IgM水平明显高于术前及对照组(t分别=3.78、3.14、3.94、3.23、3.84、3.20,P均<0.05)。术后30d,两组血氨水平明显高于术前(t分别=6.42、5.07,P均<0.05),两组间血氨水平比较,差异无统计学意义(t=0.90,P>0.05)。两组轻微并发症及严重并发症的比较,差异均无统计学意义(χ^2分别=0.35、0.56,P均>0.05)。结论TIPS与PSE联合治疗可改善肝硬化伴脾亢患者外周血细胞水平,减轻患者免疫抑制作用,不增加患者术后并发症风险。 展开更多

本试验旨在研究α-硫辛酸(α-LA)对热应激绵羊生长性能、抗氧化及免疫的影响,以期阐述其抗热应激的机理。选择4~5月龄、体重35~40kg的绵羊15只,随机分为对照组(CTL)、低剂量组(LA-L,添加600mg/kgα-LA)、高剂量组(LA-H,添加900mg/kgα-L... 本试验旨在研究α-硫辛酸(α-LA)对热应激绵羊生长性能、抗氧化及免疫的影响,以期阐述其抗热应激的机理。选择4~5月龄、体重35~40kg的绵羊15只,随机分为对照组(CTL)、低剂量组(LA-L,添加600mg/kgα-LA)、高剂量组(LA-H,添加900mg/kgα-LA),在热应激条件下(7~8月)饲喂40d。结果表明:1)与对照组相比,α-LA添加组平均日采食量增加,低剂量组和高剂量组分别比对照组增加了7.3%和2.7%(P>0.05);高剂量组血清中丙氨酸转氨酶(ALT)、门冬氨酸转氨酶(AST)活性及α-LA添加组血浆中丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-8(IL-8)含量显著下降(P<0.05);α-LA添加组血浆中三碘甲腺原氨酸(T3)、热休克蛋白70(HSP70)、免疫球蛋白G(IgG)含量及超氧化物歧化酶(SOD)活性显著升高(P<0.05)。2)与对照组相比,α-LA添加组肝脏中炎性相关因子IL-1β、IL-8、核因子-κB(NF-κB)mRNA表达量显著下降(P<0.05),抗炎因子干扰素-γ(IFN-γ)mRNA表达量显著升高(P<0.05)。综上,α-LA显著增强热应激绵羊的抗氧化能力,减少炎症相关因子的表达,增强抗炎因子的表达,提高机体免疫机能,且与α-LA剂量有关。 展开更多

Cardiotoxicity with conventional anti-cancer therapies has been well studied in the past and clear guidelines for management of these side effects are available in the literature. However, cardiotoxicity with novel agents such as ICIs has been fairly under-reported and/or underestimated and we are yet to formulate clear guidelines for management of these rare side effects. In the last few years, there has been an overall increase in the number of cases of cardiotoxicity related to ICIs. In this literature review, we describe the mechanism of action of the most widely used ICIs and their related cardiotoxicities. The increase in number of case reports about the potential of cardiotoxicities with these novel agents clearly indicates the need for a new insight into the field of cardio-immuno-oncology. 展开更多

Objective: Tumor microenvironment, especially the host immune system, plays a pivotal role in tumor initiation and progression. Profiling of immune signature within tumor might uncover biomarkers for targeted therapies and clinical outcomes. However, systematic analysis of immune-related genes in gastric cancer(GC) has not been reported.Methods: Expressions of a total of 718 immune-related genes were generated in 372 stomach adenocarcinoma(STAD) patients from The Cancer Genome Atlas(TCGA) database using RNA-sequencing data. Integrated bioinformatics analyses were performed to identify prognostic factors as well.Results: Survival analyses revealed 73 genes, which were significantly associated with patient’s overall survival(OS). Taken together with clinicopathological parameters, we established a predictive model, containing 10 immune-related genes, which were NRP1, C6, CXCR4, LBP, PNMA1, TLR5, ITGA6, MICB, PBK and TNFRSF18,with powerful efficiency in distinguishing satisfactory or poor survival of STAD patients. Moreover, the top 3 ranked prognostic genes, NRP1, TGFβ2 and MFGE8, were also significantly associated with patient’s OS by an independent validation achieved from Kaplan-Meier plotter database.Conclusions: We profiled prognostic immune signature and established prognostic predictive model for GC,which could reflect immune disorders within tumor microenvironment, and also may provide novel predictive and therapeutic targets for GC patients in the near future. 展开更多

Methods: A total of 160 patients with type 2 diabetes mellit... Objective: To investigate the effects of Liraglutide on endothelial function, immune function and related factors in patients with type 2 diabetes mellitus. Methods: A total of 160 patients with type 2 diabetes mellitus who received treatment in department of endocrinology of our hospital from December 2015 to June 2018 were collected. They were randomly divided into the control group and the observation group, 80 cases in each group. The patients in the control group were treated with metformin. The patients in the observation group were treated with liraglutide on the basis of the control group. The levels of endothelin (ET-1), nitric oxide (NO), immunoglobulin A (IgA), immunoglobulin M (IgM), immunoglobulin G (IgG), hypersensitive C-reactive protein (hs-CRP), insulin-like growth factor-1 (IGF-1), tumor necrosis factor- (TNF-α), Vaspin and HbAlc were detected and compared. Results: After treatment, compared with the treatment group, the serum levels of ET-1, hs-CRP, IGF-1, TNF-α and HbAlc of patients in the control group and the observation group were significantly decreased, and the levels of NO, IgA, IgG, IgM and Vaspin increased significantly. The change trend of the above indexes of patients in the observation group was more obvious than those in the control group (P<0.05). Conclusion: Liraglutide combined with metformin in the treatment of type 2 diabetes mellitus can significantly enhance endothelial function and immune function of patients, and alleviate inflammation, and it has good clinical efficacy. 展开更多

Hepatocellular carcinoma (HCC) is a highly aggressive malignant disease, with a poor clinical prognosis. Many standard therapies are often considered for HCC treatment today;however, these conventional therapies often fail to achieve sufficiently effective clinical results. Today, HCC therapy is set to undergo a major revolution, owing to rapid developments in cancer immunotherapy, particularly immune checkpoint inhibitor therapy. Cancer immunotherapy is a novel and promising treatment strategy that differs significantly from conventional therapies in its approach to achieve antitumor effects. In fact, many cancer immunotherapies have been tested worldwide and shown to be effective against various types of cancer;HCC is no exception to this trend. For example, we identified a specific cancer antigen called glypican-3 (GPC3) and performed clinical trials of GPC3-targeted peptide vaccine immunotherapy in patients with HCC. Here, we present an overview of the immune mechanisms for development and progression of HCC, our GPC3-based immunotherapy, and immune checkpoint inhibitor therapy against HCC. Finally, we discuss the future prospects of cancer immunotherapy against HCC. We believe that this review and discussion of cancer immunotherapy against HCC could stimulate more interest in this promising strategy for cancer therapy and help in its further development. 展开更多

The incidence of gastrointestinal (GI) tumors is increasing year by year, and its pathogenesis is closely related to the intestinal flora. At present, the use of antibiotics is very common in the clinic. And cancer patients with low immunity are vulnerable to all sorts of infections, such as respiratory tract infections and urinary tract infections. Moreover, cancer patients easily run into fever and neutropenia induced by myelosuppression. Therefore, antibiotics are used extensively and even overused in many conditions. However, because of the special anatomical location of the gastrointestinal tract, the antibiotic usage will bring changes to the intestinal flora. Besides, with the expanding popularity of immunotherapy, various factors affecting the efficacy of immune checkpoint inhibitors (ICIs) have been extensively explored, including cancer-associated inflammation and the local and systemic factors that lead to immunosuppression. Some biomarkers for ICIs, including the expression of PD-L1, tumor mutation load, and microbiota, also have been investigated, and many studies have confirmed that gut microbiota can affect the efficacy of immunotherapy. But further studies on the influence of antibiotics directly on immunotherapy are rare. In this review, we discuss the relationship between GI tumors and antibiotics, the current status of immunotherapy in GI tumors, and the influence of antibiotics on immunotherapy. 展开更多
